Ранее в экспериментах показано действие гиберрелловой,индолилуксусной кислоты и кинетина на рост волокна в культуре сем почки хлопчатника. Использование этих фитогормонов позволило автору найти оптимальный способ выращивани сем по- ч ки в культуре. В асептических услови х сажали на поверхность питательной среды сем почки и ставили в биологический термостат при 32° С. Волокно выращивали до 14 дней, а затем пересаживали в другие колбочки стакой же питательной смесью, но с добавлением ДНК + РНК + АТФ. Такой способ обеспечивает рост и накопление общей массы волокна, в отличие от метода Бисли, в течение 36 дней. Кроме того, введение в питательную смесь феруловой кислоты в смеси с гибберелловой обеспечивает накопление общей массы волокна более, чем в 4 раза по сравнению с контролем. Общую массу волокна определ ли то- луидиновым методом. Дл этого брали 10 сем почек, на поверхности которых сформировалось волокно, помещали в коническую колбочку на 100 мл смеси на 1 сем почку. Затем колбочку ставили на качалку и встр хивали в течение 2-х мин. После встр хивани сем почки с волокном промывали в воронке с фильтром сначала той же питательной смесью, а затем водой. После промывани сем почки переносили в колбочки с раствором толуидина синего в буфере с рН 4,5, состо щего из 10, 14 мл 0,ОШ лимонной кислоты + 9,86 мл 0,016 М Na2HP04 2НаО. Раствор толуидина наливали из расчета на каждую пробу 4 мл на каждую сем почку с волокном. Окрашивание производили в течение 15-20 с, после чего отфильтровывали и промывали осадок дистиллированной водой. Затем помещали сем почки с волокном в раствор Корнуа, налитого в колбы Эрленмейера из расчета 5 мл раствора на 1 сем почку, т. е. на 10 сем почек с волокном необходимо использовать 50 мл раствора Корнуа. Через 1 ч после помещени сем почек в раствор Корнуа измер ли оптическую плотность окрашенного раствора на СФ-4 при длине волны 640 мм. One hour after placing the seeds of the kidneys into the Cornwall solution, the optical density of the colored solution on SF-4 was measured at a wavelength of 640 mm.
