Abstract
The invention relates to medicine, in particular to immunology, and can be used to study the pathogenetic role of immune complexes in breast milk. The aim of the invention is to simplify the detection of immune complexes in women during lactation. The goal is achieved by using breast milk as a biological fluid, which is centrifuged, then treated with 2% acetic acid, which makes it possible to remove fats and casein, which significantly affect light scattering during photocolorimetry, as well as by subsequent precipitation of immune complexes in 2% solution of PEG-6000. The proposed method is quite informative, highly sensitive, easy to implement s / s
Description
The invention relates to medicine, in particular to immunology, and can be used to study the pathogenetic role of immune complexes in breast milk.
The aim of the invention is to simplify the detection of immune complexes in women during lactation due to the use of breast milk as biological fluid.
The invention is carried out as follows.
Грудное молоко предварительно центрифугируют в течение 30 мин при 6000 об/мин, а затем обрабатывают 2%-ной уксусной кислотой с последующей инкубацией смеси 2 ч при температуре 37°С (можно на ночь).Breast milk is pre-centrifuged for 30 minutes at 6000 rpm, and then treated with 2% acetic acid, followed by incubation of the mixture for 2 hours at 37 ° C (overnight).
Иммунные комплексы определ ют в надоса- дочной жидкости методом преципитации в растворе ПЭГ-6000The precipitate was removed by centrifuging for 8 hours at 8,000 rpm. Immune complexes are determined in a supernatant liquid by the method of precipitation in a PEG-6000 solution.
Diagnosis: a history of miscarriage ARVI at 22 weeks with a high temperature. The child has intrauterine infection.
Breast milk (1.5-2.0 ml) was centrifuged at 6000 rpm for 30 minutes, the fatty film was removed, and a 2% solution of acetic acid was added to 1.0 ml of a supernatant in a ratio of 1: 1, The resulting mixture was incubated for 2 hours at 37 ° C, and then centrifuged at 8000 rpm for 1 hour. The amount of 2% solution of PEG (mol.m., 6000). Samples with the addition of 0.5 ml of 0.87% sodium chloride solution are used as a control. After stirring, the samples are incubated at 4 ° C for 2 hours (overnight) and centrifuged at 6000 rpm for 30 minutes. Photometric measurements are made on a spectrophotometer (SF-26) at a wavelength of 380 nm in a cuvette with an optical path length of 1 cm. The result obtained is expressed in arbitrary units of extinction (the difference of the test and control samples, multiplied by 100). In this case, it was equal to 10 conventional units.
The proposed method was investigated 49 samples of breast milk. In the studied samples, the content of immune complexes ranged from 0 to 13 arbitrary units.
