Description
The invention relates to soil microbiology, in particular to methods for determining the effect of chemical compounds on the development of soil microorganisms, and can be used to control the side effects of pesticides on soil microflora when predicting and possibly preventing the negative effects caused by the ingestion of drugs in the soil.
The aim of the invention is to accelerate and simplify the method.
According to the proposed method, the usual soil suspension and test solutions of pesticides (in water, organic solvents) are successively introduced into the nutrient medium, followed by incubation and nephelometry of the samples.
Example. Determination of the influence of acrex pesticides and heterophos on soil bacteria.
In tubes containing 10 ml of co-peptone broth (MPB), 1 ml of soil suspension (dilution 1: 100 in sterile tap water) and 1 ml of acrex or heterophos solutions in acetone (final concentration 0.1 mg drugs on the active substance in 1 liter of the environment - really occurring concentrations of pesticides).
Став т два контрол : Ki - МПБ + водна почвенна суспензи разведени 1:100 (на интенсивность развити бактерий в отсутствие прзпарата и растворител ); Кг МПБ + + почвенна суспензи + ацетон (1 мл) (наThere are two controls: Ki - BCH + water soil suspension dilution of 1: 100 (for the intensity of the development of bacteria in the absence of the product and solvent); Kg BCH + soil + suspension + acetone (1 ml) (on

вли ние органического растворител на интенсивность развити бактерий),the effect of organic solvent on the intensity of bacterial growth),
All operations are performed under sterile conditions. Immediately after mixing the contents of the tubes, samples were taken for nephelometry, which was carried out at 540 nm in 3 mm cuvettes. The tubes are then placed in a thermostat, where they are incubated for 48 hours at 30 ° C (the temperature is optimal for the development of soil bacteria), and samples are taken for analysis after 24 and 48 hours.
Полученные данные подвергают статистической обработке и оценивают действие испытанных концентраций пестицидов по их вли нию на развитие (размножение) бактерий ,The obtained data are statistically processed and the effect of the tested concentrations of pesticides is evaluated by their effect on the development (reproduction) of bacteria,
1.The research results are summarized in table. one.
The statistically significant difference in the percent transmittance of the samples (Table 1) compared with Ki and Ka, which was observed most clearly after 48 hours of incubation, indicates that the pesticide preparation suppressed the development of soil bacteria.
П р и м е р 2. EXAMPLE 2 Comparison of the sensitivity of the proposed and known methods for determining the effect of pesticides on soil bacteria.
Результаты во всех случа х фиксируют в од- ни сроки (до 48 ч).Comparisons of the sensitivity of the proposed and known methods are carried out under identical conditions with the concentrations of acrex and pesticides Hermofos 0.1 mg / kg / l. The results in all cases are fixed in one time frame (up to 48 hours).
Определение численности бактерий провод т на м сопептонном агаре (МПА).The proposed method is used according to Example 1. In a known method, a bacterial suspension from a Bacillus megaterium culture is used, the use of which as a test culture is due to the fact that this soil bacterium is a fast-growing BCH medium. Bacteria counting is carried out on mocapton agar (MPA).
2.The research results are summarized in table. 2
According to the results given in table. 2, it can be seen that the proposed method is not inferior in sensitivity, and in a number of cases exceeds the known ones. Moreover, it differs from the prototype method in its simplicity of execution, since it does not require the isolation of the microorganism test culture into a pure culture and incubation.
Таким образом, предлагаемый способ позвол ет определ ть вли ние пестицидов на почвенные бактерии, не уступа известным способам в чувствительности, превосходит их по скорости и простоте исполнени .Thus, the proposed method allows to determine the effect of pesticides on soil bacteria, not exceeding the known methods in sensitivity, surpasses them in speed and simplicity of execution.
