Abstract
This invention relates to medicine and can be used for low temperature preservation of bone marrow hematopoietic cells for subsequent transplantation. The aim of the invention is to increase the efficiency of the method by increasing the proliferative activity of the hematopoietic bone marrow cells. To do this, the bone marrow is frozen with a cryoprotectant first from 0 to –20 ° C, then from –20 to –196 ° C, kept in nitrogen vapor and thawed in a water bath at 28–32 ° C until complete thawing of the suspension, which is diluted to a concentration of 1 . 10 5 cells / ml and cultured.
Description
This invention relates to medicine and can be used for low temperature preservation of bone marrow hematopoietic cells for subsequent transplantation.
The aim of the invention is to increase the efficiency of the method by increasing the proliferative activity of the hematopoietic bone marrow cells.
Example 1. Bone marrow was frozen with cryolrotector 15% PEO-400 according to a two-stage program: from O to -20 ° C at a rate of 1 ° C / min and from -20 ° C to -196 ° C at a speed of 300 C / min . Then the ampoules with the bone marrow were removed from liquid nitrogen and placed in nitrogen vapors, where they were exposed for 5-40 minutes at various times. After that, the ampoules were immersed in a water bath with a temperature of 30 ° C and heated for 50–60 s.
т.е. Контролем служит иативкый костный мозг, культивируемый в дозе 1 -10 кл./мл.those. until complete thawing of the suspension. The suspension was then diluted to a concentration of 1-10 cells / ml and cultured. The counting of the formed structures (clusters and colonies) in the agar was determined on the seventh and 14th day of cultivation. The control is the bone marrow cultured at a dose of 1 -10 cells / ml.
The formation of the maximum number of structures in agar not on the seventh, but on day 14 indicates the inhibition of the proliferative activity of the hematopoietic cells, the maximum number of structures in the agar is formed on the 7th day.
Example 2. The bone marrow was frozen as in Example 1, exposed in nitrogen vapor for 20 min, and then placed in a water bath with different temperatures and warmed when settled.

This is mixing. Further manipulations were carried out analogously to example 1. The control is native. Bone marrow.
The proliferative activity of the cells increased when kept in nitrogen vapor for 20-25 min and when thawed in a water bath at 28-32 ° C.
When preparing the bone marrow for transplantation using the proposed method, the maximum number of structures in the agar is formed on the seventh day, and with the known method - for 14 days, at that, the preservation of the cells does not fall.
The reduction (or complete absence) of the inhibition of the proliferative activity of the hematopoietic cells of the bone marrow ensures the possibility of timely restoration of hemopoiesis in the remission
