Abstract
The invention relates to the field of reproduction biology and can be used in studying the maturation process of female germ cells, as well as in the development of methods for cultivating and fertilizing oocytes from the ovarian follicles of the ovary outside the body in order to obtain early-stage embryos. The aim of the invention is to accelerate and simplify the method while keeping oocytes viable. After separating the oocyte, a part of the surrounding tubercle is separated from the follicle, treated with 0.8% hypotonic sodium citrate solution, then placed on a glass slide, dispersed, a drop of 1% acetorecein applied to the cells, covered with a cover glass and examined under a microscope. The presence of lipid granules in the cytoplasm of the cells of the malignant tubercle indicates atretic changes in the follicle and oocyte. with you

Изобретение относитс к биологии размножени и может использоватьс при изучении оогенеза, а также при разработке методов культивировани и оплодотворени in vitro ооцитов из п6лостнь1Х фолликулов ичника с целью получени эмбрионов ранних стадий .The invention relates to reproduction biology and can be used in the study of oogenesis, as well as in the development of in vitro cultivation and fertilization methods for oocytes from the ovary follicles of the ovary to obtain early-stage embryos.
The aim of the invention is to accelerate and simplify the method while keeping oocytes viable.
. Example. Experiments were carried out to determine atretic changes in oocytes from abdominal follicles of the ovary of a black-mongrel cow. .
Oocytes surrounded by cells of the tubercle of the mesentate are extracted from the cavity follicles of the ovary by cutting their walls. Then, under a microscope from each oocyte, using a two sharpened dissecting needles, a fragment of the tubercle of the tubercle is separated, placed for 3 minutes in a 0.8% hypotonic solution of sodium citrate, then transferred with a minimal amount of this solution onto a glass slide and dispersed dissecting needle. Then a drop of 1% acetoorsein is applied to the cells, covered with a cover glass and after 1 minute they are examined under a microscope at a magnification of 20x10 and 40x10. Short-term treatment of a fragment of the yachi beech with a hypotonic solution of
This leads to swelling of the cells and weakening of the intercellular contacts, which makes it possible to disperse the cells on the slide. The cells observe the presence or absence of lipid granules in their cytoplasm, which are a significant sign of atretic changes in the follicles.
The proposed method makes it possible to assess the presence or absence of atretic changes according to this trait and, thus, to judge the functional state of the oocyte, as well as to select oocytes for this condition for cultivation and fertilization outside the body.
