Description
The invention relates to methods of genetic selection studies. The aim of the invention is to improve the accuracy of the assessment due to the possibility of determining the ratio of biotypes in a heterogeneous material. The method is carried out as follows.
Two homogeneous varieties of wheat or biotype with gliadin alleles (blocks) are taken, the ratio of which must be studied in a heterogeneous selection sample. Flour obtained from their grain (or gliadin alcohol extracts) is shifted in the desired ratio (if you choose a 10% interval, then nine displacement options are needed and two source: sample. Apply equal amounts of extract on starch or sex acrylamide gels. The same amount is produced. electrophoresis, fixation, coloring and washing of electrophoregram from excess paint. The next day, electrophoregrams are prepared for photographing or used in natural form. The electrophoregram of a heterogeneous selection sample is compared with Scientists scale and determining the relation alleles (blocks) gliadin, from which it is composed.
П р и м е р. 2.PRI me R. It is necessary to determine the ratio of biotypes in heterogeneous breeding samples, if it is known that the original parents, respectively, included alleles for 1A chromosome GA4-1A1, and for 1B1B1-1B2.
They take homogeneous samples with these alleles, for example, grade A - Bezosta 1 (alleles 1A4 and 1B1) and grade B - Dneprovsk 521 alleles (1A1 and 1B2). Grind separately the grain of these samples, about 1000 grains (30-50 g). The resulting flour serves as the starting material for the composition of mixtures of biotypes with a known ratio. For example, you need an interval of 10%. To do this, take the following options: first A 100%,. Second A 90% B 10%, third A 80% / B 20%, fourth A 70 / B 30%, ..., eleventh

А 0%/Б 100%. A 0% / B 100%. Gliadin extract is obtained from these flour mixes and electrophoresis is carried out. Electrophograms are photographed or stored in saline.
When analyzing heterogeneous breeding samples, their electrophoregrams are compared with a scale and how many biotypes are determined by it, for example, with the 1A4 and 1A1, 1B1 and 1B2 alleles. as a fraction

белков) .the scale is also constructed in combination with other variants of gliadin alleles (or other proteins).
Вместо обычно проводимых 100 анализов дл одного гетерогенного образца необходимо всего 11 анализов .The method significantly speeds up the process of determining the ratio of biotypes in a heterogeneous selection material and reduces the cost of conducting a large number of analyzes. Instead of the usual 100 tests for a single heterogeneous sample, only 11 tests are needed.
