Abstract

FIELD: molecular biology. SUBSTANCE: method involves solving a sample of general histone sulfate received from calf thymus in hydrochloric acid adding drop-by-drop perchloric acid to reach the end concentration of 0.5 M. Insoluble H2A, H2B, H3 and H4 histones precipitate is formed. The precipitate is separated from H1 histone soluble in perchloric acid by means of centrifuging. The sample having H2A, H2B, H3 and H4 histones is solved in a solution consisting of 0.02 N hydrochloric acid, 0.05 M sodium chloride solution with pH 1.7. P-30 Acrylex chromatography is performed at the room temperature. Protein elution from the column is carried out by means of source solution. Recording the amount of protein output from the column is made in spectrophotometric way at 230 H wave length. Protein fractions corresponding to each protein peak are united, subjected to dialysis against 0.01-0.02 N hydrochloric acid at 4 C and lyophilized. Protein output from a column is 95% EFFECT: high bactericide activity of the preparation. 1 dwg
