The invention relates to medical parasitology, in particular to methods of sanitary and helminthological examination of environmental objects.
The aim of the invention is to improve the accuracy of the method.
The essence of the method is as follows.
Пробы обрабатывают в стандартных клинических пробирках , используют малогабаритную клиническзпо центрифугу ОПн-3.The entire supernatant of a flotation solution with a specific density of 1.40-1.42 is optimal for diffusion. The helminth eggs from the sediment into the supernatant and the layer-by-layer distribution of the latter, depending on their own specific gravity, are transferred to clean tubes and diluted with water, reducing the specific gravity of the solution to 1.05. After centrifugation, the resulting precipitate is concentrated and microscopic. Samples are processed in standard clinical tubes, using a small-sized clinical centrifuge OPN-3.
In this case, 6 ml of a 3% aqueous solution of potassium or sodium alkali are pre-poured into 20 clinical tubes, after which 2-2.5 g of the processed material is added to each tube. The contents of the tubes are thoroughly mixed and centrifuged, the supernatant is drained and the precipitate is washed once with water, then 3 ml of the whole salt solution, containing sodium and ammonium nitrate with a specific density of 1.40, is added to the precipitate, the contents are mixed and centrifuged . The resulting supernatant is poured into clean tubes.
To the transferred solution of flotation liquid, a certain amount of water is added to each tube, the tubes are shaken and centrifuged, after which the supernatant is drained, and the resulting precipitate from 20 tubes is concentrated in 4 To the precipitate, each layer is layered with ether. The tubes are shaken and centrifuged, the supernatant layer is drained, and the precipitate is microscoped.

The reagents are mixed with water in the following proportion: 800 g of NaNO, j + 400 g of NH NOj + ln, which corresponds to the molar ratio of 9.4: 3, respectively, and brought to a boil, remove scum with a spoon. The solution is boiled over low heat for 10-15 minutes, after which it is cooled and the specific gravity is measured with a hydrometer, which should be 1.40-1.42. The solution is stored at room temperature in closed containers for 7-10 days.
Processing soil samples.
In 20 clinical centrifuge tubes with a volume of 10 ml, 6 ml of a 2% solution of caustic soda are drunk. Then, the amount of soil is added to each tube so that the volume of the solution poured into the tube increases to 7 ml. Such an increase of 1 lll corresponds to a soil weight of 2 to 2.5 grams. In the study of samples in 20 clinical tubes, the total weight of the sample will be from 40 to 50 grams. Glass tube thoroughly mix the contents of the tubes, after which the tubes are centrifuged at 1500 rpm for 3 minutes. Subsequently, the centrifuging mode is kept constant.
0After centrifugation, the supernatant was decanted, and 8 ml of water was added to the precipitate. The contents are thoroughly mixed and centrifuged. 5 The supernatant is discharged, and 3 ml of a solution containing sodium and ammonium nitrate are added to the precipitate at a molar ratio of 9.4: 5, respectively. In this case, the specific density of the solution 1.40-1.42 provides for the transfer of the helminth eggs from the sediment to the supernatant layer of the liquid. Eggs with a specific gravity of less than 1.40-1.42 are floated, into a surface film, and with a specific density of more than 1.40 are distributed throughout the entire column of supernatant. The contents of the tubes are mixed and centrifuged. The centrifuged tubes are left at rest for 5 minutes, after the supernatant is poured into the next clean row of tubes (one to one), and then they are only used with it. To the transferred solution of flotation liquid, water is added to each tube to the 10 ml mark. In this case, the specific density of the solution pa0

The tubes are shaken and centrifuged at 1500 rpm for 5 minutes. This technique allows the concentration of helminths in the sediment. The supernatant was drained, and the sediment from each 10 tubes was transferred to 2 of them. The pellet is collected with a Pasteur pipette. Thus, out of 20 test tubes, only 4 remain, in which all the sediment containing the helminth eggs is concentrated.
To each tube, 7 ml of water are added to the sediment and 1 MP of ether is layered on top. The tubes are shaken vigorously for 30-40 seconds and centrifuged at 1500 rpm for 3 minutes.
Then the supernatant layer of the liquid is drained, the walls of the tube are wiped with a cotton swab, the tube is in a horizontal position.
