Description
For the manufacture of the vaccine, 8 virulent and immunogenic strains of pasteurellosis isolated from cattle, sheep and pigs are taken. As a mat seed, all strains are grown for a day on the Hottinger broth individually, and then seeded all together in one 500-liter tanker designed by visitors, where they are grown for 48 hours during aeration with sterile air. In the process of growing in a tanker, after 24 hours, peptone water is added to the culture, bringing the nenton to 1% relative to the volume of the nutrient medium. After 48 hours A 10% solution of alum is added to the grown culture at the rate of 20 ml per 1 l of the culture and, after the culture precipitates, 0.2-0.25% formalin.
A culture in a tanker with alum added to a pH of 7.2-7.4 is kept at a temperature of 38 ° for 24 hours and then packaged directly from the tanker into vials.
The vaccine is tested for sterility, harmlessness and immunogenic properties (in blue).
The vaccine is administered to animals subcutaneously, twice, in doses of: at the first vaccination of cattle, adult sheep and pigs, 5 ml each, and 3 ml of pigs and piglets there; with re-vaccination, after 10 days, in doses: cattle, 10 ml each, sheep and pigs, regardless of age, 8 ml each.
The in-depth method simplifies the manufacturing processes for the manufacture of a vaccine (the period of crop cultivation is significantly reduced, there is no need to use a large amount of glass cylinders for the production of a culture medium and culture).
