Известен способ экстракорпоральной детокси кации организма путем использовани взвеси изолированных клеток печеии, отделенных от крови полупроиицдемш мембраной 1. Однако .известный способ не всегда и не везде может быть использован в силу своей сложности, об зательного наличи сложного оборудовани , т. е. центров, где имеютс annaiраты искусственна почка, перитонеального диализа. Кроме того, не всегда возможно провести вмешательство на брюшной полости (это перитонеальш ш диализ), так как имеетс спаечный процесс в брюшной полости, которые ограш1чивак т возможность проведени этой операции. Значит перитонеальный диализ имеет свои ограничени и не всегда может быть .использован дл лечени больных с острой печеночнсж недостаточностью. При и щользовании этгих способов клеточна взвесь аллогенной печени примен етс всего лишь раз и затем выбрасываетс . Поэтому способ требует большо го количества заготовки биологического матери ала и запаса клеточной взвеси. Цель изобретени - повторное использование клеток печени. Указанна цель достигаетс тем, что согласно способу экстракорпоральной детоксикаоди орга низма путем использовани взвеси изолированных клеток печени, отделеннь1х от крови полупроницаемой мембраной, взвесь изолированных клеток печени и питательной среде помещают в замкнутые оболочки из полупроницаемой мембраны объемом 1,0-1,2 см , сформированные гранулы внос т в колонку, заполненную кровезаменителем, пропускают через нее кровь, по окончании перфузии гранулы отмывают Ч помещают в питательную среду дл хранени . Способ осуществл ют следующим образом. Взвесь изолированных клеток печени (полученную ферментно-механическим способом) развод т в среде 199 до концентрации 210 (20 млн/мл), заполн ют ими .с.точной градуировкой и системой гибких сили коновых . Одновременно-Провод т подго товку полупроницаемой целлофановой пленки, рассекают ее на определенной формы квадраты, которые первоначально помещают в физиологический раствор, а затем перенос т в раствор диоцида .1:2000 с последующим промьшанием дастиллированной водой. Участок пленки как бы надевают на конец силиконового шланга и, отступив на 1 см от его конЦа, накладьшают лигатуру, после чего в эту полость ввод т 1 м клеточной взвеси и, зат гива шгатуру, удал ют силиконовый шланг. Избыток пленки и шовHojro материала удал ют. Готов т 250-300 гранул и помещают их во флакон со средой 199. Производ забор необходимого количества гранул, заполн ют ими колонку, через которую провод т перфузию крови. Дл этих целей вьщел ют сосуды (артерию и вену) и катетеризируют дл формировани артериовенозного шунта. Колонку с гранулами подключают к шунту таким образом. Чтобы артериальна кровь в колонку поступала снизу вве|рх, заполн ее на всем прот мсенди, и выходила через верхнюю канюлю, котора соедин етс с венозной системой. Перфузию продолжают в течение 2 ч со скоростью 100-120 мл/мин. Затем колонку отключают от шунта и кровь возвращаетс в систему. После этого осуществл ют промывку колонки и гранул и последние помещают на 1-2 сут в среду 199. Этим самым П) создаем все услови дл восстановлени их запасов и ф)нкций, так как они наход тс в культуральНйй среде, где есть все необходимые метаболиты.дл их восстановлени и обеспечени жизнеспособности. Следовательно, клет-. ки могут восстановить свой метаболический балансИ могут быть использованы повторно дл детоксикации крови. Поэтому процессы диффузии и осмоса, ультрафильтрации будут осуществл тьс между гранулами- и средой зна штельно быстрее , чем Б системе аппарата Искусственна почка. После сохранени клеток в среде 199. их вновь помещают в колонку, последшою подключают к артериовенозному шунту и провод т перфузию в течение 1,5-2,0 ч. Затем колонку и гранулы промывают раствором Хенса и последние помещают в среду 199. Через 1-2 сут Гранулы вновь используютдл операии детоксикации организма.
