Abstract

Aspergillus niger CCM 8210 microorganism is usable for the preparation of citric acid of high purity. The productivity provides a yield of 90 - 93 kg of citric acid per 100 kg of sugar.

Description

Príklady uskutočnenia vynálezuDETAILED DESCRIPTION OF THE INVENTION

Príklad 1Example 1

Pripraví sa hydrolyzát čistého kukuričného škrobu s dextrózovým ekvivalentom DE = 15 %, z ktorého sa pripraví kultivačné médium. 1 liter média obsahuje 180 g cukru, 2,8 g síranu amónneho, 0,3 g dihydrogenfosforečnanu draselného, 0,15 g chloridu vápenatého a 0,08 g heptahydrátu síranu zinočnatého. Sterilné kultivačné médium s pH 2,7 až 2,8 sa v 1001 fermentore zaočkuje s 2 až 3 % obj. 40 až 48 hodinového inokula kmeňa Aspergillus niger CCM 8210 a kultivuje sa pri teplote 32 až 33 °C, obvodovej rýchlosti miešadla do 5.2 m.s1 a vzdušnení 0,2 až 0,35 VVM počas 155 až 160 hodín. V priebehu fermentácie sa vytvorí v médiu 165 g.ľ1 kyseliny citrónovej.A pure corn starch hydrolyzate with a dextrose equivalent of DE = 15% is prepared from which a culture medium is prepared. 1 liter of medium contains 180 g of sugar, 2.8 g of ammonium sulfate, 0.3 g of potassium dihydrogen phosphate, 0.15 g of calcium chloride and 0.08 g of zinc sulfate heptahydrate. A sterile culture medium of pH 2.7 to 2.8 is inoculated with 2 to 3% v / v in a 100 L fermenter. 40 to 48 hour inoculum of Aspergillus niger strain CCM 8210 and cultured at 32-33 ° C, stirrer peripheral speed up to 5.2 ms 1 and aeration 0.2-0.35 VVM for 155-160 hours. 165 g / l of citric acid are formed in the medium during the fermentation.

Príklad 2Example 2

Pripraví sa hydrolyzát kukuričnej múky s dextrózovým ekvivalentom DE = 40 %, z ktorého sa pripraví kultivačné médium. 1 liter média obsahuje 180 g cukru, 2,0 g dusična-Corn flour hydrolyzate is prepared with a dextrose equivalent of DE = 40% from which the culture medium is prepared. 1 liter of media contains 180 g of sugar, 2.0 g of nitrogen-
