elektroforesen, trots de reaktioner som sker vid respektive
elektrod. De viktigaste faktorerna som bestämmer buffert-
kapaciteten är valet av buffertkomponenter, koncentrationen
av dessa samt buffertlösningens volym.
Traditionellt genomföres elektroforesen som nämnts med
utrustningar som har speciella bufferttankar, vilka fylls
med önskad volym av respektive buffert. Det är emellertid
svårt att använda lösningar med hög koncentration vid vissa
applikationer p g a de vätskeströmningar som uppstår genom
kapillärkrafterna bl a i kanterna på gelmatrisen. Genom att
hålla en låg elektrodbuffertkoncentration kan man undvika
störningar eller rent av kortslutning i separationsgelens
elektriska fält på grund av sådan vätskeströmning. Låg
elektrodbuffertkoncentration kan kompenseras genom en ökning
av buffertvolymen vilket emellertid förutsätter att elektro-
foresutrustningen har tillräckligt stora buffertkärl.
Speciellt i sådana system där man arbetar med förhållandevis
små gelmatriser, t ex i mer eller mindre automatiserade
system, innebär hanteringen av buffertlösningar praktiska
pproblem och det föreligger ett behov av att minska hante-
ringen av vätskevolymer så mycket som möjligt.
I US37l5295 har man identifierat detta problem och föreslår
att bufferten blandas med någon substans som gör att man får
bufferten i "halvfast" form. En föredragen utföringsform
innebär att bufferten inkorporeras i silikapartiklar var-
efter en halvfast substans tillföres. Oberoende av vilken
"halvfast" form som väljes, överföres en lämplig volym av
denna till en fördjupning i vardera ändan av elektrofores-
utrustningens stödplatta varefter separationsgelen placeras
på plattan så att det blir direkt vätskekontakt mellan
separationsgel och buffert. Det skall observeras att ut-
rustningen i detta fall, liksom ovan nämnts, kräver sär-
skilda buffertbehållare. Huvudsakliga skillnaden jämfört med
det normala förfarandet är att bufferten ej utgöres av en
lösning. Värt att notera i sammanhanget är att, trots att
detta patent publicerades redan 1973, har metoden ej kommit
att utnyttjas i någon väsentlig grad.

Vi har nu funnit att ett betydligt förenklat förfarande
uppnås om buffertsubstansen inkorporeras i ett gelmaterial,
som placeras i kontakt med separationsmatrisen vid respek-
tive ände, varefter elektroderna av traditionell typ anslu-
tes till buffertgelerna. Speciellt fördelaktigt är förfaran-
det vid horisontell elektrofores då den nya tekniken möjlig-
gör att en buffertreservoar i form av ett gelstycke helt
enkelt placeras ovanpå separationsmatrisen vid respektive
ände. Provet sättes därefter på separationsgelen, och sedan
det elektriska fältet lagts på, vandrar provkomponenterna i
detta, i det utrymme av separationsgelen som avgränsas genom
de två buffertgelerna. Analogt kan buffertgelerna användas
även vid andra typer av elektrofores genom att de bringas i
kontakt med motstående ändar av matrisen.
Till fördelarna med tekniken enligt föreliggande uppfinning
hör att elektroforesutrustningen ej behöver innehålla kärl
för flytande eller halvfast elektrodbuffert, vilket gör
utrustningen enklare och vidare att de buffertgeler som
används vid utövandet av uppfinningen är synnerligen enkla
att handskas med, dvs att de har en god mekanisk häll-
fasthet. Genom att elektrodbufferten är ingjuten i "fast"
gel förekommer ingen vätsketransport p g a kapillärkraf-
terna. Detta gör det möjligt att använda betydligt högre
koncentrationer av elektrodbuffert än vad som annars skulle
vara möjligt, vilket medför att buffertvolymen kan hållas
låg. En annan fördel med "fast" gelbuffert är att elektrod-
reaktionernas produkter hindras att nå separationsgelen där
de skulle kunna tänkas påverka resultatet. Vid elektrodreak-
tionerna förändras buffertsammansättningen nära elektrodytan
och under elektroforesens gång förändras buffertlösningen
allt längre ut från elektrodytan. I Fig l àskádliggörs detta
förlopp under en elektroforetisk separation.
(l) är separationsmatrisen i form av en gelplatta. De två
elektrodbuffertinnehállande gelerna (2) och (3) är placerade
i kontakt med separationsmatrisen vid två motsatta ändar av

denna. De två elektroderna (4) och (5) har direktkontakt med
buffertgelerna (2) respektive (3). Den elektroforetiska
separationen påbörjas när ett elektriskt fält appliceras
mellan elektroderna och den förändring i elektrodbuffertarnas
sammansättning (skuggat omrâde) som sker på grund av elektrod-
reaktionerna åskådliggörs i fig 1 a-c, vilka visar status
vid 0,25 respektive 100 % av totala elektroforestiden.
Gelmaterialet för användning som stabilisator för elektrod-
bufferten väljes ur den stora grupp av i och för sig kända,
och åtminstone vid rumstemperatur, vattenolösliga polymerer,
vilka skall uppfylla kriteriet att ha låg egenladdning.
Detta är en förutsättning för att gelmatrisen ej skall
fungera som en s k elektroendosmotisk pump. Exempel på
lämpliga material är naturligtvis de agaroskvalitêer som
utvecklats speciellt för elektroforetiska tillämpningar och
vilka uppfyller nämnda kriterium, t ex Agaros IEF (Pharmacia AB,
Uppsala, Sverige). Koncentrationen av agaros i den komposi-
tion som utgöres av buffertinnehållande vattenhaltig gel
ligger lämpligen i intervallet 1 - 4 viktsprocent, förse
trädesvis omkring 2 %. Andra material som ligger nära till
hands att använda är polyakrylamid med en halt av omkring
10 % och en tvärbindningsgrad som ligger i intervallet
2 - 7 %.
De buffertar som inkorporeras i gelmaterialet är sådana som
normalt användes vid elektroforetisk separation. Exempel på
sådana lösningar är t ex en blandning av trishydroxymetyl-
aminometan (TRIS), glycin och saltsyra; alanin; ättiksyra;
och olika natriumfosfater med tillsats av natriumdodecyl-
sulfat (SDS). Lämpliga koncentrationer av de ovan nämnda
liksom ytterligare exempel återfinnes i Polyacrylamide Gel
Electrophoresis, Laboratory Techniques (Pharmacia AB,
Uppsala. Sverige). Innehållet av elektrodbuffert i respek-
tive gelstycke ligger vanligen i intervallet 80 - 99 vikts-
procent.

En övre gräns för koncentrationen av buffertkomponenter i
gelmaterialet ges av mättnadsgränser i vattenlösning för
dessa komponenter, samt av att det i vissa fall sker
oëpskade reaktioner som t ex utsaltning av gelmatrisen.
Detta sker i fallet agaros vid hög koncentration av
buffertsalter. En undre gräns för koncentrationen av
buffertkomponenter i gelmaterialet kan bestämmas genom
metodologiska studier, t ex mätning av pH i bufferten vilket
visar när dess buffertkapacitet ej längre räcker till. Dessa
gränser för olika kombinationer av gelmaterial och
buffertkomponenter kan fackmannamässigt utprovas i varje
enskilt fall.

En buffertlösning för nativ gradientgel elektrofores be-
reddes genom att 15 gram trishydroxymetylaminometan (TRIS)
och 65 gram alanin löstes i 500 ml vatten. 10 gram agaros
tillsattes och bufferten kokades under omrörning tills
agarosen löst sig. Lösningen fylldes på formar med måtten
40 x 10 x 6 mm och ställdes att svalna. Sedan gelbufferten
stelnat blir den med denna dimension lämplig att applicera
på separationsgeler med ungefärliga dimensionen 40 x 40 x
0,3 mm som exempelvis innehåller en 0,026 M TRIS-, 0,033 M
ättiksyra-buffert.
Exemgel 2
En buffertlösning för SDS gradientgel elektrofores bereddes
genom att 12,1 gram trishydroxymetylaminometan (TRIS),
17,9 gram tricin och 2,8 gram natriumdodecylsulfat (SDS)
löstes i 500 ml vatten. 10 gram agaros tillsattes och
bufferten kokades under omrörning tills agarosen löst sig.
Lösningen fylldes på formar med måtten 40 x 10 x 6 mm och
ställdes att svalna. Sedan gelbufferten svalnat är den
färdig för applicering på separationsgeler med dimensioner
enl ovan.

Förfarande enligt krav 2 k ä n n e t e c k n a t a v att innehållet av elektrodbuffert i respektive gelstycke ligger i intervallet 80 - 99 viktsprocent.
