Abstract
1,152,856. 17α - (1<SP>1</SP>,3<SP>1</SP> - Alkadiynyl) - 17#- acyloxy steroids. BRITISH DRUG HOUSES Ltd. 9 May, 1968 [6 June, 1967], No. 26049/67. Headings C2C and C2U. Novel steroids having in the 17-position the structure (wherein R is C 1-5 alkyl and R<SP>1</SP> is acyl of at most 20 carbon atoms) are prepared by acylation of the corresponding free 17#-ols with the appropriate acid chloride or anhydride. Other groups in the molecule may also undergo reaction during the acylation, e.g. hydroxy groups may be esterified and keto groups enol esterified, but they may be regenerated selectively by controlled hydrolysis. Alternatively, such groups may be protected before acylation, e.g. as ketals, enol ethers or tetrahydropyranyl ethers, and regenerated by hydrolysis subsequently. (1.Adamantyl)-acetic anhydride is prepared from (1-adamantyl)-acetic acid and acetic anhydride. The 17α - (1<SP>1</SP>,3<SP>1</SP>- alkadiynyl) - 17# - acyloxysteroids of the invention, which are stated to have estrogenic, progestational, claudogenic, ovulation - inhibiting and gonadotrophininhibiting properties, may be incorporated in pharmaceutical and veterinary compositions, e.g. tablets, injections, vaginal sponges and tampons.
