Abstract

A method for determining whether , beta-lactamase activity is present in a prokaryotic cell is provided, which comprises the steps of: providing a fluorescent substrate for , beta-lactamase; providing a prokaryotic cell which comprises a gene encoding a heterologous esterase enzyme, which enzyme has the ability to convert the substrate from a diffusing form to an active form; applying said substrate, in its diffusing form, to said prokaryotic cell, said application enabling said heterologous esterase enzyme to convert the substrate to its active form; culturing said cell under conditions such that any beta-lactamase activity present in the cell cleaves said substrate, yielding a fluorescent signal; and detecting fluorescence from said cell.
