Abstract

A method for determining concentration of free thrombin in a sample as a function of time uses a substrate that reacts with thrombin to form a product. A physical property of the product, such as optical absorbance or fluorescence emission, is measured as a function of time. A time derivative of the property is computed, preferably using a time averaging technique, to determine a rate of reaction from substrate to product. The time derivative is then used to determine the concentration of alpha2MT as a function of time and the time derivative of the concentration of alpha2MT used to determine a concentration of free thrombin. The computation minimises the effects of variability in the alpha2MT -dependent decay parameter, k and the parameter kcat2 expressing a rate of turnover of substrate by alpha2MT.
