Abstract

FIELD: microbiology, molecular biology. SUBSTANCE: DNA of brucelli contains the unique repeated nucleotide sequences showing complementarity with that of synthetic oligonucleotide /CAC/.. Hypervariable sequences can be detected by digestion of brucella DNA with restriction enzyme NcoI followed by elecetrophoresis and hybridization with DNA-probe /CAC/.. Bands pattern (sites of unique nucleotide sequence) are specific for different brucella strains that ensures to differentiate thereof. EFFECT: increased safety and specificity of method for identification and differentiation of brucelli.

Изобретение относится к области микробиологии, молекулярной биологии и касается способа дифференциации различных штаммов бруцелл вида Вrucel1а аbоrtus. Целью изобретения является повышение надежности и специфичности способа идентификации и дифференциации различных вакцинных штаммов бруцелл и возбудителя бруцеллеза крупного рогатого скота. Сущность изобретения состоит в том, что ДНК бруцелл содержит в своем составе уникальные повторяющиеся последовательности нуклеотидов. Они комплементарны к последовательности синтетического олигонуклеотида (САС) 5. Гипервариабельные последовательности обнаруживаются рестрикцией ДНК бруцелл ферментом Nсоl, электрофорезом их и гибридизацией ДНК-зондом (САС) 5.
