Abstract

FIELD: medicinal biochemistry. SUBSTANCE: method involves sample taking off, separation of serum, supernatant chromatography at reversed-phase elution in isocratic regime with 4% methanol solution in acetic acid and detection in UV at 290 nm. Content is determined by areas peaks. Blood serum is treated with isopropanol at ratio 1:5 at stirring. Chromatography is carried out by gradient elution, firstly with mixture acetonitrile/isopropanol at ratio 1:1, detection - at 210 nm and then with methanol solution in 1% acetic acid. Identification is carried out by retention time values. EFFECT: enhanced sensitivity, decreased analysis time. 1 dwg

Claims (1)

Способ относится к области медицины, а именно - к медицинской биохимии. Техническим результатом предложенного способа является повышение чувствительности и сокращение времени анализа. Способ определения содержания атерогенных компонентов в сыворотке крови включает взятие пробы, отделение сыворотки, хроматографирование супернатанта в обращенно-фазном режиме элюирования в изократическом режиме 4 %-ным раствором метанола в уксусной кислоте при УФ-детекции 290 нм и определение содержания по площадям пиков. Новым в способе является обработка сыворотки крови изопропанолом в соотношении 1 : 5, встряхивание. Отличия способа заключаются в хроматографировании в режиме градиентного элюирования первоначально смесью ацетонитрила с изопропанолом в соотношении 1 : 1 при 210 нм, а затем раствором метанола в 1 %-ной уксусной кислоте. Идентификацию проводят по временам удерживания.The method relates to medicine, namely to medical biochemistry.
