BACKGROUND OF THE ART
Liver disease is a type of disease that is widespread throughout the world, and China is a country that experiences a high incidence of liver disease. Currently, in China there are many patients with liver lesions and inflammation caused by various causes, mainly viral hepatitis. According to statistics, the direct economic loss per year from chronic hepatitis (including liver cirrhosis and liver cancer caused by late-stage chronic hepatitis) in China reaches 900 billion yuan. Over the past few years, there has also been a year-on-year increase in the incidence of drug-induced liver disease, alcoholic and non-alcoholic fatty liver disease, and autoimmune liver disease. Finding safe and effective drugs for the prevention and treatment of liver diseases is always an extremely important research topic for various research institutes and pharmaceutical companies around the world.
(Institute of Materia Medica Chinese Academy of Medical Science), Bicyclol is a type of first-line chemical medicine for the treatment of hepatitis in China, protected by independent intellectual property rights in China, which was first developed by the Institute of Materia Medica Chinese Academy of Medical Science, and when used in the clinic, it has a number of advantages due to its high efficiency in protecting the liver and enzymatic degradation, certain activity against the hepatitis virus, a convenient route of administration without any significant adverse reactions and broad pharmacological activity. Bicyclol-related scientific achievements have been consistently recognized year after year with numerous awards, including the "Achievement Award for Achieving National Key Challenges in Science and Technology in the Ninth Five-Year Plan" (2002), "Ten Most Important Events in the Chinese Pharmaceutical Industry in 2002", Beijing Science and Technology Progress First Prize (2005), National Science and Technology Progress Second Prize (2007) and other similar awards, and the patent rights for the compound bicyclol are protected in sixteen countries/regions, such as the United States, European Union, Japan, Korea and other countries, and in the Taiwan region, and it is sold to countries such as Ukraine. Since the day bicyclol was announced on the pharmaceutical market at a press conference held at the Great Hall of the People in 2001, it has brought enormous social and economic benefits to China.
(Institute of Materia Medica Chinese Academy of Medical Science) (Wu Song, Sun Hua, et al., Bicyclol Derivative and Preparations and Applications Thereof, 201610922563.5). However, bicyclol has insufficient biological solubility in water, as a result of which it does not have high bioavailability. Researchers from the Institute of Materia Medica Chinese Academy of Medical Science optimized the structure of bicyclol and found that bicyclic morpholine and its salts have good pharmacological activity and pharmacokinetic properties (Wu Song, Sun Hua, et al., Bicyclol Derivative and Preparations and Applications Thereof, 201610922563.5). Based on previous results, the researchers isolated the racemate of bicyclic morpholine and found that levorotatory bicyclic morpholine and its salts had better pharmacological activity and pharmacokinetic properties in terms of anti-inflammatory effects and liver protection than the racemate and dextrorotatory morpholine and its salts.

91,7%.Compound 1 (ie bicyclol, 5.1 g, 13.1 mmol) was charged into a 100 mL three-neck flask equipped with a magnetic stirrer and thermometer, then 50 mL of dried DMF was added and the solids were completely dissolved. After cooling the reaction system to 0°C in an ice bath, SOCl 2 (4.5 mL, 61.8 mmol) was slowly added dropwise while ensuring that the system temperature did not exceed 5°C. After addition dropwise, the reaction mixture was kept in an ice bath for 30 min until thin layer chromatography (TLC) analysis indicated complete conversion of the starting materials. The reaction mixture was poured into approximately 100 g of crushed ice and stirred vigorously to precipitate a large amount of white solids, which were filtered, and the filter cake was washed with small quantities of distilled water and ether, and dried by draining. The product was naturally air dried and weighed to give a total of 4.9 g of white solids (compound 2), yield: 91.7%.
92,3%. MS-FAB [M+H]+=460,1.Morpholine (1.28 g, 14.7 mmol) was charged into a 50 ml round bottom flask and then 25 ml of acetone and 2.2 ml of triethylamine were added with stirring at room temperature, then compound 2 (2.76 g, 6.8 mmol). The reaction was carried out at room temperature for 5 hours and the reaction mixture was further kept overnight. TLC analysis indicated complete conversion of the starting materials and pink insolubles were formed in the system. After filtration, the filtrate was distilled off under reduced pressure to remove the solvent. The resulting yellow oil was separated by a vacuum distillation column (petroleum ether:ethyl acetate=2:1), and the product components were collected to give a total of 3.1 g of colorless oil (compound 3), yield: 92.3%. MS-FAB [M+H] + =460.1.
98,0%, [α]25= -130,4 (CH2Cl2). Compound 3 (3.0 g, 6.54 mmol) was weighed and dissolved in 45 ml ethyl acetate and L-dibenzoyltartaric acid (L-DBTA, 1.2 g, 3.27 mmol) was added. At room temperature, the mixture was stirred to precipitate white solids, and after 30 minutes, filtration was performed to collect the solids, which were then dried at 60°C for 1 hour and then weighed, yielding a total of 1.25 g of white solids (levorotatory bicyclic morpholine:L-dibenzoyltartaric acid=2:1), yield: 56.8%. Chiral high performance liquid chromatography (HPLC) analysis indicated that the enantiomeric purity (%ee) of intermediate 4 was 98.0%, [α] 25 = -130.4 (CH 2 Cl 2 ).

4. Experimental results
ConA 4.1. Effects of IMM-H014 Enantiomers on Increased Serum ALT Biomarker Associated with ConA-Induced Liver Injury
По сравнению с (±) IMM-H014 в той же дозе, снижающие воздействия (+) IMM-H014 и (-) IMM-H014 на вызванное ConA повышение уровней АЛТ в сыворотке мышей превосходили воздействия (±)IMM-H014 (который также проявлял значительную эффективность, а процент снижения АЛТ составлял 88,6%), при этом снижающие воздействия (-)IMM-H014 на АЛТ имели статистически значимое различие (P<0,05) по сравнению с воздействиями (±)IMM-H014.Serum ALT levels directly and positively correlate with the degree of liver injury and are a well-established serum biomarker of liver injury. The results in Table 1 showed that administration of ConA at a dose of 20 mg/kg caused significant liver damage in mice, and serum ALT levels were significantly increased compared with the control group (P<0.001). Administration of both (+)IMM-H014 and (-)IMM-H014 significantly reduced ConA-induced increases in serum ALT levels (P<0.001). The percentage reduction in ALT when exposed to IMM-H014 enantiomers was 95.3% and 97.3%, respectively, and ALT contents were reduced to the level of the control group. Both (+)IMM-H014 and (-)IMM-H014 showed significant protective effects against ConA-induced immune liver injury, and the activity of (-)IMM-H014 was slightly higher than that of (+)IMM-H014. Compared with (±)IMM-H014 at the same dose, the reducing effects of (+)IMM-H014 and (-)IMM-H014 on ConA-induced increases in serum ALT levels in mice were superior to the effects of (±)IMM-H014 (which also exhibited significant efficacy, and the percentage reduction of ALT was 88.6%), while the reducing effects of (-)IMM-H014 on ALT had a statistically significant difference (P<0.05) compared with the effects of (±)IMM-H014.

ConA4.2. Effects of IMM-H014 Enantiomers on Increased Serum AST Biomarker Associated with ConA-Induced Liver Injury
An increase in the level of AST in the blood serum is also one of the important markers of damage to hepatocytes, in particular, damage to the mitochondria of hepatocytes, and in the case of mitochondrial damage, the level of AST in the blood serum increases significantly, thereby reflecting the severity of the damage to hepatocytes. The results are presented in Table 2. Administration of ConA at a dose of 20 mg/kg caused significant damage to the mitochondria of mouse hepatocytes, and serum AST levels were statistically significantly increased compared with the placebo control group (P<0.001). All enantiomers of (+)IMM-H014, (-)IMM-H014 and (±)IMM-H014 exhibited the ability to reduce ConA-induced increases in serum AST levels. Compared with the model group, the percentage of AST reduction in the (+)IMM-H014, (-)IMM-H014 and (±)IMM-H014 administration groups was 44.8%, 72.9% and 22.2%, respectively. while the (-)IMM-H014 group was characterized by a statistically significant difference (P<0.01) compared to the model group. The activity of (+)IMM-H014 and (-)IMM-H014 in reducing AST was superior to the activity of (±)IMM-H014, and the activity of (-)IMM-H014 was optimal and was characterized by a statistically significant difference (P<0.05) compared with group (±)IMM-H014 at the same dose.

ConA4.3. Effects of IMM-H014 Enantiomers on Increased Serum LDH Biomarker Associated with ConA-Induced Liver Injury
In liver injury, serum LDH levels may also reflect the status and extent of hepatocyte injury. The results are shown in Table 3. Administration of ConA at a dose of 20 mg/kg by tail vein injection caused severe damage to hepatocytes, and serum LDH levels were statistically significantly increased compared with the placebo control group (P<0.001). Administration of (-)IMM-H014 significantly reduced the elevated serum LDH level induced by ConA, and the percentage of LDH reduction was up to 54.4%, which was statistically significant difference (P<0.01) compared with the model group . Administration of (+)IMM-H014 also had a reducing effect on elevated serum LDH levels, and the reduction percentage was 27.6%, which was not statistically significantly different compared with the model group. Administration of (±)IMM-H014 at the indicated dose had only a weak reducing effect on elevated LDH levels, and the percentage reduction was 6.4%. Regarding LDH reduction, the activity of (-)IMM-H014 was still superior to that of (+)IMM-H014 and (±)IMM-H014, and there was a statistically significant difference (P<0.05) between (-) IMM-H014 and (±)IMM-H014 at the same dose.

2. Determination of triglyceride and cholesterol content in liver tissues
Mice underwent thoracotomy to remove the liver, which was washed with saline at 4°C. A portion of the liver tissue was processed into a 10% liver tissue homogenate using a lysate and an electrical homogenizer, after which one portion of the homogenate was used to measure its triglyceride and cholesterol content using a triglyceride test kit and a cholesterol test kit, and the other portion of the homogenate was analyzed for determination of protein content using the BCA protein quantitation kit. The content of triglyceride and cholesterol in liver tissues was corrected taking into account protein content.

4.1. Effects of IMM-H014 enantiomers on ethionine-induced increases in cholesterol (TC) content in mouse liver tissue
Активность (-) IMM-H014 была немного выше, чем активность (±) IMM-H014.Ethionine, due to the fact that it interferes with the metabolism of methionine and additionally affects the synthesis of apolipoprotein, can lead to the fact that cholesterol, triglyceride and other similar substances synthesized in hepatocytes cannot be transferred into the blood, which causes the accumulation of lipids in hepatocytes, and as a result A model of non-alcoholic fatty liver hepatosis caused by drug exposure is being formed. The results are presented in Table 4. Ethionine at a dose of 250 mg/kg can cause a statistically significant increase (P<0.05) in cholesterol content in liver tissue compared to the placebo control group. Administration of both (+) IMM-H014 and (-) IMM-H014 reduces ethionine-induced accumulation of TC in liver tissue, and the percentage reduction in TC when exposed to enantiomers of IMM-H014 (i.e. (+) IMM-H014 and (- )IMM-H014) were 27.2% and 32.4%, respectively, with the (-)IMM-H014 group having a statistically significant difference (P<0.05) compared with the model group. (±)IMM-H014 also had a statistically significant inhibitory effect on ethionine-induced increase in TC in liver tissue, and it had a statistically significant difference (P<0.05) compared with the model group. The activity of (−) IMM-H014 was slightly higher than that of (±) IMM-H014.

Степень выживаемости клеток (%)=(средняя величина OD в группе введения/ средняя величина OD в контрольной группе с растворителем) ×100%.The MTT method was used. HepG 2 cells were inoculated in a 96-well cell culture plate, and after 24 hours of culture, non-toxic concentrations of (+)IMM-H014, (-)IMM-H014 and (±)IMM-H014 were added, and at the same time 4 -acetamidophenol (APAP, final concentration 8 mM). To conduct the experiment, a positive control group with bicyclol, a control group with a solvent, and a model group were formed. Cultivation was continued for 24 hours. 100 μL of the culture solution was collected and centrifuged, and then the LDH level was determined on a fully automatic chemistry analyzer using an LDH detection kit. The remaining culture solution was discarded, and 100 μL of MTT solution (0.5 mg/mL) was added to each well to continue culture for 4 hours. The MTT solution was discarded and 150 μL DMSO was added to each well. The mixture was stirred using a vibrator, and the absorbance value (OD) was measured at a wavelength of 570 nm in a microplate reader. Cell survival rate (%)=(average OD value in the injection group/average OD value in the vehicle control group) ×100%.

LDH (lactic dehydrogenase) is one of the important enzymes in cellular energy metabolism. When cells die, the cell membrane ruptures and LDH is released from the cytoplasm, with the level of LDH being proportional to the degree of cell damage. The results are shown in Figure 1. A study was conducted on the effect of 8 mM APAP on HepG2 cells for 24 hours, and it was found that the level of LDH in the cell culture supernatant was statistically significantly increased compared to the placebo control group (P<0.01) , which also indicated significant damage to hepatocytes. Exposure to (+)IMM-H014, (-)IMM-H014 and (±)IMM-H014 at a dose of 10 µM reduces LDH levels, while LDH levels in the case of (-)IMM-H014 and (±)IMM-H014 had statistically significant differences (P<0.05, P<0.05) compared with the model group, and the reducing effect in the case of the (-)IMM-H014 group was slightly higher than that of the (±)IMM-H014 group at that same dose.
