Изобретение относится к экспериментальной медицине, а именно к области воспроизведения в эксперименте болезней человека в пожилом возрасте. Изобретение может быть использовано для моделирования хронической сердечной недостаточности (ХСН) в стареющем организме с целью изучения возрастных особенностей ремоделирования миокарда и динамики изменения специфических маркеров на различных сроках формирования ХСН. Постарение населения большинства развитых стран обострило необходимость изучения механизмов развития ХСН в старом и суперстаром возрасте и апробация фармакологических веществ, разработанных для рациональной терапии и профилактики, требует разработки адекватной экспериментальной модели сердечной недостаточности у крыс возрастом 24 месяца.The invention relates to experimental medicine, namely to the field of experimental reproduction of human diseases in old age. The invention can be used to model chronic heart failure (CHF) in an aging body in order to study age-related features of myocardial remodeling and the dynamics of changes in specific markers at different stages of CHF formation. The aging of the population of most developed countries has exacerbated the need to study the mechanisms of the development of heart failure in old and super-old age, and the testing of pharmacological substances developed for rational therapy and prevention requires the development of an adequate experimental model of heart failure in rats aged 24 months.
Известны способы моделирования ХСН у молодых животных возрастом от 4 до 6 месяцев [Teerlink J.R. 1994; Цветкова А.С., 2019; Капелько 2014; Zhang XJ, 2021]. Данные способы являются наиболее близким к заявляемому по технической сущности и достигаемому результату. Недостатком этих способов при моделировании ХСН у крыс возрастом 24 месяца является гибель 90% экспериментальных животных уже в первые часы.There are known methods for modeling CHF in young animals aged 4 to 6 months [Teerlink J.R. 1994; Tsvetkova A.S., 2019; Kapelko 2014; Zhang XJ, 2021]. Цель достигается следующим образом. Животным под кожу в области холки вводят изопротеренол гидрохлорид в дозе 10мг/кг 4 раза в течение 11 суток по следующей схеме: 1-я инъекция на 1-й день эксперимента, 2-я инъекция на 4-й день эксперимента, 3-я инъекция на 8-й день эксперимента, 4-я инъекция на 11-й день эксперимента. Определение морфологических изменений в тканях миокарда, мозга, легких и почек проводят через 60 суток после последней инъекции. Новым в предлагаемом изобретении является то, что для постановки модели изопротеринол гидрохрид вводят в дозе 10 мг/кг 4 раза с интервалом между инъекциями 2 суток, что меньше в 17 раз используемой для молодых животных дозы препарата и позволяет значительно увеличить процент выживания животных в первые сутки после его введения. The epithelium of the proximal renal tubules is characterized by granular degeneration (9). In the group of control rats, such features of the morphological picture of the kidneys were absent.
На фиг. 4 представлена гистологическая картина легких крыс контрольной группы (а) и группы с ХСН (б). В препаратах лёгких крыс с ХСН отмечается неравномерное полнокровие внутриорганных капилляров (10), в части полей зрения определяются дистелектазы (11), явления фиброза межальвеолярных перегородок (12) и перивазальный фиброз (13). В группе контрольных крыс, такие особенности морфологической картины лёгких отсутствовали.In fig. В эксперименте на крысах-самцах возрастом 24 месяца показано, что при подкожном введении изопротеренол гидрохлорида в дозе 10 мг/кг по предложенной схеме на 60 сутки после последней инъекции в тканях миокарда, мозга, легких и почек сформировались изменения этих органов типичные для ХСН. Предлагаемый способ обеспечивает развитие приближенной к клиническим условиям картины изменений в жизненно важных органах и может быть использован для моделирования ХСН у крыс возрастом 24 месяца.An experiment on male rats aged 24 months showed that with subcutaneous administration of isoproterenol hydrochloride at a dose of 10 mg/kg according to the proposed regimen, 60 days after the last injection, changes in these organs typical of CHF were formed in the tissues of the myocardium, brain, lungs and kidneys. The proposed method ensures the development of a picture of changes in vital organs that is close to clinical conditions and can be used to model CHF in rats aged 24 months.
