These features make it possible to consider bacteria of the genus Bacillus as effective producer strains, which requires effective methods for manipulating the bacterial genome.

При титре культуры клеток 109 после индукции клетки титруют до разведения 10-3 и высевают на агаризованную среду LB с соответствующим индуктором (ксилозой или ИПТГ) и 20 мкг/мл тетрациклина и инкубируют на 30°С в течении 1-2 суток. После инкубации на чашках с разведением 10-3 вырастает порядка 20-50 клонов, которые пересевают на агаризованную LB среду с соответствующим индуктором и 20 мкг/мл тетрациклина. Из клонов выделяют хромасомальную ДНК методом [33], ставят ПЦР с праймерами Р27/Р28 и отправляют на секвенирование.
