Procedures: A control group was selected to receive a blank solution (5% CMC-Na solution once daily by gavage) and a group receiving crystalline Form I (180 mg/kg once daily by gavage). After 2 days of acclimatization, the prepared animals were randomly divided into 4 groups of 10 animals each. On day O, H22 cells were inoculated subcutaneously at 1×10 4 /mouse into the right axilla. Treatment began 24 hours after vaccination. The long and short diameters of the tumor were recorded daily and the tumor size was calculated using the formula: long diameter × short diameter 2 × 0.5236. Body weight was measured every 2 days. 21 days after the start of treatment, blood was taken from the eyeballs after anesthesia with chloral hydrate. Mice were sacrificed and tumors were rapidly collected. At the end of the experiment, the tumors were weighed for summarization with the corresponding statistics shown in the following table:
22 (n=10)Inhibition of tumor growth by crystalline form I in mice using H22 cells (n=10)
From the above results, it can be seen that the tumor growth inhibition of the crystalline Form I obtained according to the present invention is up to 88.7%. The particular crystalline form is believed to further improve the therapeutic activity of the compound. Presumably, the reason may be that the crystalline form I has significantly reduced the amount of impurities that are unfavorable to activity or are harmful, and the crystalline form I also has good compatibility with solvents upon administration, resulting in easier absorption of the drug.

The procedures for determining HBV DNA, HBV RNA, HBsAg and HBeAg in cell culture supernatant were as follows:
1) The required number of cells was seeded into a 12-well plate. After 24 h, the medium was replaced with fresh medium containing no double antibodies or FBS. HBV plasmids were transfected with Lipo2000. After 6 hours, the medium was replaced with medium containing double antibodies and 10% FBS to establish a continuous culture.
After 48 h, 1 ml of cell culture supernatant was collected to detect HBsAg and HBeAg. The indicated cells were washed at least 5 times with PBS. 1 ml of the wash solution from the last wash with PBS was collected and used as a baseline to determine the extent of plasmid washout. Note: When washing the cells, PBS was slowly added to the dish along the wall of the dish without resuspending the cells. The dish was then gently shaken to wash the cells, and the remaining PBS was removed after each wash to minimize the amount of remaining plasmids.
The cells were transferred to a large culture dish and cultured in culture medium for 3 days. To detect HBV DNA and HBV RNA, 1 ml of cell culture supernatant was collected. If detection was not made immediately, the samples were stored at -20°C.
1 ml of the collected cell culture supernatant and the last PBS wash solution, which were collected after 48 hours, were centrifuged at 5000 rpm for 5 minutes. 200 μl of the supernatant after centrifugation was added to 5 μl of DNase I (deoxyribonuclease I) + 5 μl of DNase I buffer, mixed well and subjected to brief centrifugation. The supernatant was incubated for 1 hour at 37°C, mixed every 30 minutes and subjected to short-term centrifugation.
After 1 hour, 5 μl of EDTA was added and the system was incubated at 65°C for 10 minutes before the reaction with deoxyribonuclease I was stopped.

(SRB).Mouse liver fat storage cells (mHSC) were cultured using 20% FBS in an incubator at 37°C, 5% CO 2 . In the cell proliferation experiment, the cell density was 5000 cells per well, treatment was performed after induction with TGF-β1 at a concentration of 5 ng/ml for 24 hours, and inhibition of cell proliferation was detected using sulforhodamine B (SRB).
2. Immunoblotting
(SDS-PAGE).mHSC cells were transferred to dishes at a rate of 10 6 cells per dish and processed 12 hours after adhesion. After 48 h, the cells in the supernatant were collected and combined with adherent cells. Cells were lysed on ice for 30 minutes using RIPA lysate containing protease inhibitor and centrifuged at 16,000 rpm for 15 minutes, retaining the supernatant. The concentration of the cell lysate was determined using BSA (BCA) and a certain amount of loading buffer was added to prepare a protein sample at a concentration of 2.5 μg/μl. The content of a-SMA, collagen I and collagen III was determined using SDS-PAGE.
3. Animal study
20 male Balb/c mice, 6 weeks old, were randomized into 4 groups. After a stepwise gradient induction using 25% CCU for 4 weeks (three times a week), the BSL group (90 mg/kg) and the BSM group (180 mg/kg) were treated once daily, as well as the control and model groups. a 5% solution of CMC-Na was introduced. After 8 weeks of induction, mice were sacrificed, blood collected and centrifuged at 3000 rpm for 15 minutes to obtain serum for TGF-β1, TrxR1, AST, ALT, ALP, TP and expression (animal liver fibrosis model is shown in Fig. 6). A portion of the liver was fixed with 4% paraformaldehyde and subjected to immunohistochemical staining with hematoxylin and eosin (HE) and Masson staining. The expression of α-SMA and collagen I in liver tissue was determined by immunohistochemistry. Part of the liver was preserved in liquid nitrogen.
In FIG. 7 shows the measured values of IC 50 , and from the result in FIG. 7 shows that since TGF β1 plays a very important pathological role in fibrotic lesions, and HSC cells are composed of 5% normal liver cells, liver injury usually leads to the transformation and activation of HSC cells, with TGF β1 being a strong factor in fibrosis. The inhibitory effect of the crystalline form of the present invention on HSC cells is one of the important factors for the anti-fibrotic effect.
In FIG. 8 shows a comparison of various biochemical parameters and important protein markers in serum after administration of BS to fibrosis model mice (the corresponding results are shown in the following table). As can be seen from FIG. 8 and the results shown in the following table, the average body weight of mice in the model group and treatment groups was significantly reduced, especially in the model group, compared with that in the control group. At the end of the study, the average body weight in each of the four groups was 24.76 ± 4.84 g, 14.08 ± 1.51 g, 17.00 ± 0.68 g, and 18.04 ± 2.23 g, respectively.
Table 1. Body weight and biochemical parameters of blood serum of mice for all groups at the end of the study

In FIG. Figure 9 shows the results of immunohistochemical staining of the liver and tissues after experiments on the control and model groups, as well as the group receiving the BS drug in a low dose (BSL), and the group receiving the BS drug in a medium dose (BSM). As can be seen from the results of group (A), mice in the control group showed a smooth, glossy, dark red, soft liver surface, indicating a normal liver condition. At the end of the study, mice in the model group showed decreased activity, weight loss of up to 43.2%, matte, light, hard and granular liver surface and fibrotic condition, indicating the successful creation of the model. These symptoms in the liver of mice in the treatment groups improved, and the granularity on the surface of the liver of mice decreased.
The results of hematoxylin and eosin (HE) staining show that liver cells showed severe swelling and severe inflammation in the model group compared with the control group, and most of the border plates of liver cells were destroyed (>50%); in contrast, in the BSL and BSM groups, the symptoms of hepatocyte necrosis and inflammation were significantly reduced compared with the model group.
Masson staining results show that mice in the model group showed bridging fibrosis between the portal areas, and fibrotic gaps were also formed; in the BSL and BSM groups, symptoms gradually decreased compared to the model group; the control group demonstrated only a few fibers around the portal area.
In FIG. 10 shows the expression of (A) α-SMA and (B) collagen 1A1 in the liver for different groups of mice at the end of the experiment in a model of liver fibrosis at various concentrations of crystalline form I. FIG. 10, based on the results of α-SMA staining and collagen 1 staining, it can be seen that the expression level of the corresponding proteins in the liver of mice in the model group was significantly higher compared with the control group, while these symptoms in the BSL and BSM groups were significantly reduced, which is clearly significantly distinguishes them from the model and control groups.
