The cleaved pAd5/3-D24 virus backbone was assembled with the CD40L-GA fragment using the Gibson assembly master mix (NEB, E2611) according to the manufacturer's instructions for generating pAd5/3-D24-CD40L. The Gibson assembly reaction mixture was transformed into NEB® 5 alpha competent E. coli cells (NEB, C2987H) according to the manufacturer's instructions. Positive colonies were screened by PCR and correct recombination was further confirmed by construct sequencing. To generate the final construct, i.e. a virus containing both the CD40L and OX40L genes, the fragment including OX40L (OX40L-GA) was amplified using the following primers: Gibson OX40L FW and Gibson OX40L REV (amplification of the region in pShuttle-OX40L corresponding to nucleotides with 11 to 3287), see the primer sequences in Table 4b. Most importantly, when analyzing the functionality of OX40L expressed from viruses, a clear downstream gene activation was seen using HEK-293 cells stably expressing the OX40 receptor (FIG. 4). Binding of OX40L to OX40 triggers an intracellular signaling pathway in these cells that, through activation of NF-κB, leads to the expression of the firefly luciferase reporter gene. Bioluminescence levels obtained using A549 cells infected with OX40L-expressing viruses clearly show that the OX40L protein is functional and activates downstream signaling when bound to OX40 compared to bioluminescence levels obtained using virus without a transgene or negative cell control.
