BACKGROUND OF THE INVENTION
(ADC).Duocarmycins are members of a family of antitumor antibiotics that include duocarmycin A, duocarmycin SA, and CC-1065. They are known for their powerful anticancer properties but are not commonly used on their own due to their extremely high toxicity. Duocarmycins are currently being investigated as cytotoxic drugs in antibody-drug conjugates (ADCs).
ADCs have the potential to address the large unmet need for effective new cancer treatments by targeting a potent cytotoxic drug specifically to cancer cells, thereby increasing efficacy while reducing potential systemic toxic side effects of the small molecule drug.
One of the key aspects of the future commercial success of ADCs is the method for synthesizing a cytotoxic drug and the corresponding linker-drug design, in which a linker fragment is attached to a cytotoxic drug to facilitate conjugation to an antibody that is suitable for production on an industrial scale.

е. SYD985 или (vic-)трастузумаб дуокармазин, был успешно использован в нескольких доклинических исследованиях (M.M.C. van der Lee et al., Molecular Cancer Therapeutics, 2015, 14(3), 692-703; J. Black et al., Molecular Cancer Therapeutics, 2016, 15 (8), 1900-1909) и стадии I клинических испытаний (ClinicalTrials.gov NCT02277717). (TULIP; ClinicalTrials.gov NCT03262935).first disclosed in WO2011/133039 as compound 18b on page 210, ll. 21-27 is an example of a potent analogue of CC-1065. ADC vc- seco -DUBA with anti-HER2 antibody trastuzumab, i.e. SYD985 or (vic-)trastuzumab duocarmazine has been successfully used in several preclinical studies (MMC van der Lee et al. , Molecular Cancer Therapeutics, 2015, 14(3), 692-703; J. Black et al. , Molecular Cancer Therapeutics , 2016, 15(8), 1900-1909) and Phase I clinical trials (ClinicalTrials.gov NCT02277717). SYD985 treatment is currently being directly compared to physician-choice treatment in Phase III clinical trials in patients with HER2-positive locally advanced or metastatic breast cancer (TULIP; ClinicalTrials.gov NCT03262935).
Synthesis of the linker drug vc- seco -DUBA is described in WO 2011/133039 as a four-step process. Preparation of vc- seco -DUBA from this method on a laboratory scale of 50-100 mg gave the linker drug in an overall yield of only 21-25%. The last two stages, i.e. steps 3 and 4 of this process are critical to the overall yield of vc- seco -DUBA from the entire process, showing an overall yield of only about 50%. On an industrial scale, the yield of this four step process will be even lower.
Therefore, there is a need for an improved process for preparing vc- seco -DUBA. In particular, there is a need for a process that is efficient in terms of yield and chemical purity, cost effective in terms of reagents and reaction conditions, and that is suitable for industrial scale production.

DETAILED DESCRIPTION OF THE PRESENT INVENTION
Duocarmycins are a class of structurally related toxins first isolated from the broth culture of Streptomyces species. They are members of a family of anticancer antibiotics that include duocarmycin A, duocarmycin SA, and CC-1065. Duocarmycins bind to the minor groove of DNA and subsequently cause irreversible DNA alkylation. This disrupts the structure of nucleic acids, which ultimately leads to the death of tumor cells.
CC-1065WO2011/133039 specifically discloses a potent linker drug vc- seco -DUBA of formula (I) (compound 18b on pages 210, ll. 21-27) which contains the duocarmycin derivative CC-1065
The present invention relates to an improved process for producing vc- seco -DUBA with surprisingly high yields, which can be successfully applied on an industrial scale.
The chemical synthesis of vc- seco -DUBA linker-drug in Example 10 of WO 2011/133039 is described as a four step process
, ,
в котором PNP-Cl представляет собой 4-нитрофенилхлорформиат, Et3N представляет собой триэтиламин, Boc представляет собой трет-бутилоксикарбонил, TFA обозначает трифторуксусную кислоту, CHCl3 обозначает хлороформ и DMF обозначает N,N-диметилформамид.in which PNP-Cl is 4-nitrophenyl chloroformate, Et 3 N is triethylamine, Boc is t-butyloxycarbonyl, TFA is trifluoroacetic acid, CHCl 3 is chloroform, and DMF is N,N-dimethylformamide.
On a laboratory scale of 50-100 mg, this four-step process shows an overall yield of only 21-25%. On an industrial scale, this yield will be much lower.
Неожиданно было обнаружено, что эта модифицированная процедура стадии 3 и использование нового промежуточного соединения привели к значительному увеличению выхода vc-seco-DUBA.The low overall yield of this four-step process may be largely due to the low combined yield of only about 50% laboratory scale of the last two steps, i.e. steps 3 and 4. stage 3, gives a new intermediate compound, which can be isolated by crystallization. Surprisingly, this modified step 3 procedure and the use of a new intermediate resulted in a significant increase in the yield of vc- seco -DUBA.
(II)Usually the crystallization step is introduced into chemical synthesis when it is necessary to increase the purity of the product. However, the introduction of such a stage usually reduces the yield of the specified product, since a significant amount of the product remains in the mother liquor. Unexpectedly, the inventors of the present invention found that the introduction of a crystallization step into the synthesis of vc- seco -DUBA as described above in the present description, resulting in a new intermediate compound of formula ( II )
не только приводит к увеличению чистоты (с 94-96% до ≥99,0%), но также показало неожиданное, значительное увеличение выхода vc-seco-DUBA (с 53% до ~79%).not only resulted in an increase in purity (from 94-96% to ≥99.0%), but also showed an unexpected, significant increase in the yield of vc- seco -DUBA (from 53% to ~79%).
(II).Therefore, in one embodiment, the present invention relates to a compound of formula ( II ).
(III)In a second embodiment, the present invention relates to a process for preparing a compound of formula (II) comprising reacting a compound of formula ( III )

IgG1-κ.In the context of the present invention, any antibody, in particular any antibody having therapeutic activity, or any antibody known in the art of ADC, or any antigen-binding fragment thereof, such as a F(ab') 2 or Fab' fragment, a single chain (sc) antibody, scFv, single domain (sd) antibody, diabody or mini-antibody can be used for (wild-type or site-specific) vc- seco -DUBA conjugation. The antibodies can be of any isotype, such as IgG, IgA or IgM antibodies. Preferably the antibody is an IgG antibody, more preferably an IgG 1 or IgG 2 antibody. Antibodies can be chimeric, humanized or human. Preferably the antibodies are humanized. Even more preferably, the antibody is a humanized or human IgG antibody, most preferably a humanized or human IgG 1 monoclonal antibody (mAb). Preferably, said antibody has κ (kappa) light chains, i. e. humanized or human IgG 1 -κ antibody.
In humanized antibodies, the antigen binding complementarity determining regions (CDRs) in the heavy chain (HC) and light chain (LC) variable regions are derived from antibodies of non-human origin, typically mouse, rat or rabbit. These CDRs of non-human origin can be placed in the human framework (framework region (FR) FR1, FR2, FR3 and FR4) of the HC and LC variable regions. Selected amino acids in the human FR can be substituted for the corresponding original non-human amino acids, for example, to improve binding affinity while maintaining low immunogenicity. Alternatively, non-human scaffolds are retained and selected non-human FR amino acids can be substituted for their corresponding human amino acids to reduce immunogenicity while maintaining antibody binding affinity. Thus, humanized variable regions are combined with human constant regions.
These antibodies can be produced by recombinant, synthetic, or other suitable methods known in the art.

WO2017/137628.Conjugation of the vc- seco -DUBA linker drug to the antibody can be carried out as described, for example, in WO2011/133039, WO2015/177360 and WO2017/137628.
Wild-type ADCs are produced by conjugating a linker drug to an antibody through free cysteine side chain thiols obtained by reduction of interchain disulfide bonds. The preparation involves partial reduction of solvent-exposed interchain disulfides, followed by modification of the resulting thiols with maleimide-containing linker drugs. The cysteine binding strategy results in a maximum of two drugs per reduced disulfide. Most human IgG molecules have four solvent-exposed disulfide bonds and therefore a range of zero to eight drugs per antibody is possible. The exact amount of drugs per antibody is determined by the degree of disulfide reduction and the number of linker-drug molar equivalents used in the subsequent conjugation reaction. Complete reduction of all four disulfide bonds results in a homogeneous eight-drug/antibody construct, while partial reduction typically results in a heterogeneous mixture with no, two, four, six, or eight drugs per antibody.
Site-specific ADCs are generated by conjugating a linker drug to an antibody through the side chains of engineered cysteine residues at appropriate positions in the mutated antibody. The engineered cysteines are usually completed with other thiols such as cysteine or glutathione to form disulfides. These closed residues must be released before drug attachment can occur. Attachment of the drug to engineered residues is either achieved by reducing both native interchain and mutant disulfides, then reoxidizing the native interchain cysteines using a mild oxidizing agent such as CuSO4 or dehydroascorbic acid, followed by standard conjugation of the freed engineered cysteine to the linker drug, or using mild reducing agents that reduce mutant disulfides at a faster rate than interchain disulfide bonds, followed by standard conjugation of the free engineered cysteine to the linker drug. Under optimal conditions, two drugs per antibody (ie, drug-antibody ratio, DAR, equal to 2) will be attached (if one cysteine is inserted into the heavy chain or light chain of the mAb).

After 30 minutes of stirring, the precipitated crude product was filtered on a pressure filter. The filter cake was thoroughly washed with water and dried on the filter under vacuum and a gentle flow of nitrogen.
Концентрированные растворы объединяли, фильтровали через фильтр 0,2 мкм и добавляли в воду для осаждения чистого vc-seco-DUBA в виде мелкодисперсного желтого порошка (выход: 35-45%; чистота: ≥99,0%).The crude vc- seco -DUBA product was first subjected to low pressure flash chromatography (stationary phase silica gel 0.040 to 0.063 mm; mobile phase dichloromethane:methanol=90:10). The appropriate fractions (with a purity of UPLC-IN vc- seco -DUBA ≥90%) were collected in a flask, filtered and evaporated. Further purification was carried out using preparative chromatography (stationary phase - silica gel from 0.015 to 0.040 mm; mobile phase - dichloromethane:methanol=90:10-85:15). The appropriate fractions (with purity UPLC-IN vc- seco -DUBA ≥90%) were collected in a flask and the solvent was changed to DMA. The concentration was carried out at a maximum temperature of 25°C. The concentrated solutions were combined, filtered through a 0.2 μm filter and added to water to precipitate pure vc- seco -DUBA as a fine yellow powder (yield: 35-45%; purity: ≥99.0%).
