Abstract
Description
The field of technology to which the invention belongs
The present invention relates to a stable liquid preparation of antibodies, and the antibody is included in a double buffer system, including "phosphate-amino acid buffer".
State of the art
Advances in biotechnology have paved the way for the development of a large number of antibodies for therapeutic use. However, for any antibody-based therapeutic agent, the stability of an antibody preparation is one of the most important criteria for ensuring its safety as well as its effective administration.
Liquid protein/antibody preparations are generally preferred due to their ease of administration and compliance with many commercially available devices when administered. However, antibodies, being larger and more complex than traditional "small molecular weight" drugs, are also more unstable in solution. Antibodies are more sensitive to pH, temperature, and oxidation and can undergo various covalent and non-covalent modifications and/or degradations in solution. In particular, antibody degradation is faster in liquid formulations, leading to physical and chemical instability of the molecule. Examples of chemical instability include deamidation, hydrolysis, oxidation, or disulfide change, while physical instability may result from denaturation, aggregation, adsorption, or precipitation.
In addition, liquid formulations with high antibody/protein concentrations show high viscosity and increased aggregation in solution, which affects the stability and potency of the molecule. Thus, antibodies in liquid/solution raise their inherent questions for incorporation into formulations for therapeutic applications.
The aim of the present invention is to address such questions in the development of resistant antibody preparations.
The essence of the invention
The present invention relates to a stable liquid preparation of antibodies, and the preparation includes a double buffer system, including as a buffer phosphate and amino acid.
Components. The amino acid component in the double buffer system is the counterion to the phosphate component. This "phosphate-amino acid" double buffer system allows the stabilization of the antibody in the concentration range from about 10 mg/ml to about 200 mg/ml. In particular, the invention relates to a stable liquid formulation in a phosphate amino acid buffer comprising sorbitol, a surfactant and optionally arginine.
Иными словами, состав препарата по изобретению защищает антитела (например, тоцилизумаб) от окисления даже без использования в композиции какого-либо (каких-либо) известного(ых) антиоксиданта(ов).Specifically, the above formulation of the invention is devoid of any known antioxidant(s). In other words, the composition of the preparation according to the invention protects antibodies (for example, tocilizumab) from oxidation even without the use of any known antioxidant(s) in the composition.
In addition, the present invention relates to a stable liquid preparation of antibodies with low viscosity, and the preparation includes a "phosphate-amino acid" buffer system, and wherein the viscosity of the preparation is less than 10 cps and preferably less than 5 cps.
In addition, the antibody in the preparation, included in the "phosphate-amino acid" buffer system, is stable under storage conditions such as 2-8°C for at least 6 months or at 25°C for at least 6 months or at 40°C for at least 2 weeks or at 40°C for at least 4 weeks.
The content of aggregates in the composition of the antibodies obtained in the "phosphate-amino acid" buffer system is less than 5%, and the content of monomers is at least 95% under the above storage conditions.

Detailed description of the invention
ОпределенияDefinitions
The term "antibody" refers to a glycoprotein comprising at least two heavy (H) chains and two light (L) chains linked by disulfide bonds, or an antigen-binding portion thereof. The term "antibody" as used herein encompasses whole antibodies or any antigen-binding fragment (i.e., "antigen-binding portion") or fusion protein thereof.
The term "resistant" drug refers to a drug in which the antibody retains its physical stability and/or chemical stability and/or biological activity after storage.
The term "antioxidant" as used herein includes an agent that inhibits oxidation and thus is used to prevent degradation of drugs by the oxidative process. Such compounds include, for example and without limitation, methionine, cysteine, carnosine, ascorbic acid, ascorbyl palmitate, citric acid, butylated hydroxyanisole, butylated hydroxytoluene, hydrophosphoric acid, monothioglycerol, propyl gallate, sodium ascorbate, sodium citrate, sodium sulfide, sodium sulfite, sodium bisulfite , sodium formaldehyde sulfoxylate, thioglycolic acid, sodium metabisulphite, EDTA (edetate), pentetate. The disclosed preparation according to the invention is devoid of an antioxidant, in particular methionine.
Resistance studies provide data on the quality of an antibody under the influence of various environmental factors over time. The ICH "Q1A: Stability Testing of New Drug Substances and Products" indicates that data from accelerated stability studies can be used to evaluate the impact of short-term deviations above or below the labeled storage conditions that may occur during the transport of antibodies.
Various analytical methods are available to measure the physical and chemical degradation of an antibody in pharmaceuticals. An antibody "retains its physical stability" in a pharmaceutical formulation if it does not show signs of aggregation, precipitation and/or denaturation after visual inspection of color and/or clarity, or when measured by UV light scattering or size exclusion chromatography. An antibody is said to "retain its chemical stability" in a pharmaceutical formulation when it exhibits no or minimal formation of product variants, which may include variants resulting from chemical modification of the antibody of interest, such as deamination, oxidation, etc. Analytical methods such as ion exchange chromatography and hydrophobic ion chromatography can be used to study the chemical variants of a product.
The term "monomer" as used herein describes antibodies composed of two light chains and two heavy chains. The monomer content of an antibody composition is typically analyzed by size exclusion chromatography (SEC). In the SEC separation principle, large or high molecular weight (HMW) molecules are eluted first, followed by smaller or lower molecular weight molecules. In a typical SEC profile for an antibody composition, aggregates, which may include dimers, polymers, etc., are eluted first, followed by the monomer, and truncated antibody variants or degradants may be eluted last. Under some conditions, the aggregate peak or the degradant peaks may not elute as basic single peaks but as overhangs or abnormally broad peaks. In order to maintain the appropriate activity of an antibody, in particular a therapeutic antibody, it is desirable to reduce the formation of aggregates or degradation products and therefore control the monomer content at the intended level. The ability to inhibit the formation of aggregates and the content of degraders, measured at different time points during stability studies, may indicate the suitability of the drug candidate for the antibody of interest. A TSK-GEL G3000SWXL (7.8 mm x 30 cm) column from TOSCH on aqueous HPLC can be used to perform SEC.
[Chromatographic analysis of the acidic and basic species of recombinant monoclonal antibodies; mAbs; Page number 581; Volume 4; Issue 5]. The term "major peak" as used herein refers to a peak that elutes in abundance (major peak) during cation exchange chromatography. The peak that elutes during cation exchange chromatography earlier than the main peak, with a charge that is acidic relative to the main peak, is called an acid variant peak. A peak that elutes later than the main peak during cation exchange chromatography, with a charge that is alkaline relative to the main peak, is called an alkaline variant peak. The content of the main peak can be determined by ion exchange chromatography (IEC). There are two types of IEC available, namely cation and anion exchange chromatography. Positively charged molecules bind to anion exchange resins, while negatively charged molecules bind to cation exchange resins. In a typical cation exchange chromatographic profile, the acidic variants of the antibody composition will elute first, then the main peak, and then the alkaline variants will elute last. Acid variants are the result of antibody modifications such as deamidation of aspartic residues. Alkaline variants are the result of incomplete removal of the C-terminal lysine(s) residue(s), incomplete cyclization of the N-terminal glutamine, isomerization of asparagine, and also, especially, oxidation of methionine residues present in the Fc region of the antibody [Chromatographic analysis of the acidic and basic species of recombinant monoclonal antibodies; mAbs; Page number 581; Volume 4; Issue 5]. Typically, in an antibody, a lysine residue is present at the C-terminus of both the heavy and light chains. An antibody molecule containing a lysine in both the heavy and light chains is a K2 variant, an antibody molecule containing a lysine residue in any one heavy or light chain is a K1 variant, and an antibody molecule lacking one is a K0 molecule. The enzyme carboxypeptidase B (enzyme CP-B) acts on the C-terminal lysine residues present in the K2 and K1 variants and thus converts them into K0 molecules. According to the circumstances of the case, IEC analysis can be performed on samples digested with the enzyme carboxypeptidase B (CP-B). In a typical stability study, it is expected that a stable preparation during the study leads to a reduction in the formation of charged variants (acidic and alkaline variants) and therefore minimizes any decrease in the content of the main peak.
Dynamic Light Scattering (DLS) measures fluctuations in scattering intensity from particles undergoing random Brownian motion as a function of time. Information about the diffusion coefficient and particle size can be obtained from the analysis of such fluctuations. More specifically, the method makes it possible to measure the size characteristics of proteins in a liquid medium. The values of the diffusion coefficient obtained by this method are directly proportional to the stability of the molecule in a given solution.
Differential scanning fluorometry (DSF) provides the ability to quickly determine the stability of proteins with high throughput and allows direct comparison of different proteins or the same protein under different research conditions. DSF controls the thermal unfolding of proteins in the presence of a fluorescent dye and is typically performed using a real-time PCR tool. Fluorescent dyes that can be used for DSF are highly fluorescent in a non-polar environment, such as hydrophobic sites on unfolded proteins, compared to an aqueous solution where the fluorescence is quenched. Fluorescence intensity is plotted as a function of temperature; this gives a sigmoid curve that can be described by a two-step transition.

Detailed Description of Embodiments
The present invention relates to a liquid antibody preparation comprising a double buffer system comprising phosphate and amino acids as buffer components. The "Phosphate-Amino Acid Buffer System" is suitable for incorporating the antibody into the formulation at low as well as high concentrations. In addition, the buffer system allows the drug to be less viscous even at high antibody concentrations. In addition, the disclosed buffer is advantageous in terms of "more active recovery" of the antibody in relation to the "production method". This method, after purification of the antibody, first includes a "buffer exchange" step, in which the buffer from the next method step is replaced with a buffer that absorbs the antibody for incorporation into the formulation. But this “replacement” step is often accompanied by a significant loss of the antibody. However, when the "following buffer" is replaced with the "phosphate-amino acid" buffer of the present invention, this antibody loss is significantly reduced.

Tocilizumab suitable for storage in the pharmaceutical composition of the present invention is prepared by standard methods known in the art. For example, tocilizumab is produced by recombinant expression of immunoglobulin light and heavy chain genes in a mammalian host cell, such as Chinese hamster ovary cells. The expressed tocilizumab is then collected and the crude product is subjected to standard subsequent process steps which include purification, filtration and optional dilution or concentration steps. For example, the crude tocilizumab product can be purified using standard chromatographic techniques such as affinity chromatography, ion exchange chromatography, and combinations thereof. The purified tocilizumab solution can be further subjected to one or more filtration steps, and the resulting solution is subjected to subsequent formulation studies.
Example 1 Single Buffer vs Phosphate Amino Acid Buffer
I, (a) и (b)]. Также проверяют содержание главного пика с использованием ионообменной хроматографии [результаты приведены на фигуре II].Tocilizumab (at a concentration of 35 mg/ml), obtained at the final stage of the subsequent process, is replaced with a buffer and concentrated to 230 mg/ml in single buffers, as well as in a double buffer, such as a phosphate-amino acid buffer. The composition of the buffer formulations is shown in Table 1. After concentrating tocilizumab in various buffers to 230 mg/mL, the samples are tested for high molecular weight (HMW) particles and monomer content using size exclusion chromatography [results are shown in FIG. I, (a) and (b)]. Also check the content of the main peak using ion-exchange chromatography [the results are shown in figure II].

Example 2 Preparations with high concentration of tocilizumab (180 mg/ml) in a double buffered phosphate-amino acid system
In order to obtain a stable liquid high concentration formulation of tocilizumab, as part of the experiment, various combinations of double buffers on a "phosphate-amino acid basis" were prepared at concentrations of 20 mM. Tocilizumab (at a concentration of 30.02 mg/ml), obtained at the final stage of the subsequent process, replace the buffer with various phosphate-amino acid buffers (listed in table 2) and concentrate to 180 mg/ml in the indicated double buffers. The buffer composition for tocilizumab changed to histidine buffer is retained as a control, since the approved and marketed formulations with 180 mg/ml tocilizumab Actemra® or RoActemra® are histidine buffered formulations. In addition, one of the double buffers is maintained as a "non-phosphate amino acid" buffer, which serves as a negative control.

Example 7 Stable formulations of tocilizumab (180 mg/ml) lacking antioxidants
Образцы периодически извлекают в различные моменты времени и анализируют на содержание кислых, щелочных частиц и главного пика с использованием хроматографии IEX [результаты приведены в таблице 11 а-с].In general, oxidation, specifically oxidation of methionine residues, generates alkaline variants eluting during IEX chromatography as an alkaline peak (ie, later than the main peak). From the experiment described above, it can be seen that tocilizumab preparations containing sorbitol and arginine are the most stable compared to preparations that do not contain arginine. In addition, different concentrations of sorbitol and arginine contained in tocilizumab preparations without any antioxidant (methionine) were evaluated for the formation of alkaline particles. The composition of the preparations is shown in table 10. All samples are subjected to stability studies at 2-8°C, 25°C and 40°C. Samples are removed periodically at various time points and analyzed for acidic, alkaline and main peak content using IEX chromatography [results are shown in Table 11 a-c].

Example 8 Tocilizumab (20 mg/mL) formulations in phosphate-amino acid double buffered system comprising various excipients
As part of the experimental solution, to prepare preparations with low concentration of tocilizumab (20 mg/ml), various "phosphate-amino acid" buffers are prepared at a concentration of 20 mM, to which various excipients are added, such as sugars/polyols and surfactants. Optionally, a salt, namely sodium chloride, is added to some "phosphate-amino acid" buffer combinations. The buffer for tocilizumab (at a concentration of 30.02 mg/ml) obtained at the final lower stage of the method is replaced with various double buffer systems, including phosphate-amino acid buffers and pharmaceutically acceptable excipients [the composition is given in table 12]. Samples are then diluted/adjusted in an appropriate buffer medium containing excipients to achieve a tocilizumab concentration of 20 mg/mL. Tocilizumab included in a buffer composition containing phosphate buffer, sucrose and polysorbate 80 is used as a reference, since the approved drug at a dosage of 20 mg/ml Actemra® or Roactemra® is presented in this buffer composition.
