Пробы крови после отбора и перемешивания 15 минут, как описано выше, были исследованы с помощью автоматизированного гематологического анализатора ABX (Horiba, ABX Pentra 60C +) с целью общего клинического анализа крови (ОАК) с определением 5 диф, включая концентрацию лейкоцитов, концентрацию тромбоцитов, средний объем тромбоцитов (MPV), концентрации и фракции нейтрофилов, лимфоцитов, моноцитов, эозинофилов и базофилов, концентрацию эритроцитов, гематокрит, концентрацию гемоглобина, описательные параметры эритроцитов (MCV - средний объем эритроцита, MCH - среднее содержание гемоглобина в эритроците, MCHC - средняя концентрация гемоглобина в эритроцитарной массе, RDW - ширина распределения эритроцитов). Были получены образцы от нескольких доноров. Использовали измеренную концентрацию тромбоцитов для сравнения с данными, полученными при ручном подсчете концентрации тромбоцитов в пробах с илопростом и гепарином (в устройствах для проб PICO70), и в пробах с антикоагулянтом ЭДТА. The measured platelet concentration was used for comparison with data obtained by manually counting the concentration of platelets in samples with iloprost and heparin (in PICO70 sample devices), and in samples with the anticoagulant EDTA. The ABX analyzer may underestimate the concentration of platelets when they are aggregating, although due to the shear force in the flow, some of the weakly connected platelets may be separated. For control, EDTA blood samples without platelet clots were used as a reference measurement for the ABX automated measured platelet concentration.

Пример 5. Анализ влияния антитромбоцитарного препарата на показатели анализатора ABL90Example 5. Analysis of the effect of an antiplatelet drug on the performance of the ABL90 analyzer
Каждый эксперимент проводился в отдельный день с кровью от одного добровольного донора. Для каждого эксперимента были подготовлены три устройства для проб в виде шприцев PICO70 (Radiometer Medical ApS) (не модифицированных, содержащих золотой шарик и гепариновый брикет). Один PICO70 использовался в качестве немодифицированного контроля (Контроль), один PICO70 был заполнен разведенным антитромбоцитарным препаратом в объеме 15 мкл, с достижением указанной конечной концентрации в пробе крови. Третий образец PICO70 был заполнен растворителем в объеме 15 мкл (определенный растворитель, который используется для разведения тестируемого антитромбоцитарного препарата: референс-проба). Отбор проб венозной крови производили в шприцы для проб PICO70 через иглу-бабочку и закупоривали вентилируемой крышкой (VTC), при этом в самозаполняющиеся пробоотборники PICO70 набирали цельную кровь в объеме 1,5 мл. Сразу после заполнения шприцев пробы крови восемь раз переворачивали, чтобы гарантировать надлежащую гепаринизацию крови. Все измеренные значения указаны в Таблице 3. После этого пробы крови центрифугировали для отделения фракции плазмы. Плазму всех проб трижды поочередно исследовали с помощью анализатора ABL90 для оценки концентрации свободного гемоглобина в плазме, которая указывает на возможный нежелательный гемолиз эритроцитов в образцах цельной крови. For each experiment, three PICO70 (Radiometer Medical ApS) syringe sample devices (unmodified, containing a gold bead and a heparin cake) were prepared. The third PICO70 sample was filled with 15 µl of diluent (the specific diluent that is used to dilute the antiplatelet drug under test: a reference sample). Venous blood samples were taken into PICO70 sample syringes through a butterfly needle and sealed with a vented cap (VTC), while 1.5 ml whole blood was drawn into PICO70 self-priming samplers. Immediately after filling the syringes, the blood samples were inverted eight times to ensure proper heparinization of the blood. The blood samples were stirred using a SAM (Radiometer Medical ApS) stirrer, while in the blood sample, a gold-plated steel ball was moved to ensure homogeneous and repeatable mixing for 15 minutes after blood collection. After that, blood gas parameters were determined in the ABL90 analyzer (Radiometer Medical ApS). Measurements were also carried out in alternative samples (control, reference sample, drug sample) five times in each sample (5 repeated measurements), carefully turning the blood samples manually to keep the blood in a state of homogeneous mixing during the measurements. All measured values are shown in Table 3. After that, the blood samples were centrifuged to separate the plasma fraction. The plasma of all samples was analyzed three times in turn using the ABL90 analyzer to assess the concentration of free hemoglobin in plasma, which indicates a possible unwanted hemolysis of erythrocytes in whole blood samples. In hemolyzed samples, the effect is shown, for example, on the measured K + concentration.

Эптифибатид, тирофибан и илопрост не влияли на оцениваемые в анализаторе ABL показатели, тогда как MgSО4 влияет на различные показатели (pH, N+, K+, Ca2+, Cl- и Lac) (различие доли максимального влияния >2). Eptifibatide, tirofiban, and iloprost had no effect on the ABL analyzer values, while MgSO4 affected various parameters (pH, N+, K+, Ca 2+ , Cl - and Lac) (difference in the proportion of maximum effect >2).
