Abstract
Description
The invention relates to biotechnology and concerns the production of plant protection products, in particular, a method of reproduction of a producer of microbiological preparations against dangerous pests of lepidoptera phytophagous insects.
Diatraea saccharalis [8].Microsporidia are known for their high pathogenicity for insects, and many species of these parasites reduce the indicators of viability, reproductive activity and resistance to negative environmental factors and persist for a long time in host populations due to horizontal (among individuals of one generation) and vertical (in a number of generations) transmission due to various methods of insect infection (Becnel, Andreadis, 1999 [1]). This makes it possible to achieve a long-term effect on the populations of pests of agriculture and forestry, such as the cabbage whitelet Pieris brassicae, the meadow moth Loxostege stisticalis, the gypsy moth Lymantria dispar, etc. [2-4]. In particular, the species of the genus Nosema are important in limiting the number of harmful lepidoptera, including the spruce budworm Choristoneura fumiferanae [5-7], as well as the sugarcane pest Diatraea saccharalis [8].
[10].Microsporidia Nosema pyrausta is an important factor in the dynamics of the abundance of the dangerous maize pest, the corn moth Ostrinia nubilalis, from the Crambidae (Pyraloidea) family of moth grass [9]. In addition, this pathogen exhibits high biological effectiveness in relation to another representative of the specified family, a particularly dangerous agricultural pest, the meadow moth Loxostege sticticalis, for the control of which a highly virulent strain of N. pyrausta was created, deposited in the State Collection of Microorganisms FGBNU VIZR under registration number M-1 VIZR. For the mass production of spores of this entomopathogen, the method of artificial infection of caterpillars of the typical host of the corn moth is used, followed by the cultivation of infected insects in laboratory conditions and the isolation of spores from pupae, which makes it possible to obtain about 50 million spores from one pupa [10].
[11].To feed insects at the caterpillar stage, a standard composition of an artificial nutrient medium (IPA) is used, which includes corn flour, wheat germ, brewer's yeast, a complex of vitamins, agar, and antibiotics per liter of water [11].
This method of breeding insects, as the closest in terms of a set of essential features, was used as a prototype. Its main disadvantage is the relatively low productivity of the microbiological plant protection agent.
Thus, the current technical problem is the low productivity of the microbiological plant protection agent when breeding infected insects. The present invention seeks to solve this problem by providing a method with an increased spore yield of N. pyrausta microsporidia per 1 pupa of the host insect.
The technical result is a significant increase in the productivity of microsporidia spores.
[12].The technical result is achieved by increasing the nutritional value of the fodder substrate by partially replacing one of the components, corn flour, with soy flour, which is characterized by an amino acid composition close to that of animal products [12].
1:1.Thus, the technical result is achieved by the fact that in the method for the production of spores of the microsporidia Nosema pyrausta for the control of polyphagous lepidoptera pests, including the cultivation of artificially infected caterpillars of the corn moth Ostrinia nubilalis at a temperature of + 24 ° C for 30 days on agar nutrient medium containing cereal flour , wheat bran, yeast extract and a mixture of vitamins and antibiotics, and the isolation of the producer's spores from pupated insects, it is proposed to use corn and soy flour as cereal flour in a 1: 1 ratio.
Дополнительным отличием способа является то, что состав компонентов агаризованной питательный среду следующий, в масс %:An additional difference of the method is that the composition of the components of the agar nutrient medium is as follows, in wt%:
35%.As a result of long-term studies, the authors found that insects fed on IPA with the addition of soy (IPA-soy) gain weight faster, resulting in the formation of larger pupae containing on average 70 million spores per pupa. This makes it possible to increase the productivity of microsporidia spores by an average of 35%.
Nosema pyraustaExample 1. Obtaining biomass of microsporidium Nosema pyrausta
Ostrinia furnacalis corn moth eggs from a permanent laboratory culture were placed on a layer of standard agarized IPA (Table 1), poured into 0.5 L glass vessels, which were placed upside down because hatching caterpillars move upward due to a negative geotactic reaction, which allows them to quickly find food.
Groups of 2nd instar caterpillars in the amount of 25 individuals 1-3 days after molting were placed on a piece of office paper folded in several layers in a hermetically sealed container for 2 hours without food. Spores of microsporidia N. pyrausta in an amount of 2.5 × 10 6 were mixed with a 0.5 g portion of IPA and glued to the bottom of a glass vessel with a 0.5 L jar. The previously planted caterpillars were transplanted into a jar with food containing spores, the jar was covered with a layer of dense cloth, fixed with an elastic band, and turned upside down. After complete consumption of the food with spores, the caterpillars were transplanted into cans with standard IPA or with IPA soybeans (Table 2) and kept at a temperature of + 24 ° C and a photoperiod of 18C: 6T for 20-30 days until pupation. Surviving pupae were collected 3-5 days after pupation and kept in a refrigerator for 5-10 days. Pupae were individually placed in a 1.5 ml plastic Eppendorf tube, 100 μL of distilled water was added, and homogenized with a plastic pestle for 30 sec. The tubes were centrifuged at 4000 g for 5 min. The supernatant was decanted, the precipitate was resuspended in 1 ml of distilled water, and the concentration of spores was calculated in the Goryaev chamber. This method was used to obtain from 23 to 80 million spores per pupa, on average 52.0 ± 3.51 million (the number of analyzed insects N = 20), while feeding the caterpillars on standard IPS; and from 19 to 128 million spores per pupa, on average 69.9 ± 6.79 million spores (N = 19). The revealed differences in the spore productivity of microsporidia during feeding of the host insect on PSI of various composition are statistically significant at the 5% significance level.
Crambidae. Thus, when using the proposed method of reproduction of N. pyrausta microsporidia, a high spore productivity of the parasite is achieved, significantly exceeding that when using an analogue, in particular, a method of growing infected caterpillars on a standard IPA (Table 2, line 1). The productivity of N. pyrausta, expressed as the number of spores formed per 1 pupa of the host insect, when insects are grown on IPA with the addition of soybeans is 35% higher than that of standard IPA. This method can be recommended for the production of microsporidia spores, promising for the protection of crops from the polyphagous lepidoptera pests of the Crambidae family.
