Abstract
Description
The invention relates to the field of microbiology, more specifically to a method for producing biomass of methane-oxidizing microorganisms and a line for its production. The invention can find application in the industrial production of bioprotein in the form of biomass of aerobic microorganisms for biotechnological purposes when using a gaseous substrate as an organic raw material, for example, a natural methane-containing gas, and as methane-oxidizing microorganisms in cultures such as Methylococcus capsulatus, Methylococcus, Methylosinus, Methymoncy and etc.
The use of microorganisms as biological agents for the production of biomass and its derivatives is an important component of biotechnology. Solving the problem of bioprotein production by microbiological means is a promising direction, since microorganisms are distinguished by a high protein content, balanced in amino acid composition. The high intensity of microbial metabolism is characterized by the rate of protein formation in bacteria, which is much higher than in plants and animals.
Under optimal fermentation conditions, methanotrophic bacteria actively assimilate natural gas methane, multiply rapidly and build up their mass, rich in valuable protein, a set of amino acids, vitamins and other biologically active substances. In the eighties, the VNIIsintezbelok Institute (now GosNIIsintezbelok) created a technology for producing an excellent protein-vitamin feed product from this biomass with the well-known domestic name - haprin. As a result of the isolation of active cultures of methanotrophs, as well as the achievement of certain successes in the development of continuous cultivation technology, it became possible to obtain relatively high yields of bacteria at such flow rates that provide an economically acceptable productivity of the process.
The development of bioprotein production in Russia will ensure independence from protein imports for feed purposes and create a national source of microbial protein that is not subject to climatic influences (see the Comprehensive program for the development of biotechnology in the Russian Federation for the period until 2020 (approved by the Government of the Russian Federation of April 24, 2012) No. 1853p-P8) At the same time, the development of the microbiological industry in general, and specifically the technology of continuous cultivation of bioprotein, is largely determined by the improvement of a set of necessary measures related to the development of fermentation methods and systems, the design of effective bioreactor designs, the development and acquisition of appropriate bacterial strains, the creation of the necessary seed culture and a suitable nutrient medium.
S, Grebennikova I. A. "Technology of microbial synthesis" A course of lectures in the specialty "Biotechnology." - Minsk: BSTU, 2014; Netrusov A.I. "Microbiology".
From the prior art, reflected in the patent bases of the Russian Federation and other countries, technical solutions are known related to the creation of new methods of fermentation of microorganisms, systems, apparatus and installations for their cultivation (see, for example, patents RU: 2064016, 2021350, 161690, 161691, 2580646, 786326, 2678425, 2562146,2626592, 2050788,2266676,2580646, 2144952, 2352626, 160091.2 699 293.2 654 109; US patents: 3957585, 4204042, 4656138, 4752564; EP patents: 1419834, GB 1353 30516, Eurasian patent No. 001948, etc.).
The study of the patent situation in relation to the claimed object is aimed at identifying the closest analogs in the fields related to the production of BVA and bioprotein in a broad sense, and, more specifically, feed protein in the form of biomass of methane oxidizing or methanassimilating aerobic microorganisms using natural methane-containing gas as organic raw materials.
12.05.1976).A known method of producing biomass, including the cultivation of microorganisms, such as yeast, on a nutrient medium containing paraffinic hydrocarbons as a carbon source, growth stimulants necessary for growth of mineral salts, selection of the culture medium, dividing it into fractions: solid - yeast and liquid, liquid return fractions for the initial stage of cultivation, maturation of yeast, additional separation of yeast into liquid and solid yeast fractions and drying of yeast. In order to increase the yield of biomass while improving its quality, the culture medium before separating it into fractions and the liquid fraction obtained after separation is kept at acidic pH (2.7-3.5), and after ripening (at pH 2.7-5 , 5) the yeast suspension is heated to 62-68 ° C for 5-15 minutes (see RF patent 506613, publ. 05/12/1976).
The disadvantage of the known technical solution is the fact that lowering the pH to 2.7-3.5 of the returned spent culture liquid allows to reduce the level of extraneous microflora in the process, but does not prevent the accumulation of products that inhibit the growing process. In addition, such treatment of the returned culture liquid will lead to a significant increase in the consumption of ammonia water to stabilize the pH of the process.
27.12.2005).Known technological line for the production of feed yeast. The line includes a block for growing yeast with separate selection of yeast suspension and after yeast brew, blocks of flotation thickening, heating the thickened suspension, its final concentration and drying the yeast, connected in series with it. A feature of the known technical solution is the connection of a flotation thickening unit with a growing unit, a return line of the floated post-yeast mash, and a yeast suspension heating unit is installed between the flotation thickening and final yeast concentration units, and the unit for heating it is made in the form of a mixer with hot water or post-yeast mash, and the block for final concentration of the yeast suspension is in the form of a settling tank (see RF patent No. 2266676, published on December 27, 2005).
The disadvantages of the known technical solution include the implementation of the yeast suspension heating unit in the form of a mixer with hot water and, given that flotation separation does not allow for a relatively high degree of thickening, additional thickening with hot water further reduces the concentration of thickened biomass, resulting in an increase in load at the subsequent stages of the production of feed yeast.
04.09.2019).A known method for producing biomass of methane-oxidizing bacteria, providing for their cultivation in a fermenter under aeration conditions on a nutrient medium containing methane as a carbon source, sources of nitrogen, phosphorus, mineral salts and trace elements, the separation of biomass from the culture liquid, partial return of the spent culture liquid to the growing stage , dehydration and drying of biomass. A feature of the known method is that the cultivation of biomass is carried out in the alternating aeration mode in order to activate molecular oxygen by using the reduced coenzyme NAD-H (nicotinamide adenine dinucleotide) with keeping the biomass in time without oxygen supply and with oxygen supply in a ratio of 1: 4 minutes to 1 : 6 minutes, and methane is continuously fed into the maximally turbulent aerated zone of the fermenter (see RF patent No. 2 699 293, published 09/04/2019).
The disadvantage of this method is the complication of the technological process of obtaining biomass of methane-oxidizing bacteria, associated with the use of the reduced coenzyme and the frequency of supply of oxygen to the biomass in a minute mode. Taking into account the fact that even with a biomass concentration of 7-8 g / l, the depletion of oxygen in the environment occurs within 20-30 seconds, so that the oxidation of methane without oxygen becomes impossible. Thus, the process will be periodically under anaerobic conditions, with increased foaming and increased release of metabolic products into the culture liquid. This will not allow to ensure high performance of the biomass production process; in addition, increased concentrations of metabolic products in the spent culture fluid (OCL) will require a decrease in the amount of returned OCF.
The closest technical solution to the proposed one is a method for obtaining biomass of methane-oxidizing microorganisms, including their continuous cultivation on a water-mineral nutrient medium with the supply of methane-containing gas and a gas containing free oxygen, using the spent medium after concentrating the biomass, and drying it (see. RF patent No. 2064016, published on July 20, 1996-prototype).
The peculiarity of the known method is that the process of growing microorganisms is carried out at a pressure in the growing medium, above atmospheric, with partial use in the process of growing the spent medium after concentrating the biomass, while the rate of supply of nutritional components of microorganisms to the process and the selection of a suspension containing biomass from the process of continuous cultivation of microorganisms is set in proportion to the flow supplied to stabilize the pH of the cultivation medium. The ratio of the flow of the spent medium returned to the growing process, after concentration, to the flow of the suspension withdrawn from the process is set so as to provide the maximum value of the flow supplied to stabilize the pH of the growing medium, and the pressure in the growing medium is set at a minimum level to ensure the optimal concentration of dissolved oxygen in the growing medium, but not lower than the pressure required for the transportation and energy utilization of gas streams leaving the process. A device that implements the known method includes means for preparing a nutrient medium, a bioreactor for growing microorganisms using methane-containing and oxygen-containing gases, means for concentrating and drying biomass.
In the known technical solution, one of the essential features is the partial use of the spent medium in the process of growing microorganisms after concentrating the biomass, which depends on the ratio of the flow of the spent medium returned to the growing process to the flow of the suspension taken from the process. The mentioned ratio depends on a number of factors influencing the process of growing microorganisms, such as the pH of the growing medium, the amount of dissolved oxygen, working pressure, etc. For these reasons, the device for implementing the known method is distinguished by a relatively complex system of automatic control of the parameters of the process of obtaining biomass of methane-oxidizing microorganisms. In addition, in the known technical solution, the composition of the spent medium returned to the growing process, in terms of the content of organic substances, can vary within significant limits (in terms of chemical oxygen consumption). Since the spent medium contains a significant amount of accompanying microflora, it is necessary to take into account the correspondence between the composition of the spent medium and the culture used, which is possible under the condition of additional purification of the spent medium before the start of the nitrification process at lower values of the chemical oxygen consumption.
The problem to be solved by the invention is to increase the technical and economic efficiency and competitiveness of the method for the production of biomass of methane-oxidizing microorganisms of cultures, for example, Methylococcus capsulatus, with the possibility of optimizing the process of cultivating the protein mass in terms of the productivity of the finished product and the environmental friendliness of its production.
The technical result achieved when solving the problem is to eliminate the shortcomings of the known technical solutions, increase the efficiency and productivity of industrial production of bioprotein while reducing costs associated, among other things, with the consumption of purified water per unit of finished product. This cost reduction is achieved by returning the spent medium or spent culture liquid (OKL) after concentrating the biomass in the nutrient medium during the industrial cultivation of microorganisms. An additional technical result consists in a significant reduction in discharges of liquid technological waste in the production of bioprotein.
The specified technical result is achieved by the fact that in the method for producing biomass of methane-oxidizing microorganisms, including their continuous cultivation on a water-mineral nutrient medium with the supply of methane-containing gas and gas containing free oxygen, using the spent medium after concentrating the biomass, and drying it, according to the invention, after concentrating the biomass, the spent medium is separated and cooled to a temperature of 30-32 ° C, then subjected to continuous aerobic fermentation, under conditions of aseptic cultivation of the accompanying microflora, followed by separation into biomass of the accompanying microflora and a liquid phase, which is returned to the water-mineral nutrient medium for continuous growing biomass of methane-oxidizing microorganisms and covering the consumption of fresh water.
In addition, the conditions for aseptic cultivation of the accompanying microflora at the stage of aerobic fermentation can be provided by mechanical and bacteriological sterilization of air, which serves as a source of oxygen, and the separated biomass of the accompanying microflora can be partially or completely directed to inactivation and drying of the biomass after its concentration.
The specified technical result is also achieved by the fact that in the line for the production of biomass of methane-oxidizing microorganisms according to any one of claims 1.2, containing a block for the preparation of an aqueous-mineral nutrient medium, a block for growing a pure culture with means for introducing a pure culture of methane-oxidizing microorganisms, a block for continuous cultivation microorganisms, including means for supplying methane-containing gas and gas containing free oxygen necessary for the growth of microorganisms in accordance with the needs of the selected culture, a biomass concentration unit adapted to use the spent medium and a biomass drying unit, according to the invention, the biomass concentration unit is provided with two outlets , the first of which is connected through the inactivation unit with the input of the biomass drying unit, and the second is connected through the spent medium cooling unit, the aerobic fermentation unit, under the conditions of aseptic cultivation of the accompanying microflora, and the bio separation unit. the mass of the accompanying microflora and the liquid phase, with the input of the unit for continuous cultivation of biomass of methane-oxidizing microorganisms, combined with the input for covering the flow of fresh water, and the separation unit is equipped with two additional outputs for connection with additional inputs of the unit for concentrating the biomass of methane-oxidizing microorganisms and the unit of aerobic fermentation.
Such an implementation of the method for producing biomass of methane-oxidizing microorganisms and lines for its production provide a solution to the problem, with the achievement of the specified technical result due to the fact that the biomass concentration unit is made with the possibility of continuous separate selection of the spent medium or spent culture liquid to form a loop of its forced return to the continuous unit. industrial cultivation of methane-oxidizing microorganisms through series-connected blocks: a block for cooling the spent medium, mainly to a temperature of 30-32 ° C, which is optimal for intensifying the growth of concomitant microflora, a block of aerobic fermentation under conditions of aseptic cultivation of concomitant microflora and a block for separating the concomitant microflora and liquid phase into biomass to cover the consumption of fresh water. At the same time, the separation unit is equipped with two additional outputs for connection with additional inputs of the unit for the concentration of biomass of methane-oxidizing microorganisms and the unit for aerobic fermentation. The presence of a block for cooling it to the specified temperature in the forced return loop of the ALC to the specified temperature provides optimal conditions for biological treatment of the ALC in the aerobic fermentation unit with the help of accompanying microflora, a separation unit that provides continuous purification at the stage of aerobic fermentation prior to the start of the nitrification of the ALC without contact with the environment. ...
The composition of the ALC, depending on the growing regimes at the stage of industrial cultivation of a methane-oxidizing culture, in terms of the total organic matter content in terms of chemical oxygen demand (COD), can range from 700 to 3000 mg / l. At the same time, the ALC contains the bulk of the accompanying microflora formed during the cultivation of the culture used. As a result, the process of biological treatment of ALC at the next stage of fermentation is a continuous aerobic process, when air that has undergone mechanical and bacteriological purification or oxygen is used as a source of oxygen. As a result, the cultures of microorganisms used for the oxidation of organic substances contained in the processed ALC correspond to the composition of the accompanying microflora when growing a production culture in the block for industrial growing of methane-oxidizing microorganisms.
In accordance with the inventive concept, the block of the additional stage of aerobic fermentation is a closed bioreactor of any known design, having a low energy consumption and functioning under conditions of aseptic cultivation of the accompanying microflora. At the same time, the depth of cleaning the ALC at the stage of aerobic fermentation is carried out, as indicated, before the start of the nitrification process, usually up to COD in the range of values of 100-200 mg / L. To ensure a stable quality of cleaning the ALC, for example, in the case of a sharp increase in the content of organic substances in the original AFC, the possibility of supplying additional biomass from the separation unit to the aerobic fermentation unit is provided.
The proposed implementation of the line for the production of biomass of methane-oxidizing microorganisms, as indicated, eliminates the disadvantages of the known technical solutions in terms of dumping excess ALC. These disadvantages are also inherent in technologies for the production of feed protein haprin, including the serial connection of blocks for the production of a pure culture of microorganisms, preparation of solutions of mineral nutrition, industrial fermentation, concentration of biomass, inactivation and drying of the finished product. The implementation of the proposed technical solution is directly related to an increase in the efficiency and productivity of industrial production, with a simultaneous decrease in the cost of the target product, due to the introduction into the process line of the return circuit of the spent medium through the aerobic fermentation unit to the industrial cultivation unit for methane-oxidizing microorganisms.
FIG. 1 is a block diagram of a line for the production of biomass of methane oxidizing microorganisms or feed protein.
The feed protein production line contains
The
The functioning of the proposed line for the production of feed protein is as follows.
The production of protein mass from methane-containing raw materials in the proposed line for the production of feed protein is associated with a number of stages, in which the preparation of the necessary components for the growth of biomass of methane-oxidizing microorganisms and its processing into the target product takes place. feeding solutions of nutrient salts, as well as methane-containing and oxygen-containing gases to block 3 of industrial production of methane-oxidizing microorganisms, in accordance with the needs of the selected culture, ensure the cultivation of biomass in the adopted modes. As indicated, a gaseous substrate, for example, natural methane-containing gas, can be used as organic raw materials, and cultures of Methylococcus capsulatus, Methylococcus, Methylosinus, Methylomonas, Methylocystis, etc. can be used as microorganisms.
Preparation of oxygen-containing (air) and carbon-containing raw materials (methane) is carried out in additional blocks, including means for separating air into oxygen and nitrogen, purification of methane-containing natural gas and their dosed supply to block 3 of industrial production of methane-oxidizing microorganisms (not shown). In most cases, natural gas is used - methane and / or methane-containing gases containing at least 90% methane. With regard to the use of oxygen-containing gas, it is necessary to take into account the following condition - the oxygen content in the gas should provide optimal specific values in relation to the current and predicted concentration of microorganisms in the culture liquid of the fermenter of
As indicated, in addition to methane (natural gas) and oxygen (air), an aqueous solution of ammonia, phosphoric acid, solutions of mineral nutrition elements and a biologically treated spent medium for dilution are continuously supplied to block 3 of the industrial production of methane-oxidizing microorganisms. In this case, the amount of biologically treated waste medium used for diluting the culture liquid is at least 90-95% of the total enterprise demand for process water. The speed of the liquid flow in the bioreactor of
A feature of methane-oxidizing bacteria is their high sensitivity to the concentration of these organic substances in the liquid phase, on which the accompanying microflora develops, partially reducing their level during its growth. In practice, a technologically interesting growth of a pure culture without accompanying microflora, which feeds on organic compounds contained in the liquid phase, is impossible. The species composition of the accompanying microflora depends on the growing regimes of the main crop and the content of methane homologues in natural gas. In this regard, approximately 70-90% of the main culture and 30-10% of the accompanying microflora are cultivated in industrial fermentation plants. For this reason, a decrease in the level of metabolic products in the returned culture fluid, as a result of its processing in
Continuous selection of the spent culture liquid from the
In the known technical solutions, the separated spent medium, or OLC, constitutes up to 95% of the initial flow through the bioreactor. As indicated, the ALC contains exocellular metabolic products and background concentrations of mineral salts. In addition, it should be borne in mind that the ALC contains up to 70-100 mg / l of phosphorus (the required degree of purification to MPC in the reservoir is 2 mg / l), ammonium nitrogen 40-100 mg / l (MPC in the reservoir is 0.5 mg / l ), sulfates - more than 100 mg / l (maximum concentration limit in the reservoir is 100 mg / l). Therefore, cleaning the OKL for its disposal is not only technically difficult, but also very costly. In addition, during the release of phosphorus, a fairly large amount of sludge is formed, the disposal of which is also a problem. Typically, the concentration of biomass in the suspension at the bioreactor outlet is 1.5-3.0%, depending on the mode of its operation. Thus, in the case of large-scale industrial production of bioprotein, the cost of 1 m 3 of fresh water is 3-4 times higher than the cost of 1 m 3 of natural gas, which, in a direct-flow mode, significantly affects the cost of production of fodder protein.
In order to exclude the accumulation of exocellular metabolic products and oxidation products of methane homologues, when returning to the biomass growing process, the ALC is subjected to biological treatment in
The expediency of using the proposed functioning of the return circuit of the OKL in
To intensify the growth of the accompanying microflora at the stage of aerobic fermentation in
The composition of the ALC, depending on the operating modes of the bioreactor, at the first stage of growing a production culture in
