Abstract
Description
The invention relates to a method for producing an antibacterial material for use as hygienic and protective materials, in particular in the production of antibacterial dressings in field hospitals.
[3]Poly-L-lactic acid is a biodegradable polymer material with good biocompatibility, controlled degradation rate and required mechanical strength, well suited as a base material for dressings. Poly-L-lactic acid is not antibacterial. To impart antibacterial properties to products made of poly-L-lactic acid, methods of immobilization of various components with antibacterial properties are used, both on the surface and inside the product. [one]. Zinc oxide is an antibacterial material with good biocompatibility, inhibits the growth of bacteria, viruses and fungi, and promotes the metabolism of human skin. [2]. The most practical method for obtaining antibacterial materials based on poly-L-lactic acid and zinc oxide nanoparticles is the electrospinning method. The fabric made using this method has a large surface area, high porosity, which leads to an increase in the rate of wound healing and is attractive for applications in biomedicine, for scaffolds in tissue engineering, drug delivery systems, and dressings. [3]
[4].There is a known method of producing an antibacterial material based on polylactic acid fibers and zinc oxide nanoparticles (0.9-13.2 wt.%), Which consists in immobilizing zinc oxide nanoparticles on the surface of polylactic acid fibers by soaking polylactic acid nanofibers in a dopamine solution for 1 -10 minutes with the formation of polydopamine on the fiber surface, followed by soaking the resulting material in a dispersion of zinc oxide nanoparticles for 1-10 minutes. To control the content of zinc oxide nanoparticles, the processes of soaking nanofibers in a dopamine solution and in a dispersion of zinc oxide nanoparticles can be repeated many times with cleaning steps between soaking cycles. The obtained composite materials based on polylactic acid had good antibacterial properties and good biocompatibility. [four].
The disadvantages of this method can be attributed to the laboriousness of the manufacturing procedure, due to the repeated stages of soaking the polymer in a solution of dopamine and zinc oxide nanoparticles to achieve the required concentration on the fiber surface. In addition, it is difficult to track the amount of zinc oxide applied to the fiber surface.
[5].There is a method of obtaining an antibacterial material by the method of sequential impregnation of a polylactic acid web obtained by electrospinning with an aqueous colloidal solution of zinc oxide nanoparticles, including successive multiple stages of impregnation and drying at 20 ° C until the concentration of zinc oxide nanoparticles on the surface of the web reaches 1 mg / cm 2 . The obtained materials were tested as model dressings with bactericidal properties in relation to the gram-positive strain of Staphylococcus aureus and the gram-negative strain of Escherichia coli. [five].
The main disadvantage of this method is the laboriousness of the application procedure, which consists in multiple stages of impregnation and drying.
Surface treatment of polylactic acid fibers with an antibacterial component is most often used to obtain materials with antibacterial properties. However, processed materials often do not have adequate antibacterial efficacy because the antimicrobial material tends to fall off or wear off during storage. This problem can be solved by the direct introduction of zinc oxide nanoparticles into the polymer fiber, which will also make it possible to achieve a long-term (prolonged) antibacterial effect due to the gradual decomposition of polylactic acid or the release of zinc oxide nanoparticles.
[6]A known method of producing an antibacterial material by the method of electrospinning of a spinning solution, which is a suspension of nanoparticles of zinc oxide and polylactic acid in chloroform, with a component content of 77 wt. % polylactic acid and 23 wt. % four types of commercial zinc oxide nanoparticles with different surface treatments, rod-shaped, 10-3 nm in diameter and 100 nm in length. The materials obtained showed antimicrobial activity against the pathogenic microorganism of Staphylococcus aureus. [6]
The main disadvantage of this method is the selection of a solvent and surface treatment of nanoparticles in order to create a stable suspension for electrospinning and obtain fibers with the required strength characteristics and antibacterial properties.
Of the proposed, the closest to the claimed technical solution in terms of the technical essence and the achieved technical result is a method for producing an antibacterial material by electrospinning a solution of polylactic acid in trifluoroethanol containing 1-5 wt. % of zinc oxide nanoparticles synthesized by the microwave solution method. Antibacterial activity was assessed by direct contact with 4 ml of
The disadvantages of this method include the use of an expensive solvent trifluoroethanol for the preparation of a spinning solution. The complexity and multistage procedure for the synthesis of zinc oxide nanoparticles using an autoclave and a microwave reaction system, additional stages of washing and drying nanoparticles to obtain a nanopowder.
The present invention is based on the problem of creating a method for producing an effective antibacterial material by electrospinning, having a high ability to suppress the growth of pathogenic microorganisms, due to the inclusion of antibacterial nanoparticles of zinc oxide obtained by pulsed laser ablation inside the fiber of poly-L-lactic acid. The method of pulsed laser ablation of a zinc target in air is simple in hardware design, and zinc oxide nanoparticles obtained by this method have an active defective surface, without additional stabilizers, which contributes to an increase in the antibacterial effect. Nanoparticles are immediately obtained in the form of a dry powder, which eliminates the stages of drying and calcining the active (antibacterial) component. The purity of the material allows it to be used for biomedical purposes. The zinc oxide nanoparticles obtained in this way are well dispersed in organic solvents, which makes it possible to obtain suitable solutions for electrospinning with the required properties.
Электроспиннинг осуществляют из раствора, включающего хлороформ, поли-L-молочную кислоту, наночастицы оксида цинка, полученные методом импульсной лазерной абляцией цинка в воздухе, на установке с цилиндрическим коллектором с частотой вращения 50 об/мин, при расстоянии от инжектора до сборочного коллектора 170 мм, скорости подачи раствора 6 мл/час, напряжении на инжекторе 27 КВольт, при температуре 25°С и относительной влажности 44 %, с последующей температурной обработкой материала на сборочном коллекторе при 100°С в течение 12 часов, при следующем соотношении компонентов, масс.%:The problem is solved by the fact that the method for producing an antibacterial material includes the method of electrospinning a solution of poly-L-lactic acid with zinc oxide nanoparticles in an organic solvent. An example of a specific use of the invention is given below.
1.Example 1.
In a sealed glass reactor, 93 grams of chloroform of grade O.S. Ch (Ekros-1, Russia) was placed, then 1.2 grams of zinc oxide nanoparticles obtained by pulsed ablation of zinc in air were added. The reactor was closed and placed in an ultrasonic bath for 10 hours at room temperature. Then 4.8 grams of PL-10 grade poly-L-lactic acid (Corbion) was added to the reactor. The rector was closed and placed in an ultrasonic bath for 20 hours at room temperature. The resulting solution was subjected to electrospinning at an injector voltage of 27 KV, a spinning solution feed rate of 6 ml / h, a chamber temperature of 25 degrees relative humidity of 44%, a distance from the injector to an assembly manifold of 170 mm, and a manifold rotation frequency of 50 rpm. To obtain materials of each type, 40 ml of a spinning solution were used. After the formation of materials, the assembly manifold was subjected to heat treatment at a temperature of 100 degrees for 12 hours. Then the assembly manifold was removed and cooled to room temperature. Thus, an antibacterial material has been obtained, which is a web of randomly interwoven poly-L-lactic acid fibers containing zinc oxide nanoparticles incorporated into the polymer fiber at a concentration of 0.25 mg / cm 2 or 20 wt%. Using the antibacterial activity test against S.aureus and E.coil, the antibacterial activity value was found to be 2.55 and 0.98, respectively.
