Abstract
Description
The invention relates to biotechnology, namely to improve the stability and reproducibility of electrical parameters of biological sensors based on reduced graphene oxide. A method for increasing the stability and reproducibility of the characteristics of a biological sensor based on reduced graphene oxide includes the formation of a graphene oxide film on a polymer substrate and further local modification of the oxide-graphene film according to a given pattern with different processing parameters of the sensitive area and the electrically conductive channel. The graphene oxide film is formed by the methods of drop, centrifugal or aerosol deposition. In this case, the modification of the graphene oxide film is produced by laser radiation with the fluence parameters significantly different in the case of a sensitive region and a conducting electrode. The sensitive area of the sensor is formed with minimal fluence parameters, which, on the one hand, ensures the presence of the maximum number of oxygen-containing functional groups, and on the other hand, the formation of conductive channels. The invention makes it possible to form biological sensors with maximum stability of parameters, as well as to ensure the maximum degree of immobilization of biosensitive agents due to the minimum degree of restoration of the sensitive area.
There are known methods for modifying a surface with electromagnetic radiation from the ultraviolet to infrared range, including visible light, and both incoherent radiation sources and lasers of various types can be used for modification: both continuous action and pulsed, and constant action) with different wavelengths and pulse duration. Also, in general, the described methods are aimed at creating highly conductive and / or optically transparent regions of reduced graphene oxide.
The paper [1] describes a method for reducing graphene oxide using a focused beam of sunlight, as well as using focused radiation from an incandescent lamp. With this use of sunlight (wavelengths 250 - 2500 nm), the measured radiation flux density was from 1.77 to 2.03 W / cm 2 . At the indicated values of the flux density, the temperature rises to 150-200 ° C in 1-2 s. At the same time, the presence of a D-band (1360 cm -1 ), a G-band (1570 cm -1 ) with the ratio ID / IG = 0.20 is observed in the Raman spectra, which is quite close to graphite (ID / IG = 0.07 ) in this case, the conductivity of such samples is 1.494 × 10 3 S / m, which is also similar to graphite (10 5 S / m), while the conductivity of graphene oxide is approximately 8 orders of magnitude lower (~ 10 -5 S / m). The thickness of the graphene film ranged from 0.9 to 1.4 nm. In the case of artificial light, a 100 W halogen incandescent lamp was used, the radiation of which passed through a bandpass filter providing wavelengths in the range of 580-700 nm. In this case, the luminous flux power was about 2 W. With this effect, the temperature in the irradiated area reaches 175 ° C, and the film changes its color from black to brown, as in the case of exposure to sunlight. However, as can be judged from the description, the disadvantage of this method is the impossibility of controlling the parameters of the resulting reduced graphene oxide with high accuracy and repeatability. In addition, the light spot in this case has a sufficiently large size, which does not allow full localization, and, for example, integration into microelectronic processes. In addition, in this case, there is no possibility of controlling the type of functional groups present in the regions of reduced graphene oxide.
In [2], a method for forming a pattern of reduced graphene oxide and a method for producing an electrical circuit based on reduced graphene oxide are described. Due to their high transparency, these schemes are primarily offered for use in touch screens. A graphene oxide film is applied onto a pretreated substrate by spin-coating at a rotation speed of 400 - 700 rpm. To reduce graphene oxide, this patent uses a 248 nm nanosecond laser (
Closest to the proposed method is the method described in [3]. Namely, a method for creating a conductive and partially transparent layer based on reduced graphene oxide, including depositing a graphene oxide layer on a substrate and heating a portion of the graphene oxide layer using laser radiation with a power of 0.036 W, which causes the chemical reduction of graphene oxide to graphene with a concomitant decrease in thickness films due to ablation. The result of this approach is the production of a graphene oxide layer with the required parameters of transparency and conductivity. The possibility of regulating the resulting resistance by changing the laser power and exposure time at a point is shown. The disadvantage of this work is the focus on obtaining such electrophysical indicators as electrical resistivity and optical transmittance, which is important for creating transparent conductive coatings, but does not solve the problem of forming a transducer for chemical or biological sensors.
The specific features of the reduction of graphene oxide in these patents are the formation of a graphene oxide film from solutions with a wide range of film thicknesses on various substrates. In this case, the main attention is paid to the conductivity (resistance) of the formed regions of reduced graphene oxide.
The object of the present invention is to provide the formation of a conductive region of reduced graphene oxide with a different amount of residual functional, in particular carboxyl, groups in different places of the conductive region. In this case, the total resistance of the conducting area should be no more than 50 kOhm. The presence of residual functional groups is required to ensure further binding of sensitive substances with reduced graphene oxide, thus forming a sensor transducer.
To ensure local reduction of graphene oxide in order to form conductive regions in electronic devices, the latter must be applied in a uniform layer on the substrate surface.
The general sequence of operations for the formation of the reduced graphene oxide region consists of the following steps:
1) Preliminary mechanical cleaning of the substrate using 2-propanol (ultrapure grade) followed by washing in a stream of deionized water.
2) Drying of the substrate surface in a stream of purified air or nitrogen at a pressure of 1-3 atmospheres.
3) Preparation of a graphene oxide solution of a given concentration.
4) Application of a solution of the specified concentration. The number of iterations and the concentration of the solution are selected based on the required film thickness and application method.
5) Heat treatment (drying) of the substrate with solvent evaporation.
6) Placement of the substrate with the formed film into the working chamber of the laser modification unit.
7) Local reduction of graphene oxide with parameters providing the minimum degree of reduction. The parameters of laser radiation are selected in such a way as to ensure the minimum degree of recovery on the one hand (which ensures the presence of a large number of functional groups), and on the other hand, to ensure the formation of conducting channels and minimum conductivity.
8) Local reduction of regions of graphene oxide with a high degree of reduction, and these regions partially overlap the region with a minimum degree of reduction. The high degree of recovery ensures high conductivity and repeatability of the conductivity parameters during the formation of large batches of conductive structures based on reduced graphene oxide.
9) Control of the degree of reduction of graphene oxide is carried out by one or more of the following methods: IR spectroscopy, Raman spectroscopy, X-ray photoelectron spectroscopy, measurements of the contact angle of wetting.
As a substrate, both flexible polymer substrates can be used, preferably such as polyethylene terephthalate (PET) or polyethylene naphthalate (PEN), both providing relatively high physicomechanical and thermal characteristics, and solid substrates, namely silicon with an oxide layer, quartz or glass ... As deposition methods, drop-casting, spray-coating, spin-coating, inkjet printing, etc. can be used. with fomrization of a film with a thickness of 0.03 to 3 microns. For the reduction of graphene oxide, both continuous and pulsed lasers can be used, and the pulse length can vary over a wide range from microseconds to femtoseconds.
Thus, a method is proposed for forming the structure of reduced graphene oxide, which consists in the reduction of graphene oxide films with a thickness of up to 1000 nm using a microsecond laser with a wavelength of 442 nm and a power of 600 mW. The formation of a graphene oxide film with a thickness of up to 1000 nm on a flexible polymer substrate is performed in several iterations by the method of dropwise deposition of a graphene oxide solution. The patterning of the reduced graphene oxide region is performed using a microsecond laser with a wavelength of 442 nm and a power of 600 mW, and when the pattern of the restored region is formed, the laser fluence is changed so as to obtain a weakly reduced conducting region of reduced graphene oxide at low fluence values and a region with a high degree of reduction at the maximum value of the fluence. The restoration is carried out under the condition of exposure to one pulse at one point of the topological pattern. The minimum value of the fluence of 2.5 μJ / cm2 provides the maximum number of functional groups, which is required to form the sensitive area of the sensor structure, while the maximum value of the fluence of 93.4 μJ / cm2 provides the maximum structuredness of the topological pattern area, thus providing high conductivity required to create conductors and interconnections. To minimize possible oxidative reactions, the reduction is carried out in an inert gas atmosphere.

In the exemplary embodiment, PET substrates with a thickness of 175 μm and dimensions of 40 × 40 mm were used. In an exemplary embodiment, a graphene oxide film was deposited onto a flexible substrate by drop deposition with the formation of a film with a thickness of about 1 μm. The thickness of the film is limited by the minimum thickness, which is about 0.03 microns, since When using a 442 nm pulsed laser with a thinner film, not only ablation of the graphene material occurs, but also thermal destruction and melting of the substrate.
In the example shown, a pulsed microsecond laser with a wavelength of 442 nm and a power of 600 mW was used. The size of the laser spot was 40 × 15 μm, 1 pulse fell on one point. The pulse duration in the case of the minimum degree of recovery was 2.5 μs, which corresponds to a fluence of 2.5 μJ / cm 2 . In the case of a high degree of recovery, the pulse duration was 150 μs, which corresponds to a fluence of 93.4 μJ / cm 2 . The specific topological drawing of the restored areas is selected depending on the specific task and is set in the programs for controlling the laser installations or in graphic editors compatible with such programs.
After the formation of the film, U-shaped topological patterns of the blanks of the sensor structures were formed, where the bridge was restored with the minimum parameters of the fluence, while the remaining areas - with the maximum (Fig. 1). A study was carried out of the properties of the obtained material, namely the degree of graphitization of the presence of defects in the crystal lattice of graphene (including functional groups) by obtaining Raman spectra from the indicated regions (Fig. 2). The conductivity (resistance) of the formed regions was also measured both in the case of restoring the entire topological pattern with the same fluence, and when restoring from different bridges and other regions with different fluence parameters given earlier (Fig. 3). From the data shown in FIG. 3, it follows that upon restoration of the bridge at a low fluence, as well as further restoration of the remaining regions of the topological pattern with partial overlap of weakly and strongly restored regions, the resistance of the resulting structures decreases by 2-3 orders of magnitude and amounts to 4-12 kΩ. When recovering with the same fluence, the resistance of the structures is from 200 to 5000 kOhm.
[4].An analysis of the Raman spectra revealed the following correlation between the structure resistance and the obtained Raman spectra: at the lowest fluence, the Raman spectrum has the form typical of graphene oxide with practically the same intensity of D and G peaks, as well as an unexpressed 2D peak (Fig. 2). In the case of a high degree of recovery, the intensity of the 2D peak increases significantly, which indicates the formation of crystal structures consisting of several layers in the composition of the reduced graphene oxide film (Fig. 2). In addition, the D peak decreases, which is typical for reduced graphene oxide and indicates the removal of some defects, which, in the case of reduced graphene oxide, leads to a decrease in the amount of oxygen-containing functional groups [4].
The presence of functional groups can be anticipated by IR spectroscopy. FIG. 4 shows the Fourier-IR spectra of the structure formed at a fluence of 2.5 μJ / cm 2 . FIG. 4 shows a distinct peak at ~ 1720 cm -1 , which, according to publications [5, 6], is responsible for carboxyl groups.
Thus, we can say that at low fluence values, it is possible to ensure the formation of a reduced graphene oxide film, which is conductive on one side, and on the other, contains functional, in particular carboxyl, groups, which makes it possible to set the reduction parameters in such a way that to obtain regions of reduced graphene oxide suitable for use in various sensor structures, where covalent bonding of a sensitive substance with an electrically conductive region is required.
