Берут 1 г мицелия гриба трутовика лекарственного (Fomitopsis of ficinalis Will. Bond. Et Sing.), помещают в плоскодонную колбу с обратным холодильником, приливают 60 мл изопропилового спирта. Смесь охлаждают, фильтруют через стеклянный фильтр, получают экстракт 1. Остаток повторно экстрагируют с 30 мл изопропилового спирта в тех же условиях, получают экстракт 2. Полученные экстракты 1 и 2 объединяют и упаривают на роторном испарителе (RV 8 Flex, IKA, Германия). Нерастворимый остаток высушивают в сушильном шкафу до постоянной массы при +50°С. Получают кристаллический порошок белого цвета с легким желтоватым оттенком, без запаха, практически не растворим в воде, растворим в этаноле 95%. Взвешивают. Идентификацию полученного вещества, как агарициновой кислоты, проводили метод масс-спектрометрического анализа с помощью матрично-активированной лазерной десорбции/ионизации (МАЛДИ) с использованием масс-спектрометра Ultraflex III MALDI-TOF/TOF (Bruker, Германия). Для этого полученные образцы растворяли в этаноле и центрифугировали в течение 10 мин при 10000 об/мин, пропускали через фильтр (размер пор 5 мкм). 1 мкл экстракта смешивали с 1 мкл матрицы, наносили на мишень и высушивали на воздухе. В качестве матриц были использованы 2,5-дигидроксибензойная кислота (10 мг/мл в 33% водном ацетонитриле), альфа-цианокоричная кислота (насыщенный раствор в 33% водном ацетонитриле), 3-гидроксипиколиновая кислота (10 мг/мл в 33% водном ацетонитриле), а также применялся метод нанесения экстрактов на мишень при отсутствии матрицы. Наилучшие по чувствительности результаты были достигнуты при использовании в качестве матрицы 2,5-дигидроксибензойной кислоты в режиме регистрации отрицательных ионов. Агарициновая кислота была идентифицирована по наличию пика депротонированной молекулы [M-H]- при m/z 415.2701±10 ppm (см. фиг. 1, фиг. 3) и пикам фрагментных ионов при m/z 397, 379, 353, 335, 309 и 281 в режиме МС/МС (режим высокоэнергетичной столкновительной диссоциации, high energy CID) (см. фиг 2). Как видно из представленных результатов, заявляемый способ значительно упрощает процесс получения агарициновой кислоты по сравнению с прототипом, не требует дополнительной очистки, так как сразу обеспечивает получение чистого целевого продукта, имеющего степень чистоты 97,8 – 99,0%. Кроме того, заявляемый способ позволяет повысить выход агарициновой кислоты в 2,1 раза.
