Проблема разработки эффективных средств для лечения онкологических заболеваний остается высоко актуальной для всего мирового сообщества. Так, в Российской Федерации в настоящее время на учете с различными онкологическими заболеваниями состоит около 3,3 миллионов больных, нуждающихся в эффективном лечении. Но, несмотря на существенные успехи онкологии последних десятилетий, многие распространенные онкологические заболевания, по-прежнему, с трудом поддаются лечению. Например, рак молочной железы, ежегодно поражающий свыше 65 тысяч женщин в России, становится причиной смерти 35% заболевших, а летальность рака легких, которым ежегодно заболевает около 60 тысяч россиян, достигает 85% (Каприн А.Д., Старинский В.В., Петрова В.Г. Токсичность соединения Т1084 исследована на самцах белых беспородных мышей по тесту «острой» токсичности в возрасте 2-2,5 месяца с массой тела 22-25 г при однократном внутрибрюшинном введении (Руководство по проведению доклинических исследований лекарственных средств. Часть первая / под ред. А.Н. Миронова - М.: Гриф и К, 2012). Рабочие растворы Т1084 изготавливали на основе 0,9% асептического раствора хлорида натрия (Дальхимфарм, РФ). Наблюдение за животными осуществляли в течение 14 суток. Исследование выполнено на 81 самке мышей-гибридов F1 (СВА × C57Bl/6j) в возрасте 2-2,5 месяцев с массой тела 19-22 г, распределенных методом рандомизации в 4 экспериментальные группы: контрольную и три опытные, по 20-21 особи в каждой. Всем животным трансплантировали КЭ по методике, описанной в Руководстве по проведению доклинических исследований лекарственных средств (2012). Контрольные животные в дальнейшем не получали каких-либо экспериментальных воздействий. Животным опытных групп с 6-х по 20-е сутки роста КЭ проводили ежедневое в/б введение, соответственно, Т1023, Na-ДХА и Т1084 в эквимолярных дозах: Т1023 - 60 мг/кг; Na-ДХА - 33,6 мг/кг; Т1084 - 70,7 мг/кг. Растворы Т1023 и Т1084 изготавливали на основе 0,9% асептического раствора хлорида натрия (Дальхимфарм, РФ), растворы Na-ДХА изготавливали на основе воды для инъекций (Дальхимфарм, РФ). Противоопухолевые эффекты изучали по методике, описанной в Руководстве по проведению доклинических исследований лекарственных средств (2012).The study was performed on 81 female F 1 hybrid mice (CBA × C 57 Bl / 6j) aged 2-2.5 months with a body weight of 19-22 g, randomized into 4 experimental groups: control and three experimental, 20 -21 individuals in each. Antitumor effects were studied according to the method described in the Guidelines for conducting preclinical studies of drugs (2012).

Практически в течение всего срока наблюдения действие этих соединений сопровождалось статистически достоверным противоопухолевым эффектом. Максимальный противоопухолевый эффект развивался к концу первой недели применения Т1023 и Na-ДХА, и проявлялся торможением роста КЭ примерно на 40%. Но в последующем противоопухолевая эффективность Т1023 и Na-ДХА прогрессивно снижалась и на поздних сроках индекс ТРО составлял 21-24% (около 50% от максимальной эффективности).The results of this experiment showed that, with the administration schemes and doses of T1023 and Na-DCA, they had a similar effect on the development of CE (Table 3). И в эти сроки противоопухолевый эффект Т1084 на уровне статистической тенденции (р=0,12) на 20-25% превышал эффекты Т1023 и Na-ДХА. А в дальнейшем динамика противоопухолевого действия соединения Т1084 качественно отличалась от действия Т1023 и Na-ДХА. Если на поздних сроках торможение роста КЭ под влиянием Т1023 и Na-ДХА прогрессивно ослабевало, то действие Т1084 подавляло адаптацию опухоли к такому воздействию и поддерживало до конца наблюдения стабильное торможение роста опухоли на уровне 45-50% (90-100% от максимальной эффективности). И при этом на поздних сроках противоопухолевый эффект Т1084 статистически достоверно превосходил эффекты Т1023 и Na-ДХА.The initial stages of development of the antitumor effect of the compound T1084 were similar to the action of T1023. Целью эксперимента являлось сравнительное изучение влияния Т1023, Na-ДХА и Т1084 при их хроническом применении на рост рака шейки матки (РШМ-5) у мышей.The aim of the experiment was a comparative study of the effects of T1023, Na-DCA, and T1084 in their chronic use on the growth of cervical cancer (cervical cancer) in mice.
Исследование выполнено на 74 самках мышей-гибридов F1 (СВА × C57Bl/6j) в возрасте 2-2,5 месяцев с массой тела 19-22 г, распределенных методом рандомизации в 4 экспериментальные группы: контрольную и три опытные, по 18-20 особей в каждой. Всем животным трансплантировали РШМ-5 в область латеральной поверхности правого бедра путем подкожного введения 2⋅106 опухолевых клеток в 0,1 мл суспензии на основе среды 199 (Пан-Эко, РФ). Контрольные животные в дальнейшем не получали каких-либо экспериментальных воздействий. Как и в Примере 4, животным опытных групп с 7-х по 21-е сутки роста РШМ-5 проводили ежедневое в/б введение Т1023, Na-ДХА и Т1084 в эквимолярных дозах - 60, 33,6 и 70,7 мг/кг, соответственно. Но в последующем неоплазия стремительно приобретала практически полную резистентность к действию этих соединений. Уже через 5-7 суток от начала применения Т1023 и Na-ДХА ингибирование роста РШМ-5 резко ослабевало, и до конца наблюдения их противоопухолевые эффекты полностью отсутствовали даже на уровне статистической тенденции.The results of this experiment showed that the used experimental neoplasia was weakly sensitive to the action of T1023 and Na-DCA (Table 4). Заявленный технический результат предлагаемого изобретения заключается в обеспечении более выраженного и стабильного противоопухолевого эффекта соединения Т1084 в сравнении с прототипами - гидробромидом 1-изобутаноил-2-изопропилизотиомочевины (соединение Т1023) и натрия дихлорацетатом (Na-ДХА), а также в подавлении (или торможении) развития резистентности неоплазий.The claimed technical result of the invention is to provide a more pronounced and stable antitumor effect of the compound T1084 in comparison with the prototypes 1-isobutanoyl-2-isopropylisothiourea hydrobromide (compound T1023) and sodium dichloroacetate (Na-DCA), as well as inhibition (or inhibition) development of neoplasia resistance.
В приведенном эксперименте (см. пример 4) установлено, что сочетанное применение ингибитора NOS Т1023 и Na-ДХА сопровождается более выраженным и стабильным противоопухолевым действием на солидную КЭ мышей и подавляет развитие резистентности карциномы к действию Т1023 и Na-ДХА. Полученный результат свидетельствует, что два однонаправленных (противоопухолевых) вида фармакологической активности - NOS-ингибирующее антиангиогенное действие Т1023 и PDKl-ингибирующее гипоксия-ориентированное цитотоксическое действие ДХА, способны к фармакодинамическому синергическому взаимодействию. И в этой связи, сочетание этих видов фармакологической активности, реализуемых по различным механизмам, является перспективным подходом для разработки эффективных комплексных средств и методов таргетной терапии онкологических заболеваний. Совокупность приведенных данных свидетельствует о целесообразности синтеза соединения Т1084 - дихлорацетата 1-изобутаноил-2-изопропилизотиомочевины (см. примеры 1,2).In the given experiment (see example 4), it was found that the combined use of the NOS T1023 inhibitor and Na-DCA is accompanied by a more pronounced and stable antitumor effect on solid CE of mice and inhibits the development of carcinoma resistance to the action of T1023 and Na-DCA. And in this regard, the combination of these types of pharmacological activity, implemented by various mechanisms, is a promising approach for the development of effective integrated tools and methods for targeted therapy of cancer. The combination of the above data indicates the feasibility of the synthesis of the compound T1084 - dichloroacetate 1-isobutanoyl-2-isopropylisothiourea (see examples 1,2).
В токсикологических исследованиях установлено (см. пример 3), что замена солеобразующей кислоты с гидробромида (Т1023) на дихлорацетат (Т1084) не изменяет «острую» токсичность солей 1-изобутаноил-2-изоприлизотиомочевины, и, формально, не повышает риски негативных эффектов в случае применения Т1084 в качестве лекарственного средства. При этом в исследованиях на двух опухолевых моделях -солидной карциноме Эрлиха (КЭ; см. пример 4) и раке шейки матки (РШМ-5; см. пример 5) у мышей соединение Т1084 проявляло существенно более выраженную и стабильную противоопухолевую активность в сравнении с Т1023 и Na-ДХА.
