[1].Known nutrient medium for stationary cultivation of Bacillus subtilis - meat and peptone broth (MPB). The meat-peptone broth is prepared from meat water, to which 1% peptone and 0.5% sodium chloride are added. Meat water is obtained by boiling in 2-4 times the volume of tap water of meat or minced cattle or horses for 1 hour, followed by filtration [1].
The disadvantage of this environment is the high cost of the components used and the complex cooking technology. The use of this medium for industrial production is difficult.
[2].The closest analogue for the cultivation of Bacillus subtilis to the claimed nutrient medium is L-broth. It is prepared as follows: composition (g / l): yeast extract - 5, enzyme peptone - 15, sodium chloride - 5, distilled water - up to 1 l (pH 6.8-7.0). Sterilization was carried out at a pressure of 1.1 atm. within 40 minutes [2].

To achieve the technical result of the cultivation of Bacillus subtilis VKPM B-12079, it was proposed to use a nutrient medium consisting of a yeast extract, enzymatic peptone, sodium chloride, 96% alcoholic extract of Arabica coffee, distilled water, brought to a volume of 1 l (pH 6.8 -7.0), with the following ratio of components (g / l): yeast extract - 5, peptone - 15, sodium chloride - 5, 96% alcohol extract of Arabica coffee 2-40 ml / l, distilled water - up to 1 l. (pH 6.8-7.0). Sterilization at a pressure of 1.1 atm. for 40 minutes.
The preparation of the extract includes the extraction of plant materials. In this work, we used the Soxhlet apparatus. For 1 g of air-dry substance used 15 ml of solvent. As solvents, a 96% ethanol solution was chosen. 10 g of air-dried plant material was added to a cartridge made of white filter paper, after which it was closed on both sides and placed in a Soxhlet extractor. 150 ml of solvent was added to the flask and extraction began in a water bath. The extraction time was 3 hours, the number of cycles was 10. The alcohol extract of the plant material was obtained at the output.
The experience is illustrated by the following examples.
1.Example 1
Затем, в остывшую жидкую питательную среду (объем 50 мл) в стерильных условиях вносили навеску 0,5 г лиофилизированно высушенных спор Bacillus subtilis ВКПМ В-12079.To prepare a liquid nutrient medium, medium components were added to an Erlenmeyer flask (volume 50 ml), with a ratio of g / l: yeast extract - 5, peptone - 15, sodium chloride - 5, certain volumes of Arabica coffee extracts (see table 1), distilled water - up to 1 liter (pH 6.8-7.0). Closed with a cotton-gauze plug and sterilized at a pressure of 1.1 atm. for 40 minutes. Then, in a cooled liquid nutrient medium (volume of 50 ml) in sterile conditions weighed 0.5 g of lyophilized dried spores of Bacillus subtilis VKPM B-12079.
Cultivation was carried out in an Innova 44 shaker incubator (New Brunswick, USA) at a rotation speed of 250 rpm (eccentricity 5 cm), at a temperature of 37 ° C for 24 hours.
To determine the amount, we used the standard method of ten-fold dilutions with reseeding on nutrient agar prepared in the following ratio of components (g / l): yeast extract - 5, peptone - 15, sodium chloride - 5, agar-agar - 18, distilled water - up to 1 l (pH 6.8-7.0). Sterilization was carried out at a pressure of 1.1 atm. for 40 minutes. Then it was thermostated at a temperature of 37 ° C for 24 hours. As a control sample, medium was used without adding Arabica coffee extracts. The experiments were carried out seven times.
From the above data it is seen (Table 1) that the maximum increase in the number of bacteria is achieved by adding 20 ml of a 96% alcoholic extract of Arabica coffee to L-broth.
Thus, the proposed nutrient medium for the cultivation of the bacterium Bacillus subtilis VKPM -12079 contains available raw materials, and its use allows to increase the number of microorganisms to 2 × 10 13 CFU / ml.
