Известно устройство для сбора личинок и мелких нематод из фекалий (а.с. №1391625). Устройство содержит чашу 1 со сбегающей боковой поверхностью в виде крыльчатки 2. На поверхности крыльчатки и чаши имеются борозды 3, с наружной стороны дна между крыльями - полукруглые вырезы 4. Чаша 1 имеет вид полусферы с гладкой внутренней поверхностью. Устройство погружается в фекалии, в чашу 1 наливается вода. Колпачок с целью сбора личинок и мелких нематод из фекалий используется следующим образом. В чашку Петри устанавливают колпачок отверстием вверх. Вокруг него помещают исследуемые фекалии так, чтобы они плотно прилегали к боковым поверхностям корпуса 1 со всех сторон и по всей его высоте. Внутреннюю емкость колпачка заполняют водой до самого верха. Закрывают чашку Петри. Гельминты, активно передвигаясь в фекалиях, в поисках более чистой среды поднимаются по бороздам 3 и попадают в емкость колпачка, на дно которого вскоре оседают. Колпачок экспонируют в течение заданного времени в термостате. Для доказательства целесообразности использования колпачка были проведены экспериментальные исследования. В июне-августе 2017 г. в частном крестьянско-фермерском хозяйстве Выборгского района Ленинградской области (60.735175°N, 29.004207°Е) от овец романовской породы в возрасте от 2 до 7 лет ректально были получены фекалии. В лаборатории на базе кафедры паразитологии им. В.Л. Якимова ФГБОУ ВО «Санкт-Петербургская государственная академия ветеринарной медицины» было проведено предварительное копроовоскопическое исследование по модифицированному методу С. Дарлинга, которое показало наличие яиц нематод в исследуемых фекалиях. Затем было проведено культивирование личинок из яиц по методу А.М. Петрова и В.Г. Гагарина, для чего в чашки Петри (2 серии по 45 шт.) помещали 10-граммовую навеску фекалий, чашки закрывали и помещали в термостат для культивирования в течение 10 дней при температуре 27°С. Ежедневно чашки Петри извлекали из термостата для проветривания и увлажнения фекалий. После завершения периода культивирования (когда личинки нематод успевали выйти из яиц и претерпеть две линьки, достигнув инвазионной стадии развития, оптимальной для проведения исследования) в фекалии в чашках Петри первой серии установили устройства для сбора личинок и мелких нематод из фекалий (а.с. №1391625), а в чашках Петри второй серии - колпачки. Чаши устройств и емкости колпачков до краев заполнили чистой водой и оставили экспонировать. Через 24 часа собранных личинок из каждой пробы (фиг. 3 и 4) отдельно выбрали пипеткой, а чаши устройств и емкости колпачков вновь наполнили водой и продолжили экспонирование. Жизнеспособность личинок определяли по их подвижности и сохранности морфологических структур под микроскопом Микмед-6 при 40-кратном увеличении (фиг. 5). Затем их зафиксировали 70°C спиртом и подсчитали количество личинок. Совокупный срок экспонирования составил 72 часа. По завершении экспонирования полученных личинок происследовали на жизнеспособность, а после - зафиксировали спиртом и подсчитали их количество. Результаты исследований представлены в таблице 1.To prove the feasibility of using the cap, experimental studies were conducted. After completion of the cultivation period (when the larvae of the nematodes managed to leave the eggs and undergo two molts, reaching the invasive stage of development optimal for the study), feces in the first series Petri dishes were equipped with devices for collecting larvae and small nematodes from feces (A.S. 1391625), and in the Petri dishes of the second series - caps. The cups of the devices and the capacity of the caps to the brim were filled with clean water and left to expose. After 24 hours, the collected larvae from each sample (Figs. 3 and 4) were individually selected with a pipette, and the devices and cap containers were again filled with water and continued exposure. The viability of the larvae was determined by their mobility and the preservation of morphological structures under a Mikmed-6 microscope at a 40-fold increase (Fig. 5). Then they were fixed with 70 ° C alcohol and the number of larvae was counted. The total exposure time was 72 hours. Upon completion of exposure, the obtained larvae were examined for viability, and after that they were fixed with alcohol and their number was calculated. The research results are presented in table 1.
Применять колпачок для сбора личинок и мелких нематод из фекалий можно в целях: 1) качественной и количественной диагностики нематодозов человека и животных; 2) определения антигельминтной резистентности у паразитических нематод и скрининга препаратов против них in vitro; 3) получения образцов антигенов нематод с целью изготовления серологических диагностикумов и вакцин; 4) изучения биологии нематод и разработки на ее основе более совершенных мер борьбы; 5) секвенирования ДНК нематод в интересах эволюционной биологии (т.к. обнаружение сходных гельминтов у животных разных видов (ныне существующих или вымерших) может свидетельствовать о филогенетическом родстве или общем ареале обитания).
