Abstract
Description
This invention relates to veterinary medicine and can be used in the treatment of acute and chronic otitis media of parasitic, bacterial and fungal origin in dogs, cats, fur animals and rabbits.
In the market of veterinary drugs, ear drops (solutions for auricular use) for the treatment of otitis media in animals often contain several active ingredients: broad-spectrum antibiotics, acaricides, anti-inflammatory components. These compositions do not provide a complete relief of the pathological focus.
[2, 3].An integrated approach to therapy is necessary for the successful treatment of diseases with different etiologies. Finished dosage forms should contain, if possible, components that provide a comprehensive effect on the pathological focus while minimizing the number of injections of various drugs and the cost of working time of veterinary specialists for additional treatments [2, 3].
The causes of otitis media in animals are mainly reduced to three main agents: parasitic, bacterial and fungal. Parasitization of ear mites often leads to an excessive growth of opportunistic microflora and fungi of the genus Candida and Malassesia. Therefore, the use of ear drops with a single-component composition: acaricide, or antibiotic, or antimycotics is inappropriate and sets the goal for veterinarians to create a combined drug of polytropic action.
Ear drops are known for the treatment of otitis media in animals. Veterinary medicine Aurican (Ceva Sante Animale, France) contains diazinon, sodium prednisolone sulfobenzoate, hexamidine diisetionate and tetracycline hydrochloride as active ingredients. This composition does not provide fungicidal action.
The drug “Drops of ears Bars forte” (LLC NEC Agrovetzashchita, Moscow) contains miconazole nitrate and enrofloxacin, these components provide only antibacterial and antifungal effects.
The prototype of this invention is a drug for veterinary use "Mometomax" ("Schering-Plow Canada Inc.", Canada), containing gentamicin, mometasone and clotrimazole as active substances [1]. However, this tool is acceptable only for otitis media of bacterial and fungal origin. The disadvantage of this drug is the lack of an active substance against an ear tick, as well as the limitation of the list of target animal species for which it is possible to use the drug.
The aim of the present invention is to develop a highly effective comprehensive preparation for the treatment of acute and chronic otitis media of parasitic, bacterial and fungal origin in dogs, cats, fur animals and rabbits. This goal is achieved by the fact that in a drug that is intended for the treatment of acute and chronic otitis media of parasitic, bacterial and fungal origin, a combination of active substances from levofloxacin, clotrimazole, dexamethasone and moxidectin and targeted additives are used.
В качестве целевых добавок препарат содержит растворители (например, полиэтиленгликоль (ПЭГ-400) и эмульгатор (например, твин-80).As target additives, the preparation contains solvents (for example, polyethylene glycol (PEG-400) and an emulsifier (for example, tween-80).
This complex preparation contains clotrimazole, which has a pronounced fungicidal effect. The mechanism of its action is to reduce the synthesis of ergosterol, which is an integral part of the cell membrane of the wall of microscopic fungi, which leads to a change in its structure, properties and further death.
A broad-spectrum antibiotic - levofloxacin has a detrimental effect on a wide range of pathogenic and conditionally pathogenic microorganisms that are the causative agents of infectious and inflammatory processes. Levofloxacin is an antibiotic with a bactericidal type of action (it blocks the work of enzymes that are necessary for the synthesis of DNA of microorganisms, without which they are not capable of reproduction).
Dexamethasone is a synthetic glucocorticoid primarily used as an anti-inflammatory and immunosuppressive agent. When applied topically, the therapeutic activity of dexamethasone is due to anti-inflammatory, anti-allergic and antiproliferative effects. It reduces the permeability and proliferation of capillaries, local exudation, cell infiltration, phagocytic activity, inhibits the formation of scar tissue. All these effects are involved in suppressing the inflammatory response in tissues in response to mechanical, chemical, or immune damage.
[4].Moxidectin is a semi-synthetic compound from the milbemycin group. Providing a stimulating effect on the release of gamma-aminobutyric acid and binding to postsynaptic receptors, it causes a violation of muscle innervation, paralysis and death of ectoparasites; high efficacy of moxidectin in carnivorous acaroses was noted [4].
A comparable analysis of the proposed technical solution with the known prototype shows that the claimed tool differs from the prototype in its multicomponent composition, which stops all agents that cause otitis media in animals, as well as an expanded list of target animal species.
The invention is illustrated by the following examples.
Example 1. The drug is prepared as follows. A PEG-400 sample is loaded into the reactor; tween-80 is mixed until completely dissolved; then load samples of levofloxacin hemihydrate, clotrimazole, dexamethasone sodium phosphate and moxidectin; mix until completely dissolved.
%):The drug in the form of a solution for auricular use (in wt.%):
Example 2. For otitis media caused by ear mites, the external auditory meatus is cleaned of scabs and crusts, then 2-5 drops of the drug are instilled into each ear (for cats, small dogs, ferrets and rabbits - 2-3 drops, for medium dogs, foxes - 4 drops, large dogs - 5 drops). The auricle is folded in half and massaged at its base. If the animal shakes its head after using the drug, fix the head for several minutes to prevent the drug from splashing. The treatment is repeated 2 times with an interval of 7 days. If necessary, repeat the treatment. The drug must be administered in both ears, even if clinical signs of the disease are detected in only one ear.
The studies were carried out in veterinary clinics in Moscow and the Moscow region. The first group included 3 dogs (2 of them puppies), 16 cats and 6 ferrets, who were diagnosed with otodectosis. The second group consisted of 11 rabbits with a diagnosis of psoroptosis. After using the drug in animals from group No. 1, the general condition improved, hyperemia and swelling of the skin of the inner surface of the auricles disappeared, the auricle elasticity was restored, they became almost clean, Otodectes cynotis ticks were not found after the course of treatment, only dead ticks and parts of them were recorded . After double treatment, live ticks were also not found. Thus, 100% effectiveness of ear drops is confirmed by two acarological studies. After treatment, hyperemia and swelling of the skin of the inner surface of the auricles disappeared in rabbits, they became almost clean, Psoroptes cuniculi ticks were not found after double treatment of animals.
Example 3. In the case of fungal and / or bacterial otitis media, it is necessary to carry out hygienic treatment of the auricle and external auditory meatus before using the drug. The drug is instilled into the ear canal 2-5 drops of the drug (depending on the size of the animals), then gently massage the base of the ear. The drug is used 1 time per day for 7 days. In case of incomplete disappearance of symptoms after one course, treatment is continued for another 7 days.
Studies were carried out in veterinary clinics in Moscow and the Moscow region. The diagnosis of otitis media of bacterial and / or fungal etiology was made to 20 dogs (of which 1 puppy), 16 cats (of which 2 kittens) and 2 ferrets. After using the drug after 14 days: the ears became cleaner, the itching and inflammation almost stopped. After the drug was discontinued, 2 dogs had a relapse in the chronic process, and therefore treatment was continued for another 7 days, after which clinical recovery was observed.
The work was carried out in the conditions of JSC breeding animal farm "Saltykovsky". They treated foxes with a diagnosis of otodectosis; revealed 100% effectiveness of the drug based on two acarological studies. Also, based on histological examination data, positive morphological changes in the skin of the inner surface of the auricle were revealed (Figure 1-3).
Восстановление оптимальной микроструктурной организации тканей кожи наблюдается после применения препарата (Рисунок 3), в частности уменьшение общей толщины кожи в 2,2 раза по сравнению со значениями, представленными в группе больных животных (Рисунок 2).The restoration of the optimal microstructural organization of skin tissue is observed after the use of the drug (Figure 3), in particular, a decrease in the total skin thickness by 2.2 times compared with the values presented in the group of sick animals (Figure 2).
