Известен способ определения высокомолекулярных некрахмалистых полисахаридов в содержащем их сырье [Патент SU №1455297, опубл. 30.09.89]. Навеску исследуемого материала подвергают трехминутному гидролизу смесью 60%-ного раствора серной кислоты и этилацетата в соотношении 9:1. В полученном гидролизате определяют высокомолекулярные некрахмалистые полисахариды (способные гидролизоваться до глюкозы) фотоколориметрическим методом с использованием смеси серной кислоты, антрона и этилацетата в соотношении 20:0,01:1 соответственно. Экспериментальным путем обнаружено, что для обеспечения указанного технического результата необходимо соблюдать предлагаемые режимные параметры пробоподготовки и хроматографического разделения углеводов, содержащихся в полупродуктах спиртового производства. При реализации предлагаемых режимных параметров уменьшается время, затрачиваемое на анализ, повышается чувствительность метода, а также упрощается способ, отсутствуют потери при пробоподготовке, отсутствуют потери за счет испарения вещества при проведении анализа, прослеживается динамика изменения массовой концентрации углеводов в технологическом процессе производства спирта. Микрошприцем отбирали пробу надосадочной жидкости, разбавляли дистиллированной водой и проводили количественное определение мальтозы, глюкозы и фруктозы в подготовленных пробах хроматографическим методом с использованием жидкостного хроматографа с рефрактометрическим детектором на хроматографической аналитической колонке RHM-Monosaccharide Phenomenex НГ, заполненной сорбентом, смолой с 8% со степенью сшивки, в водородной ионной форме (300×7,8 мм, размер частиц 8 мкм), скорость потока подвижной фазы (дистиллированная вода) 0,6 см3/мин. Давление 2,3 МПа, температура в колонке 80°С. По полученной хроматограмме было установлено, что сусло содержит глюкозу в количестве 7,7 г/100 см3, мальтозу, 7,1 г/100 см3 и фруктозу, 0,96 г/100 см3.Example 1. Давление 2,3 МПа, температура в колонке 80°С. По полученной хроматограмме было установлено содержание мальтозы, глюкозы, фруктозы. Динамика изменения массовой концентрации углеводов в технологическом процессе производства спирта представлена в таблице 2.Example 2.
