Бактерицидная активность фракций супернатантов клеток оценивалась по общепринятой методике «Определение чувствительности микробов к антибиотикам методом диффузии в агар с применением бумажных дисков». В стерильные чашки Петри заливали 20 мл 2% стандартного мясо-пептонного агара. Посев тестовой культуры микроорганизмов производили общепринятым способом. На подсушенную поверхность среды засевали сплошным газоном госпитальный штамм St.aureus 155, Е.Coli 2290 и Canda albicans с использованием стерильного ватного тампона, смоченного в 2-миллиардной взвеси микробной культуры. На поверхность засеянной среды раскладывали диски, пропитанные исследуемыми субстанциями путем погружения дисков в состав до полного пропитывания.The bactericidal activity of fractions of cell supernatants was evaluated according to the generally accepted methodology "Determination of the sensitivity of microbes to antibiotics by diffusion into agar using paper discs." Установлено, что выраженной бактерицидной активностью в отношении St.Aureus 155 обладают коммерческие бактерицидные эндогенные пептиды (БЭП) LL37 (прототип) и HNP (прототип), растворенные в буферном растворе в дозе 10 мг/мл (20,1±1,9 и 22,2±2,15 мм диаметр зоны подавления роста микроорганизма соответственно). У них же в той же дозе проявляется самая высокая бактерицидная активность в отношении Е.Coli 2290 (23,2±1,7 и 24,7±2,15 мм диаметр зоны подавления роста микроорганизма соответственно). Подобной же активностью обладает полученный нами пептид ZP-2, состоящий из 12 аминокислот. У него бактерицидная активность в отношении St.Aureus 155 и Е.Coli 2290 достоверно не отличалась от известных коммерческих БЭП и составила соответственно 22,3±2,16 и 19,6±1,6 мм диаметр зоны подавления роста микроорганизмов. Отличие было лишь в том, что подобная бактерицидная активность проявилась не только в буферном растворе, но и при растворении в физиологическом растворе. The only difference was that a similar bactericidal activity was manifested not only in the buffer solution, but also when dissolved in physiological saline. When dissolving bactericidal endogenous peptides (BEP) LL37 and HNP in physiological saline, the latter do not exhibit bactericidal activity, which is also seen from table 1.
