Abstract
Description
The invention relates to medicine, specifically to pharmaceutical chemistry. A new stable X-ray amorphous form of ranitidine hydrochloride having the properties of an H 2 blocker is described. The new form is physically stable and has a more pronounced antiulcer effect compared to previously known crystalline forms.
Ranitidine: N- [2 - [[[5- (dimethylamino) methyl] -2-furanyl] methyl] thio] ethyl-N'-methyl-2-nitro-1,1-etendiamine) and its physiologically acceptable salts are first described U.S. Patent No. 4,128,658. Ranitidine found the ability to block H 2 histamine activity, which led to the prospect of its use in the treatment of diseases accompanied by disorders of the functional state of the gastrointestinal tract: gastric ulcer, peptic ulcer, postoperative complications, as well as allergic and inflammatory conditions in which histamine is known to participate as a pick.
The salt formed by ranitidine base and hydrochloric acid (hereinafter referred to as ranitidine hydrochloride) is the most important, as it is used in most manufactured dosage forms, for example, tablets for oral administration. This is the reason for the search for ways to produce ranitidine hydrochloride in such a physical condition that would provide advantages not only during the manufacturing process, but also during storage of the final product, as well as when using the finished dosage form for patients.
Например, заявка РСТ WO 96/33185, патенты США №5523423, №5338871 и №5621120.So, Ranitidine hydrochloride Form 2 was described and patented - US Pat. Nos. 4,521,431 and 4,672,133. It is indicated that in accordance with the method proposed to obtain the initial form 1 of ranitidine hydrochloride (US patent No. 4128658), in the industrial production process of the product, there are drawbacks - the product has unsuitable characteristics for filtering and drying. The above sources indicate the impossibility of producing crystalline form 1 of ranitidine hydrochloride on a commercial scale and, moreover, form 1 is unstable and spontaneously passes into stable form 2, and very quickly. Unlike Form 1, Form 2 has larger crystals, is less hygroscopic, and exhibits other technological advantages. However, repeated attempts have been made to create Form 1, which would be devoid of these shortcomings. For example, PCT application WO 96/33185, US patents No. 5523423, No. 5338871 and No. 5621120.
Initial US Patent No. 4,128,658 describes various aminoalkylfuran derivatives, including ranitidine. Example 32 of the description of US patent No. 4,128,658 sets out a method for preparing Form 1 of ranitidine hydrochloride by dissolving ranitidine in industrial alcohol (the solvent, the bulk of which is ethanol) containing hydrogen chloride. Crystallization is carried out by adding ethyl acetate. This procedure is unsatisfactory in its characteristics - from the above solvents (ethyl acetate and ethanol), ranitidine hydrochloride form 1 is unstable. This makes the process inapplicable for use on a commercial scale.
US Pat. Nos. 4,521,431 and 4,672,133 describe a crystalline form of ranitidine hydrochloride, designated as Form 2, and having more favorable characteristics for filtration and drying than Form 1 of ranitidine hydrochloride obtained using hydrogen chloride, industrial alcohol and ethyl acetate. Ranitidine hydrochloride Form 2 can be prepared by treating a ranitidine solution with, for example, lower alcohol and hydrogen chloride, followed by crystallization at elevated temperature and with the addition of an additional amount of solvent. In addition, ranitidine hydrochloride form 2 can be obtained by preliminarily isolating form 1 and dissolving the salt, for example, heating an organic solvent such as methanol, ethanol, followed by cooling and crystallization of the salt of form 2, it is possible to add an anti-solvent or add germ crystals of form 2 to induce crystallization.
The differences between Form 2 described in US Pat. Nos. 4,521,431 and No. 4,672,133 and Form 1, the product obtained in Example 32 of the description of US Pat. No. 4,128,658, are characterized by an IR spectrum and powder x-ray characteristics.
US Pat. No. 5,338,871 describes the preparation of pure form 1 of ranitidine hydrochloride by dissolving ranitidine hydrochloride in a solvent mixture containing at least one lower aliphatic alcohol, an aromatic hydrocarbon and crystallization initiator in the form of germinal crystals of pure form 1 of ranitidine hydrochloride.
In US patent No. 5621120 describes three modifications that differ in the characteristics of powder x-rays, form 1, form 2 and a standard sample corresponding to the US Pharmacopoeia.
Как следует из вышеуказанных патентов, температура плавления нестабильной формы 1 ранитидина гидрохлорида составляет 133-134°С (патент США №4128658), стабильной формы 1 ранитидина гидрохлорида - 136-137°С (патент США №5621120), а формы 2 ранитидина гидрохлорида - 139-141°С (патент США №4521431).As follows from the above patents, the melting point of the unstable form 1 of ranitidine hydrochloride is 133-134 ° C (US patent No. 4,128,658), the stable form of 1 ranitidine hydrochloride is 136-137 ° C (US patent No. 5621120), and form 2 of ranitidine hydrochloride is 139-141 ° C (US patent No. 4521431).
No therapeutic equivalence of these polymorphic modifications has been reported. On the contrary, in the description of US patent No. 5621120, it is reported that form 1 and form 2 exhibit practically indistinguishable bioavailability.
It is known that the solubility of a drug substance is one of the factors that determine its therapeutic efficacy. It is known that therapeutic efficacy depends on the bioavailability of the drug, which is a function of good and / or complete absorption. Absorption depends on the solubility of ranitidine hydrochloride. A good dissolution of ranitidine hydrochloride is all the more desirable because there is a relatively small area of the gastrointestinal tract where it is absorbed. Weak or incomplete dissolution at the moment of contact with the absorption zone causes a weak absorption and, thus, the therapeutic effect can be significantly reduced.
For a long time, they tried to prepare a dosage form containing ranitidine in amorphous form, since it is known that in amorphous forms, the solubility is usually higher than that in crystalline forms (see review by J. Haleblain, J. Phann. Sci, 64, 1269, 1975). However, the creation and use of amorphous forms is a complex problem, mainly because they are physically unstable, thermodynamically unstable and can transform into crystalline forms within a short period of time. The vast majority of substances are difficult to obtain in an amorphous state (Kitaigorodsky A.I. X-ray diffraction analysis of fine crystalline and amorphous bodies. M., 1952). PCT application WO 96/39126 indicates that the detection of a physically stable amorphous form of ranitidine that does not rapidly convert to crystalline form would be a significant step forward. In this application, to solve the problem, an adsorption complex of ranitidine hydrochloride on a carrier was obtained. The problem of obtaining a pure amorphous form has not been solved. A similar decision was taken in PCT application WO 94/20091. This application describes an inclusion complex of ranitidine hydrochloride and cyclodextrin. In these applications, the X-ray diffraction characteristics of the complexes and DSC spectra are given.
Naturally, the radiographic, thermal, and other characteristics of ranitidine hydrochloride adsorbed as a monolayer on a carrier or enclosed within a cyclodextrin molecule are significantly different from those inherent in its amorphous or crystalline forms. In addition, molecular and conformational changes caused by the influence of adsorption or other stabilizing complexes of forces do not guarantee the fact that ranitidine hydrochloride is present in the complex in the conformation characteristic of an individual amorphous form.
In addition, the introduction of an additional component in the drug is an undesirable fact. For example, upon receipt of the ranitidine-carrier adsorption complex (according to the aforementioned PCT application WO 96/39126), there are significant disadvantages: significant losses of the active component, high demands on the specific surface of the carrier, difficulties in the subsequent standardization of the complex substance, and some others. It should also be noted that the use of a large amount of carrier can adversely affect the condition of the gastrointestinal tract of a patient using the final product, an anti-ulcer agent.
Thus, we can conclude that at present the problem of obtaining stable ranitidine hydrochloride, which has therapeutic advantages compared with the known polymorphic modifications, is not solved.
The objective of the invention is the creation of a physically stable form of ranitidine hydrochloride with increased therapeutic activity. As a result of the experiments, it was unexpectedly discovered that the X-ray amorphous form of ranitidine hydrochloride possesses increased specific activity along with stability over a long period of time sufficient for commercial use. The X-ray amorphous form of ranitidine hydrochloride is a very light, “airy” white amorphous powder with a slight yellowish tint, characterized by diffuse scattering (amorphous halo) on a powder X-ray diffraction pattern in the region of 15-30 2θ ° and one wide endothermic effect on the DSC curve (differential scanning calorimetry) in the range from 107.1 ± 2.5 to 146.2 ± 2.0 ° C. Powder is faster and larger than crystalline forms, it dissolves in water.
The invention is also a method of obtaining a new x-ray amorphous form. The method consists in the fact that crystalline ranitidine hydrochloride is dissolved in an organic solvent or in water, or in a mixture of them at a temperature of 50-80 ° C, the solution is cooled to freeze, the frozen mass is ground and dried to obtain an X-ray powder of ranitidine hydrochloride having a residual moisture content of more than 2.5%. Drying the frozen mass is carried out under reduced pressure. As the organic solvent, it is preferable to use lower aliphatic alcohols, in particular ethyl alcohol.
The new X-ray amorphous form of ranitidine hydrochloride, in comparison with the previously known polymorphic forms and various complexes of this compound, has an increased specific antiulcer activity.
The obtained X-ray amorphous form of ranitidine hydrochloride can be widely used in medicine, especially for the treatment of gastric ulcer.

Industrial applicability
USP XXIII).The antiulcer activity of the claimed X-ray amorphous form of ranitidine hydrochloride was studied in comparison with its well-known modification (widely used in the treatment of peptic ulcer disease of ranitidine hydrochloride according to USP XXIII).
J. GastroenteroL-1981. Vol.16, Suppl. №69 - p. 1-8). The antiulcer activity of the studied samples was determined on a model of aspirin gastric ulcer in male Wistar rats weighing 258-337 g (Brittain RT, Daly MJ, Scand. J. GastroenteroL-1981. Vol.16, Suppl. No. 69 - p. 1 -8). The animals were kept in individual standard cages with an area of 806 cm 2 with trellised floors made of stainless wire. 18 hours before the experiment, the animals were deprived of food with free access to drinking water.
15 minutes before the induction of the ulcer, the animals were intragastrically injected with a solution of the claimed preparation in physiological saline at a concentration of 0.1 or 0.2 mg / ml based on 5 ml / kg of body weight of the animal. The animals of the control group were injected with saline. Ulcerative lesions of the gastric mucosa were caused by intragastric administration of a 6% suspension of acetylsalicylic acid in 1% starch at the rate of 5 ml / kg. Slaughter of animals was carried out 5 hours after the introduction of a suspension of acetylsalicylic acid by overdose of medical ether. In isolated stomachs with ligatures superimposed on the esophagus and pylorus, 8 ml of a 2% formalin solution was injected through a 10-mm syringe, after which the preparations were placed for 10 min in a 2% formalin solution to fix the wall of the stomach, the stomachs were dissected according to great curvature. The total length of ulcerative lesions of the gastric mucosa in each of the rats was measured at 8-fold magnification using a biological microscope equipped with a measuring ruler integrated into the eyepiece with a division price of 0.1 mm. The ulcerative index was calculated as the sum of the length of ulcerative lesions of the rat gastric mucosa.
The results of a study of the effect of preliminary intragastric administration of the claimed X-ray amorphous form of ranitidine hydrochloride in comparison with the known ranitidine hydrochloride, widely used as an anti-ulcer agent, are shown in the table.
As can be seen from the table, a significant decrease in the ulcer index of the claimed drug compared with the well-known pharmacopoeial ranitidine hydrochloride at a dose of 0.5 mg / kg is 1.2 times, and at a dose of 1 mg / kg - 1.5 times.
