Abstract
Description
The invention relates to medicine, namely to laboratory diagnostics, and is used for the semi-quantitative determination of phosphate ion in daily urine.
20.08. 2002).A known method for the determination of phosphate in biological materials, including taking a sample, its mineralization by treatment with oxidizing agents based on sulfuric acid during boiling, followed by the introduction of solutions of nitric acid, vanadate and ammonium molybdate with the formation of a colored solution and its photometry, with nitric acid being introduced in two doses of 1 / 2 of the total number (Application No. 2000118580/13 of the Russian Federation. Publ. 20.08. 2002).
However, this method is time-consuming and requires preliminary preparation of the material, conducting a standard test, using spectrophotometric equipment.
The present invention is the development of a technically simple, affordable way to determine the concentration of phosphate ion in the urine.
The problem is solved in that in the method for the semiquantitative determination of phosphate in daily urine, including taking a sample of the test material and analyzing the phosphate content in it, phosphate is determined by the precipitation of the ion with a solution of iron (III) chloride, then the results of the sample are evaluated, and visualization of the sediment indicates about the phosphate content in the urine is within normal limits, and the absence of sediment indicates the phosphate content in the urine is below normal.
The method is as follows.
Patients collect daily urine. 0.3 ml of urine is taken into a test tube, diluted by adding 0.9 ml of distilled water to the indicated volume. Then, 0.02 ml of a 0.4 N solution of iron (III) chloride is added to 1.2 ml of diluted urine. If the solution in the test tube becomes cloudy as a result of the addition of ferric chloride, then it is concluded that the phosphate content in the daily urine is within normal limits. If the solution in the test tube does not become turbid when iron chloride is added, then it is concluded that the phosphate content in daily urine is below normal.
Clinical example.
The phosphate content, measured by reaction with ammonium molybdate, in the daily urine of patients G. and N. was 28.92 and 6.55 mmol / day, respectively. When adding 0.02 ml of a 0.4 N solution of iron (III) chloride to 1.2 ml of diluted urine of a patient G. turbidity was observed, while adding a solution of iron (III) chloride to 1.2 ml of diluted urine of patient N. Transparency the solution has not changed. Based on the data obtained, it was concluded that the phosphate content in the urine of patient G. is within normal limits, while in patient I. it is reduced.
The proposed method allows for the semi-quantitative determination of phosphate in daily urine. This method is simple to execute, does not require equipment, does not require preliminary sample preparation, only one substance is used as a reagent. This makes it possible to use the method in the field for rapid diagnosis of conditions associated with a decrease in phosphate excretion.
