Abstract
Description
The invention relates to medicine, namely to oncology, and can be used to treat tumor diseases.
И. 1990).To date, there are many ways to treat tumors. Known surgical methods for treating patients of this group. The method is widely and successfully used with limited tumor lesions (corresponding to stages I-II). Surgical intervention is considered radical, which involves the removal of a tumor as a single unit with healthy tissues, regional lymph nodes and, if necessary, with part of neighboring organs (S. L. Daryalova, V. I. Chissov “Diagnosis and treatment of malignant tumors”, 1993). However, the frequency of relapses and metastases, especially with locally advanced forms of cancer, does not allow us to talk about the achievement in some cases of complete cure due to unrecognized subclinical metastases and the impossibility of an adequate organ resection due to the large tumor spread. According to some reports, cancer recurrence after radical surgery was 25-30%, and metastasis in 10-15% of patients (Knysh V.I. 1990).
Х. А., 1988).Also known is the radiation treatment method, which consists in the use of ionizing radiation, in particular gamma rays, with the inclusion of skin tumors and adjacent healthy tissues in the volume of exposure. However, gamma therapy as an independent type of treatment has a number of broad contraindications, and summing up the maximum doses of 60-70 Gy in most cases not only does not lead to complete damage to the tumor, but also damages healthy tissues, due to their low tolerance to ionizing radiation (Kozlova A.V. 1971; Sargsyan Yu.Kh. et al. 1982, Chushkin N.A., 1988).
1993).Chemotherapy also refers to known methods of treating malignant tumors. For chemotherapy as an independent type of treatment, one (monochemotherapy) or several drugs (polychemotherapy) is used. Known and widely used chemotherapy drugs have low antitumor activity, a large number of complications and reactions (vomiting, leukopenia, anemia, thrombocytopenia, immune suppression, etc.). Increasing the effectiveness of chemotherapy for tumors goes in different directions. The main ones are the creation of new drugs that are more selective for tumor cells for monochemotherapy, the development of the combined use of two or more drugs (polychemotherapy), the optimization of drug administration methods (dosing, the regimen of use), the improvement of chemotherapy as a link or stage of a comprehensive and combined treatment of tumors, the use of other drugs to reduce the side effects of anticancer drugs, etc. Much attention is paid to the mechanism effects of drugs and their pharmacokinetics (the study of routes of distribution, drug metabolism and excretion) and pharmacodynamics (the study of the effects caused by medication). Based on the data obtained, rational methods for using well-known drugs are developed and ways to create new effective antitumor agents are determined (N.I. Perevodchikova “Antitumor Chemotherapy”, M., Medicine, 1993).
The objective of the invention is the search for new drugs with pronounced antitumor activity, especially against malignant tumors, and low toxicity.
The problem is solved by the use of chlorocholine chloride as an antitumor agent.
«Effect of various xenobiotics on the in vivo incorporation of 14C-acetate into cholesterol and other lipids in the rat», Acta Biol Med Ger, 1981, 40 (7-8), р.1043-52). Chlorocholin chloride (chloromequate, TUR preparation) of the formula ClCH 2 CH 2 N + (CH 3 ) 3 Cl - is widely used in agriculture (Zadontsev A.I., Pikush G.R., Grinchenko A.A. "Chlorocholin chloride in crop production", M., Kolos, 1973, 360 pp.). The drug is used to protect potatoes from viral diseases (RU 2075289, RU 2024532), as well as to regulate the growth of various plant objects (RU 1343772, RU 2050136, RU 2082296). In laboratory conditions, the effect of the claimed compound on the synthesis, transport and accumulation of phospholipids, triglycerides and cholesterol in rats was studied (Seidler H, Hartig M, Koslowski C, Ackermann N. “Effect of various xenobiotics on the in vivo incorporation of 14C-acetate into cholesterol and other lipids in the rat, Acta Biol Med Ger, 1981, 40 (7-8), p. 1043-52). However, none of the above sources of information describes the use of chlorocholine chloride as an antitumor agent.
The drug chlorcholine chloride refers to moderately toxic compounds. Acute toxicity parameters (LD 50 ) are 54 mg / kg.
The proposed properties of chlorocholine chloride are illustrated, but not limited to the following examples.
1.Example 1
Rats were infected with sarcoma-45 (a total of 20 rats). Subsequently, all animals were divided into two groups: control, in which no treatment was carried out, and experimental, in which 5 drops of a 1% solution of chlorocholine chloride were administered twice a day for 1 month. All rats in the control group (10 animals) died within three weeks. Among the rats of the experimental group, 3 rats died in the first three weeks, the remaining 7 only after two months.
2.Example 2
Patient Sh., 72 years old. Diagnosis: primary liver cancer, untreated. The patient refused chemotherapy. Objectively: weight loss by 20 kg, weakness, anemia, pain in the right hypochondrium. On ultrasound, a 4 × 3 cm volume formation is visualized in the left lobe of the liver.
Conclusion: Primary liver cancer. Diagnostic puncture confirms the diagnosis.
Treatment with chlorocholinchloride has begun according to the scheme: 15 drops of a 1% solution of the drug 3 times a day for 3 weeks. After 2 weeks, the patient's condition improved significantly; Appetite appeared, pain disappeared, became more active. After 2 months, a control ultrasound of the liver was performed. At the site of the tumor, a formation of 0.1 × 0.2 cm remained, and after a month nothing was found.
Currently, the patient recovered by 25 kg, his blood counts are normal, and his general condition is good.
Thus, chlorocholine chloride is an effective drug for the treatment of various tumors, including malignant neoplasms. The drug is moderately toxic and does not have the side effects characteristic of many other antitumor agents.
