The invention relates to medicine, namely to a preparation from the group of fluorinated quinolones, namely toloxacin for injection.
345-346).Ofloxacin 9-fluoro-2,3-dihydro-3-methyl-10- (4-methyl-1-piperazinyl) -7-oxo-7H-pyrido / 1,2,3-de / 1,4-benzoxazine carboxylic acid a wide range of antibacterial action. Practically insoluble in water and alcohol. Available only in the form of tablets (M.D. Mashkovsky, Medicines, vol. 2, pp. 345-346).
РФ №2145497).Known dosage form ofloxacin in the form of tablets (US Pat. US No. 4639458) and in the form of ointments (US Pat. RF No. 2145497).
So in patent No. 2145497, an agent is described for treating purulent-necrotic processes in the form of an ointment containing an antibacterial (ofloxacin) and a local anesthetic (trimecain or lidocaine) agent, a gelling agent (proxanol), protease-C, polyethylene oxide-400 and water. The product is intended for external use only. In this method of producing ointment, ofloxacin was able to dissolve in water when heated only due to the presence of trimecaine.
The prototype of this invention is a composition containing a therapeutically permitted amount of ofloxacin, and auxiliary additives (US Pat. RF No. 2207115). The known composition is made in the form of coated tablets. As auxiliary substances it contains lactose, starch, hydroxypropyl cellulose, crospovidone, magnesium stearate. This composition is inherent in all the disadvantages of the tablet form of the drugs: lack of therapeutic availability, low tablet strength.
The objective of the invention is the creation of ofloxacin for injection, convenient to use, providing high therapeutic efficacy, the absence of crystallization of the active substance for a long time.
Массовое соотношение ингредиентов, масс.%:This goal is achieved through the use of a composition containing a therapeutically permitted amount of ofloxacin, and auxiliary additives, which use trilon-B, sodium chloride, water for injection. The composition is obtained in the form of a solution for injection. Mass ratio of ingredients, wt.%:
Трилон-Б 0,19-0,22Trilon-B 0.19-0.22
8,5-9,5Sodium chloride 8.5-9.5
100%Water for injection ost. up to 100%
In the course of the study, it was found that the combination of ofloxacide with trilon-B and sodium chloride in certain ratios prevents the crystallization of the active compound and increases the solubility of ofloxacin in water.
As a result of the research work, it was found that Trilon-B, a disodium salt of ethylenediaminetetraacetic acid, is able to form complex compounds with fluorine-containing compounds, namely ofloxacin, translating it into a water-soluble state.
The invention is illustrated by the following examples.
Example 1. In a capacitive heated apparatus equipped with a stirrer, a sodium chloride solution is preliminarily prepared to obtain a 0.85-0.95% isotonic solution, Trilon-B is added at the rate of obtaining a 0.0190-0.022% solution. Ofloxacin 1.9-2.1 g / l is added at a temperature of 50-60 ° C for 60 minutes. To the resulting solution add buffer (NaOH solution) pH 6.0-7.3 with the calculated amount of water. Membrane filtration is carried out using a plate holder company "Millipor". A membrane with a pore diameter of 0.2 μm “Vladipor” of the PMAC type is placed in the filter holder. For sterilizing filtration, a 0.2 μm Sartobran P hydrophilic membrane from Sartrius is used. Bottling is carried out in accordance with GOST 10782-85 with a dose of 100 ml. Capping is done with stoppers from the rubber mixture 52-369 / 1, crimped with aluminum caps. After bottling, the finished product is sterilized in an autoclave at a temperature of 120 ° C for 15 minutes.
