Abstract
Description
The present invention relates to medicine, in particular to drugs based on doxazosin, which has antihypertensive, vasodilating, antiatherosclerotic effects, is effective in the treatment of prostate adenoma in the early stage.
The substance of doxazosin - 1- (4-amino-6,7, -dimethoxy-2-quinazolinyl) -4 - / (2,3-dihydro-1,4-benzodioxin-2-yl) carbonyl / piperazine is permitted in the country and is registered in the Ministry of Health of the Russian Federation in the form of monomethanesulfonate (mesylate). US Pat. No. 4,188,390 describes doxazosin as a biologically active compound for use in drugs.
A solid cylindrical dosage form is known, with controlled release of a substance, for example, doxazosin, at almost constant speed, including, in addition to doxazosin, a carrier of sodium benzoate, ethyl cellulose, lactose, magnesium stearate and a shell of an ethylene vinyl acetate copolymer (US Pat. RF 2072835).
A product is also known for prolonged release of a pharmaceutically active substance in the form of a tablet, consisting of a core containing doxazosin and an excipient, and surrounded by a shell of a polymer film representing an asymmetric membrane formed by a phase-inversion method from polymers selected from the group consisting of cellulose ethers and esters , polyvinyl alcohol, ethylene vinyl alcohol copolymer, polyvinylidene fluoride, polyamide, polyurethane, polymethyl methacrylate and polysulfone. Adipic acid and lactose were used as filler in this form (US Pat. RF 2093147).
The main disadvantage of the known encapsulated forms of prolonged action is the difficulty of obtaining them, the use of sophisticated technological equipment.
The most preferred solid oral dosage form is tablets.
The closest technical solution to the claimed object is a well-known pharmaceutical composition in the form of a tablet, comprising the active ingredient doxazosin mesylate, corn starch and lactose as a filler, magnesium stearate, and water as a glidant (US Pat. RF. 2139053).
The known composition of the tablets according to the prototype has a low strength and a limited shelf life. The authors found that doxazosin is chemically incompatible with starch, cellulose and causes its hydrolysis with the formation of by-products, causing the formation of brown spots on the surface of the tablets.
The purpose of this invention is the creation of a simple and affordable form of the drug in the form of a tablet containing doxazosin mesylate as an active ingredient, increasing the strength of the tablet.
1231877, 1698263.This goal is achieved through the use of a pharmaceutical composition in the form of a tablet comprising the active ingredient doxazosin mesylate, excipient, magnesium stearate and lactose. In this case, a mixture of aerosil and exopolysaccharide taken in a ratio of 1: 15-20 is used as a filler, and the composition additionally contains polyvinylpyrrolidone and sodium dodecyl sulfate in the following ingredients, wt.%:
Doxazosin Mesylate - 2.0-3.0
A mixture of Aerosil with exopolysaccharide - 35-45
Magnesium stearate - 0.5-1.5
Sodium dodecyl sulfate - 0.1-0.3
Polyvinylpyrrolidone - 2.5-3.0
Lactose - The rest is up to 100%
It is known to use in the pharmaceutical industry for the preparation of medicines in the form of tablets as a surfactant sodium dodecyl sulfate and polyvinylpyrrolidone (US Pat. RF 2159107, 2039553) and as a dusting agent and improve flowability - Aerosil (US Pat. RF 1748822), but the use of these ingredients in drugs on the basis of doxazosin is unknown. The use of these ingredients individually did not lead to the goal in drugs based on doxazosin. For the purposes of this invention, an exopolysaccharide biopolymer obtained by growing a producer of Bacillus polymyxa 1459-B in a nutrient medium followed by isolation of the desired product was used. This known method for producing exopolysaccharide is described in ed. St. 1231877, 1698263.
Doxazosin tablets are made by the classical method of wet granulation, followed by drying of the granules, which are then dusted and compressed into tablets.
The exopolysaccharide is pre-dried at a temperature of 70-80 o C. All the ingredients are sieved through a sieve. A solution of polyvinylpyrrolidone is prepared by adding a calculated amount of water.
Aerosil is mixed with exopolysaccharide in a ratio of 1: 15-20. Doxazosin mesylate is added, mixed. Lactose is added. Mix everything. Then a solution of polyvinylpyrrolidone is added. Mixed. The wetted mass is sent to a granulator. Dried to a moisture content of 2-3%. Dry granules are then dusted with magnesium stearate and sodium dodecyl sulfonate. The powdered mass is tabletted.
An aerosil-added exopolysaccharide is able to bind to a macromolecule having a strong bulk structure that does not change as a result of compression. Thus, the tablet has increased strength. Due to the introduction of sodium dodecyl sulfate and polyvinylpyrrolidone into the composition, the active ingredient doxazosin is well distributed in the total mass subjected to granulation.
1 и 2. The experimental results are summarized in table. 1 and 2.
