(Farooqui и Horrocks, Brain Res. Rev. 16, 171, 1991).BACKGROUND OF THE INVENTION
Stimulatory amino acids such as L-glutamate (Glu) and L-aspartate (Asp) are the main neurotransmitters in the central nervous system of mammals. For these acidic amino acid neurotransmitters, there are many subtypes of acidic amino acid receptors. For example, they include ion channel-coupled receptors mediating neuronal depolarization, named for the typical antagonists of N-methyl-D-aspartate (NMDA), alpha-amino-5-methyl-4-isoxazolepropionic acid (AMPA), kainate and imaginary presynaptic stimulator L-2-amino-4-phosphonobutyrate (L-AP4). The fifth receptor for stimulatory amino acids is the metabotropic receptor associated with phosphoinositide metabolism. (Farooqui and Horrocks, Brain Res. Rev. 16, 171, 1991).
10, 280, 1987). NMDA receptors play a specialized role due to the unique characteristics of their associated ion channels and participate in a variety of plastic neural events, including the initiation of long-term potentiation, which is a hypothetical substrate for learning, memory, and establishing synaptic contacts during neuronal development. NMDA receptors are also involved in other processes, such as the transmission of sensory information (MacDermott and Dale, Trends Neurosci. 10, 280, 1987).
In addition to their important physiological role, stimulatory amino acids such as NMDA are also involved in pathophysiological events in the central nervous system.
[изд.]: "Heritage Disorders of Amino Acids Metabolism" New York: Macmillan pp. 501-512, 1989) для описания процесса, посредством которого стимуляторные аминокислоты могут вызывать смерть нейронных клеток. Abnormally low levels of glutamic acid (Glu) can alter normal levels of stimulation and lead, for example, to learning and memory deficits. Excessive Glu levels can lead to toxic effects. The term “stimulotoxicity” was coined by Olney (in Hyhan WL [ed]: "Heritage Disorders of Amino Acids Metabolism" New York: Macmillan pp. 501-512, 1989) to describe the process by which stimulatory amino acids can cause death neural cells.
(Said, S.I. et al., Letters to Neuroscience, 65, 943-946, 1995). Rev. Pharmacol. Toxicol. 30: 47- 71, 1990; Foster et al., в "Current and future Trends in Anticonvulsant, Anxiety and Stroke Therapy" Wiley-Liss, Inc. pp. 301-329, 1990; Rogawski and Porter, Pharmacol. Rev., 42: 223-286, 1990). There is evidence that NMDA receptors exist in peripheral tissues and that activation of these receptors may be involved in the mechanism of damage to the lungs and other organs (Said, S.I. et al., Letters to Neuroscience, 65, 943-946, 1995). This cytotoxic process is mainly mediated by overstimulation of NMDA receptors and can occur in cases of cerebral palsy, cerebral ischemia, epilepsy, Alzheimer's disease, AIDS-related dementia, traumatic brain injuries and other neurodegenerative disorders (Olney, Ann. Rev Rev. Pharmacol. Toxicol. 30: 47-71, 1990; Foster et al., In Current and future Trends in Anticonvulsant, Anxiety and Stroke Therapy Wiley-Liss, Inc. pp. 301-329, 1990; Rogawski and Porter, Pharmacol. Rev. 42: 223-286, 1990).
NMDA receptors contain several binding domains that interact with each other for proper functioning and modulation of nerve cell activity. There is a theory that the NMDA receptor forms a complex that acts as an ion channel bound to the receptor. More precisely, the function of the receptor is the binding of NMDA or natural amino acids, Glu or Asp, and the opening of the bound ion channel, which allows sodium (Na + ) or calcium (Ca 2+ ) to enter the stimulated neuron, as well as potassium (K + ) to leave.
While the ion channels of the receptors of other stimulatory amino acids (AMPA, kainate and L-AP4) are permeable only to Na + and K + , the channel of the NMDA receptor is also permeable to Ca 2+ . This feature may be important for the intended role of this receptor for both short-term and long-term plasticities, such as learning, memory, and neuropathology.
Rev. 16, 171; 1991).Intracellular Ca 2+ is responsible for the regulation of a wide variety of cellular processes (Farooqui and Horrocks, Brain Res. Rev. 16, 171; 1991). Overstimulation of brain NMDA receptors observed in cases of anoxia, ischemia and hypoglycemia leads to an accumulation of Ca ++ concentration in stimulated neurons and to a cascade of intracellular events (activation of phospholipases [PLA 2 , PLC], lipases, proteases and endonucleases), which leads to the death of neuronal cells (Farooqui and Horrocks, Brain Res. Rev. 16, 171; 1991).
Therefore, there is a need for compounds that can bind or antagonize the NMDA receptor complex or otherwise protect neurons against degeneration induced by a stimulatory amino acid receptor.

The therapeutic agents of the present invention may be administered individually or in combination with pharmaceutically acceptable carriers. The proportion of each carrier is determined by the solubility and chemical nature of the compound, by administration and standard pharmaceutical practice.
In order to obtain a consistency for administration, it is preferred that the composition of the invention is in unit dosage form. The unit dose forms for oral administration may be tablets and capsules and may contain conventional excipients. For example, binding agents such as acacia, gelatin, sorbitol or polyvinylpyrrolidone; fillers such as lactose, sugar, corn starch, calcium phosphate, sorbitol or glycine; tabletting lubricants such as magnesium stearate; disintegrants such as starch, polyvinylpyrrolicone, sodium bricollate starch or microcrystalline cellulose; or pharmaceutically acceptable wetting agents, such as sodium lauryl sulfate.

The compounds may be administered parenterally; this means intramuscularly, intravenously or subcutaneously. For parenteral administration, compounds in the form of sterile solutions containing other solutions, for example, saline or glucose, sufficient to make the solution isotonic, can be used.
The compounds may be administered orally in the form of tablets, capsules or granules containing suitable excipients such as starch, lactose, white sugar and the like. The compounds may be administered orally in the form of solutions, which may contain color and / or odor enhancing agents. The compounds can be administered sublingually in the form of tablets or lozenges, in which each active ingredient is mixed with sugar or corn syrups, odorants and colorants, and then sufficiently dehydrated to make the mixture suitable for compression into a solid form.
Solid oral formulations may be prepared by conventional methods of mixing, filling, tabletting or the like. Repeated mixing operations can be used to distribute the active agent in compositions where large amounts of excipients are used. Such procedures are, of course, traditional in the area. The tablets may be coated according to methods well known in normal pharmaceutical practice, especially intestinal membranes.
Oral liquid preparations may be in the form of emulsions, syrups or elixirs, or may be presented as dry products for reconstitution with water or other suitable vehicles before use. Such liquid preparations may or may not contain conventional additives. For example, suspending agents such as sorbitol, syrup, methyl cellulose, gelatin, hydroxyethyl cellulose, carboxymethyl cellulose, aluminum stearate gel or hydrogenated edible fats; emulsifying agents such as sorbitan monooleate or acacia; non-aqueous vehicles (which may include edible oils) such as almond oil, fractionated coconut oil, oil esters selected from the group consisting of glycerol, propylene glycol, ethylene glycol and ethyl alcohol; preservatives, for example, methyl parahydroxybenzoate, ethyl parahydroxybenzoate, n-propyl parahydroxybenzoate or n-butyl parahydroxybenzoate sorbitol acid; and, if required, traditional agents that add a smell or color.
For parenteral administration, unit dosage forms may be prepared by using the compound and a sterile vehicle, and, depending on the concentration used, may be either suspended or dissolved in the vehicle. Once in solution, the compound can be injected and sterilized on the filter before filling in a suitable vial or ampoule, followed by sealing the carrier or packaging for storage. Adjuvants, such as local anesthetics, preserving or buffering agents, may be dissolved in the vehicle before use. The stability of the pharmaceutical compositions can be enhanced by freezing the composition after filling the vial and removing water under vacuum (for example, drying by freezing the composition). Parenteral suspensions can be prepared essentially in the same way, except that the compound should be suspended in the carrier rather than dissolved, and further, sterilization is not achieved by filtration. The compound can be sterilized, however, by exposing it to ethylene oxide before suspending it in a sterile vehicle. A surfactant or moisturizing solution can be advantageously formulated to facilitate an identical distribution of the compound.

Им инъецировали I.С.V. Exp. Therm. 252 349-357, 1990).
A. Locomotor and impaired behavior
Mice were placed individually in observation cells for 60 minutes for the addictive period. They injected I.C.V. test compounds and were observed for 15 to 30 minutes. Locomotorism and behavioral disorders were evaluated in accordance with Koek Colpaert procedures (J. Pharm. Exp. Therm. 252 349-357, 1990). For each animal, the presence of locomotor activity and behavioral disorders were recorded. The statistical significance of drug-induced changes in accordance with defined behavior was tested using the methods of Fray et al. (Psycopharmacol 69 253-259, 1980). MK-801, known as having significant neurotoxicity, induced behavioral disturbances at doses as low as 1.5 nmol / mouse.
Б. B. Assessment of motor coordination by the Rotarod.
Pharmac. Assoc. 46 208-209- 1957). The effect on movement enhanced by various antagonists was evaluated using a rotarod treadmill (model 7600, EGO Basile, Italy). The method used was similar to the procedure described by Dunhamand Miya (J. Am. Pharmac. Assoc. 46 208-209-1957). The device consisted of a rail with a diameter of 2.5 cm, which is suspended horizontally at a height of 50 cm above the working surface. The rail was turned at a speed of 8 rpm. Circular dividers were placed at intervals along the rail so that five animals could be tested simultaneously. All animals developed addiction to rake for two consecutive days before experimentation.
A study in which comparative motion damage was caused by Compound # 13 was compared with dextrorphan when evaluated by rotarod. The percentage of animals exhibiting an effect on movement is higher for animals treated with dextrorphan than for animals treated with Compound # 13.
