Изобретение относится к биологии и медицине и может быть использовано при хранении пространственно организованных клеточных структур и клеточных ассоциаций (суспензий). Известен способ хранения культуры клеток в замороженном с криопротектором (глицерин или ДМСО) состоянии при -196oC (жидкий азот) [1,2], а также способы хранения клеток крови в жидком состоянии при +4oC с консервантом ЦОЛИПК-7Б [3].A known method of storing a cell culture in a frozen state with a cryoprotectant (glycerin or DMSO) at -196 o C (liquid nitrogen) [1,2], as well as methods of storing blood cells in a liquid state at + 4 o C with the preservative TSOLIP-7B [ 3].

Данный способ сохранения пространственной организации клеточных культур неизвестен из доступных литературных источников, что свидетельствует о соответствии заявляемого изобретения критерию "новизна" и "изобретательский уровень". Далее следует пример конкретного выполнения способа: культуры клеток человека, образующие 3-мерные структуры в суспензионном состоянии (лимфоцитарные и макрофагальные клеточные суспензии) смешивают со свежей питательной средой RPMI-1640 с добавлением в нее 20%-ной фетальной сыворотки и заполняют в стерильный пластиковый контейнер объемом 5 мл без воздушного зазора. Пластиковый контейнер помещают в камеру высокого давления, которая представляет собой цилиндр с завинчивающимися пробками и с электровводами для измерения давления, контроля температуры и термоэлементом для ее поддержания в заданном режиме. Полость камеры высокого давления заполнена селиконовым маслом, которое бесконтактно передает создаваемое в камере ручным способом давление (450-550 атм) образцу. Охлаждение образцов в камере достигается либо подачей через проточную систему камеры жидкого азота, либо в криостате до заданной температуры. Образец с клеточной культурой необходимый период времени (например, 6 мес или 1 год) хранится при заданных условиях. По окончании сроков хранения камеру либо вынимают из криостата, либо подогревают до комнатной температуры включением термоэлемента, после чего плавно в режиме 10-20 атм/мин сбрасывается гидростатическое давление до атмосферного. Указанная последовательность операций и плавный сброс давления необходимы для предотвращения повреждающего действия высокого давления на клеточные ассоциации и предотвращения их замерзания. В случае сброса давления до уровня атмосферного без предварительного повышения температуры образцов до комнатной температуры культура клеток может подвергнуться замерзанию при отрицательной (ниже 0oC) температуре. Учет результатов производят до и после воздействия по жизнеспособности клеток окрашиванием 0,4%-ным раствором трипанового синего и последующим подсчетом живых и мертвых клеток в камере Горяева по обычной методике, а также по сохранности организации тестируемых клеточных субпопуляций в 3-мерных клеточных структурах (кластерах), - процент которых определяют в сравнении с контрольными клеточными культурами.The following is an example of a specific implementation of the method: human cell cultures forming 3-dimensional structures in suspension (lymphocytic and macrophage cell suspensions) are mixed with fresh RPMI-1640 nutrient medium with the addition of 20% fetal serum and filled into a sterile
