Abstract
Description
The invention relates to the development of methods for the rapid detection of illegal drugs, in particular heroin, by changing the color of the color-forming reagent when it interacts with the test substance.
Heroin is usually obtained from morphine by acetylation of the latter; as a chemical compound, heroin is diacetylated morphine (alkaloid).
247). Known methods for the qualitative analysis of alkaloids (Handbook of analytical chemistry, M., 1979, p. 247).
1. Microcrystalline.
Since alkaloids salts have a strictly defined crystal shape, after planting, observe the crystal shape and compare it with reference samples (forensic chemistry).
2. The color change of the alkaloid when processing indicator.
The tertiary nitrogen atom has the basic (alkaline) properties and, depending on the structure of the alkaloid, is characterized by the basicity constant pK c . When interacting with acid-base indicators (such as, for example, bromphenol blue - BPS), the alkaloid forms an ion pair with the indicator, changing the distribution of the electron density of the indicator and its color. The interaction of the alkaloid (heroin) with BFS leads to the formation of a complex in which the BFS changes the yellow color from λ max = 436 nm to blue λ max = 542 nm.
A known method for the qualitative determination of drugs of plant origin, such as morphine (RF patent N 2024868, G 01 N 31/20, 31/22, 1994). The method consists in processing an aliquot of the analyzed sample with a solution of hydrochloric acid, then with a 1% solution of formalin in sulfuric acid. In the presence of common alkaloids, a yellow precipitate forms in the sample. After that, a new sample is taken, a 10% sodium carbonate solution is added to it, the mass is mixed until completely moistened, chloroform is added, it is shaken and a 10% solution of formalin in concentrated sulfuric acid is added to the separated chloroform layer. Staining of the chloroform layer in violet indicates the presence of morphines in the sample.
The method is complex, multi-stage and requires various reagents for the detection of morphines.
A known method for the detection of heroin, including spraying the surface of a solid carrier with a color-forming reagent placed on it with a 1-4% aqueous solution of bromphenol blue indicator (BPS), moreover, 20% of the water can be replaced by alcohol. If there are traces of heroin on the surface, the latter turns black. Thus, the presence of heroin is judged by the color change of the color-forming reagent (French patent N 2629915, G 01 N 33/52, 1989).
The disadvantages of this method include its lack of information, since the indicator changes color when interacting with any alkaloid, which complicates the identification of drugs.
The objective of the invention is to increase the specificity, information content and reliability of the method of rapid detection of heroin.
The essence of the method consists in the use of an indicator element in the form of filter paper for detecting heroin, on which a double-ring imprint of a saturated solution of bromophenol blue (BPS) in an organic solvent with methylene chloride is applied. The surface of the indicator element is sealed with a polymer film and a hole is cut out in the middle part, which is closed with a carrier with an adhesive layer, thus forming an indicator hole on which the sample of the analyte is placed. After that, the sample is wetted with methylene chloride. If heroin is present in the sample, it dissolves in methylene chloride and the solution diffuses along the paper layer to indicator rings, which in the presence of heroin change their color from yellow to blue-violet.
The indicator element is shown in the drawing, where 1 is indicator (filter) paper sealed in a polymer film, 2 is indicator rings, 3 is an indicator hole.
The BFS indicator non-specifically reacts with the analyte, since it is an ion-pair reagent for alkaloids that have similar basicity characteristics (pK in ). Therefore, a variety of alkaloids, other than heroin in structure, but have pK values close to, will give the same response.
In accordance with the invention, in order to reduce the frequency of false responses, the BFS indicator is separated from direct contact with the analyzed sample: the sample is placed in the well, and the BFS is applied at a distance from the well. In the proposed method, an analyte solution reacts with an indicator. Therefore, some alkaloids that are not soluble in methylene chloride do not diffuse to the indicator zone and do not give a false positive. Thus, the reliability of detection of heroin against the background of other alkaloids is increased.
In addition, the proposed method can reduce the likelihood of false positives in the analysis of wet samples. If a solution of the indicator is applied to a wet sample, then regardless of the chemical structure of the sample in the presence of water, the indicator will change color. Since water does not mix with methylene chloride, in the proposed ring structure of the indicator, a solution of the sample in the solvent will diffuse before the indicator and interact with it, and not the side impurities insoluble in methylene chloride.
The achievement of the technical result is achieved through the joint use of a combination of two methods: visual colorimetry and paper chromatography using methylene chloride and an indicator (ring impression) from a saturated solution of BFS in a special solvent - methylene chloride.
Thus, the novelty of the method lies in the use of a special reagent and in a certain way prepared indicator element.
The invention is illustrated by examples.
1. Example 1
An imprint of two rings with diameters of 15 and 25 mm from a saturated solution of BFS in methylene chloride is applied to the filter indicator paper. After that, the surface of the indicator paper is sealed on both sides with a layer of polyethylene and a hole with a diameter of 10 mm is cut out in the middle, which is closed with adhesive tape.
Take the indicator element (see drawing) and in the indicator hole 3 on a sticky surface place a sample of the analyte in an amount of 5 mg containing 50% heroin. The sample is sprayed with a solvent - methylene chloride. After 5 s, the color of the indicator rings 2 changes from yellow to blue-violet, which indicates the presence of heroin in the analyzed sample.
The developed method for the rapid detection of heroin is simple and more informative compared to the known ones, as it allows heroin to be detected with greater probability (reliability). Thus, the developed method is specific to heroin. The uniform nature of the color intensity of the indicator rings increases the reliability of heroin detection and reduces the likelihood of artifacts (false positives) in practical use.
