Abstract
Description
The invention relates to biotechnology, in particular to in vitro cultivation of flax, and can be used in the cultivation of anther plants to obtain new starting material.
Numerous nutrient media for the cultivation of anthers of plants are known, which contain the necessary components to induce the formation of callus and embryoids based on them (F.A. Kalinin et al. Methods of tissue culture in plant physiology and biochemistry. Kiev: Naukova Dumka, 1980. P. 238-242). It was shown that there is no universal nutrient medium that allows to obtain a sufficient result on different plants, and therefore environments are developed for specific types of plants (Plant Biotechnology: cell culture / Transl. From English V.I. Negruka; from the foreword R.G. Butenko, Moscow: Agropromizdat. 1989.280 p. (P. 41-43). The use of the main well-proven media such as N 6 (Cu S.S., 1978), MS (Murashige, Skoog, 1962), Monnier (Monnier M., 1978), B 5 (Gamborg OL, 1965) and others on Flax was accompanied by the formation of only isolated cases of anther callus or the emergence of embryoids based on them.
A 22 (Nichteriein K., et al. New methods and recent progress in the breeding of fiax / Flax as a fibre and oil bearing crop. Proceedings of the FAO European regional workshop on flax.-Brno, Czechoslovakia, 18-20 June 1991. A culture medium is known (the closest analogue) for the cultivation of flax anthers, called A 22 (Nichteriein K., et al. New methods and recent progress in the breeding of fiax / Flax as a fiber and oil bearing crop. Proceedings of the FAO European regional workshop on flax.-Brno, Czechoslovakia, 18-20 June 1991. P. 175-183; Nichterlein K., Umbach H., Friedt W. Genotypic and exogenous factors affecting shoot regeneration from anther callus of linseed (Linum usitatissimum L). Euphytica . 1991. 58. P. 157-164), which contains (mg / l): calcium chloride two-water - 435; potassium phosphate monosubstituted - 170; potassium nitrate - 1900; magnesium sulphate heptahydrate - 370; ammonium nitrate - 165; ferrous sulphate - 27.8; sodium ethylene diamine tetracytate ((EDTA) - 37.3; hexavalent cobalt chloride - 0.025; pentahydrate copper - 0.025; boric acid - 6.2; potassium iodide - 0.75; manganese sulfate - 22.3; sodium molybdenum two-water - 0.25; zinc sulphate - 8.6; menzoinositol - 100; thiamine - 0.1; glutamine - 750; sucrose - 60,000; agarose - 4000; 6-benzyladenine - 2.0; naphthylacetic acid - 1.0 .
The disadvantage of these environments is the low yield of callus anthers of flax and anthers with embryoids. An increase in the incidence rate of encapsulating anthers in flax is very important, since the technology of anther culture in vitro is quite expensive, and breeders do not always have enough valuable plant material. Increasing the frequency of callusogenesis can reduce the volume of preparation of the medium, laboratory equipment, the volume of plant material, the consumption of working time and creates the basis for the technology of mass production of regenerants in the anther flax culture.
