Abstract
Description
The invention relates to medical virology and can be used in the manufacture of drugs for the prevention of epidemic flu.
Currently, in the preparation of reassortant vaccine strains of influenza B virus, several basic donor strains are used for the production of inactivated and live influenza vaccines, which ensure the production of harmless or highly reproductive viruses with a modern antigenic structure of surface proteins. The strain of influenza virus B / Lee / 40 (1) is a multi-passage virus, used as a high reproductive donor for the construction of high-yielding reassortants to obtain an inactivated influenza vaccine. Strain B / Leningrad / 14/55/17, B / USSR / 1/69/60 (2) and B / Leningrad / 2/67/115 are used as an attenuation donor to create harmless reassortants for a live influenza type B vaccine. Strains analogous to donor B / Leningrad / 14/76/50, used to construct harmless and highly reproductive strains of influenza B virus for the production of inactivated and live vaccines, do not exist in the world.
The virus strain B / Leningrad / 14/76/50 was obtained by serial passaging of the original virus B / Leningrad / 14/76 at optimal and infraoptimal incubation temperatures, followed by repeated cloning in developing ten-day-old chicken embryos and on chicken fibroblast cell culture. As a result of selection, a clone of this strain was selected with high reproduction at 32 o C (hemagglutinating activity 1: 1024 - 2048 AE; infectious activity 9.0 - 9.25 lg EID 50 / 0.2 ml) and 26 o C (hemagglutinating activity 1: 1024 AE; infectious activity 4.5 - 5.0 lg EID 50 / 0.2 ml).
Based on the internal genes of influenza B / Leningrad / 14/76/50, a reassortant with surface proteins of the epidemic strain B / Panama / 45/90 was obtained. The specified reassortant inherited the ability to multiply at 26 o C to 4.0 - 4.25 lg EID 50 / 0.2 ml and at 32 o C to 8.5-8.75 lg EID 50 / 0.2 ml, i.e. . possessed cold adaptation and high reproductivity - the characteristics necessary for the production of live and inactivated influenza vaccines.
Thus, based on one strain of the donor, a reassortant was obtained, which is simultaneously used for the production of live and inactivated influenza vaccines.
