Синтез пептида проводили на автоматическом синтезаторе пептидов Biosearch 9600 (США) на РАС-смоле, используя Fmoc/DIPCDI метод по стандартной программе c: FDI, прилагаемой к прибору. Конденсацию Fmoc-аминокислот проводили карбодиимидным методом, для подавления рацемизации добавляли эквимолярные количества 1-оксибензтриазола. Остаток тирозина вводили в виде его о-трет-бутилового эфира. Стартовую аминокислоту триптофан присоединяли к 5 г РАС-полимера в количестве 0,76 ммоль аминокислоты на 1 г полимера. Далее Trp-полимер обрабатывали 0,4 М растворами соответствующих защищенных аминокислот, содержащими примерно 7-кратный избыток соответствующей аминокислоты и диизопропилкарбодиимида. Отщепление Fmoc-группы после конденсации проводили смесью пиперидин-толуол-диметилформамид (30:35:35). После окончания синтеза пептид отщепляли от смолы трифторуксусной кислотой, содержащей 2,5% этандиола и 2,5% воды при охлаждении льдом в течение 1,5 часов. Полученный пептид выделяли и очищали высокоэффективной хроматографией на колонке Диасорб-130 С-16 Т, 10 мкм, размером 26х250 мм в градиенте ацетонитрила в 0,05 М фосфатном буфере рН 3,0 и обессоливали на той же колонке. После лиофилизации получали 2 г белого аморфного порошка, гомогенного по данным ВЭЖХ. Аналитическую хроматографию проводили на колонке Ультрасфера ODS-3 в градиенте ацетонитрила (20 80%) в 0,05 М фосфатном буфере рН 3,0, детекцию осуществляли при 220 нм. Аминокислотный анализ кислого гидролизата (6н HCl, 20 часов) показал наличие следующих аминокислот: Leu 1 (0,95), Val 2 (1,87), Tyr 1 (1,03), Pro 1 (0,96). After completion of the synthesis, the peptide was cleaved from the resin with trifluoroacetic acid containing 2.5% ethanediol and 2.5% water while cooling with ice for 1.5 hours. The resulting peptide was isolated and purified by high-performance chromatography on a Diasorb-130 C-16 T column, 10 μm, size 26x250 mm in an acetonitrile gradient in 0.05 M phosphate buffer pH 3.0 and desalted on the same column. After lyophilization, 2 g of a white amorphous powder homogeneous according to HPLC was obtained. Analytical chromatography was performed on an ODS-3 Ultrasphere column in an acetonitrile gradient (20–80%) in 0.05 M phosphate buffer pH 3.0, and detection was carried out at 220 nm. Amino acid analysis of an acid hydrolyzate (6N HCl, 20 hours) showed the presence of the following amino acids: Leu 1 (0.95), Val 2 (1.87), Tyr 1 (1.03), Pro 1 (0.96).
