Изобретение относится к области получения высокомолекулярных биологически активных соединений, а именно к способу получения водорастворимых поликомплексов хитозана с декстрансульфатом натрия, способным специфично связывать атерогенные липопротеины из крови. Известны способы получения интерполимерных комплексов (ИПК) хитозана с различными сульфополисахаридами [1, 2] В частности, рассматриваются нерастворимые в воде поликомплексы гепарина с хитозаном и гликольхитозаном, проявляющие антитромбогенные свойства, однако нет данных об их применении для сорбции липопротеинов. Основной недостаток упомянутых комплексов связан с их нерастворимостью в воде и обычных органических растворителях, что обусловливает непригодность подобных композитов для нанесения покрытий. Поставленная задача достигается тем, что в способе получения интерполимерных комплексов хитозана с декстрансульфатом натрия путем смешения растворов исходных компонентов, выдерживания реакционной смеси и выделения целевого продукта, в качестве растворителя используют воду или 2-6 М водный раствор мочевины, в качестве исходного раствора хитозана раствор, содержащий соль хитозана с органической или минеральной кислотой, имеющий рН 6,0-7.5, и выдерживание реакционной смеси осуществляют до достижения рН 7,5-9,0, при молярном соотношении соль хитозана: декстрансульфат натрия, равном 1:2-12, причем отношение молекулярных масс соли хитозана и декстрансульфата натрия выбирают в диапазоне 0,02-0,4. Как видно из таблицы, где представлены данные экспериментов, при соотношении компонентов, больше 1:12 коэффициент седиментации раствора ИПК и содержание серы в нем не изменяются. При соотношении компонентов меньше 1:2 образуется нерастворимый комплекс в водно-солевых средах. ИПК ДС и солей хитозана с молекулярной массой меньше 10 кД непрочные или вовсе образование ИПК не происходит. При использовании хитозана с молекулярной массой выше 200 кД, поликомплекс выпадает в осадок. Верхняя граница растворимости солей хитозана от рН определяется потерей их растворимости при рН больше 7,5, а нижняя граница (меньше 5,5) нерастворимостью конечного продукта. При рН конечного раствора ИПК меньше 7,5 система становится мутной и следовательно, происходит изменение состава ИПК, а при рН больше 9,0 в системе происходит фазовое разделение. При концентрации мочевины меньше 2 моль/л частицы поликомплексов с концентрацией 1% и более находятся в ассоциированном состоянии, а больше 6 моль/л не наблюдается изменение в состоянии системы. Полученные ИПК могут быть использованы в качестве аффинного лиганда для связывания атерогенных липопротеинов в гемосорбентах, что иллюстрируют примеры 21 и 22. The obtained IPC can be used as an affinity ligand for binding of atherogenic lipoproteins in hemosorbents, as illustrated by examples 21 and 22.
