Abstract
Description
The invention relates to medicine, more specifically to microbiology, and can be used in the treatment of infectious diseases, in gastroenterology and pediatrics.
With dysbiosis, bacterial preparations that restore eubiosis are widely used. Their development is carried out along the path of selecting strains that have an antagonistic effect against pathogenic flora. Official preparations are produced in the form of starter cultures, powders, gelatin capsules, tablets, including freeze-dried eubiotics biomass. The main condition for these dosage forms is to ensure maximum viability of the microorganism included in the preparation and to ensure its inclusion in the intestinal biocenosis.
These requirements can be implemented by lengthening the course of treatment, increasing a single dose or creating compositions that include components that prevent the death of the applied strain in the gastrointestinal tract. Gelatin, pectins, etc. were proposed as such components. Recent studies have shown that used protectors can block the adhesion of microbes, thereby inhibiting their colonization in the intestine. The loss of activity of a medicinal product is possible at the stage of drying the biomass.
The stabilizing additives used during lyophilization (sucrose-gelatin, milk reverse, etc.) provide a yield of not more than 80-85% of viable culture cells. The foregoing is the basis for the selection of the optimal tread components for the creation of this kind of biological products.
Known drug based on L.bulgaricus, which is a starter for milk. Its disadvantage is that it does not have a dosage form and can be dosed.
A preparation is also known, which is a composition in which pectin is included as a protective component. Its disadvantage is that pectin causes blocking of adhesion, thereby preventing the colonization of the strain used.
A well-known drug is dry lactobacterin, taken as a prototype, available in powder form and including dried
The aim of the invention is to maintain the viability of the culture during lyophilization, as well as the protection of bacteria from the effects of gastric juice.
3%.This goal is achieved in that the composition includes lactobacilli of the
3%.The drug is prepared as follows. The native biomass of lactobacilli is obtained according to laboratory regulations, diluted to a concentration of 5x10 9 to 5x10 10 in 1 ml of sterile isotonic sodium chloride solution. 4 volumes of 4% sodium alginate are added to 1 volume of suspension after mixing, the mixture is granulated (5). The obtained granules are washed three times with water and transferred to vials with isotonic solution (80 granules in 1 vial), and then freeze-dried. Before use, the granules are rehydrated in 2 ml of boiled tap water. The prepared composition has the following composition: L.fermentum 1 - 7% (5x10 9 to 5x10 10 living cells in 1 ml of rehydrated composition), calcium alginate 90-96% and water - up to 3%.
П р и м е р. PRI me R. The stability of granules in an environment close to the conditions of their presence in various parts of the gastrointestinal tract was investigated. The pellets were incubated for 24 hours at 37 ° C in a medium with different pH values in the range from 1 to 12. When the pH - 1-7 alginate beads did not change, and at a pH of 8 and above their swelling was observed and the complete dissolution.
From this it follows that when the drug enters the stomach, where the pH is below 7, the microorganisms included in the granules remain immobilized, and when passing through the intestines a pH above 8) they are released from the granules.
The effect of various substances of interest in terms of the protective components included in eubiotic preparations on the adhesion of microbes has been investigated.
The percentage of blocking the adhesion of lactobacilli on red blood cells under the influence of alginates, pectins, dextran was determined.
The experimental results are given in Tables 1, 2. It follows from them that of all the investigated substances, alginates do not cause inhibition of bacterial attachment to target cells. This is their property that was used when choosing a protective component in the proposed composition.
The protective properties of alginate during freeze drying of biomass were studied. Comparative experiments were conducted on drying native biomass using traditional stabilizers (sucrose-gelatin supplement) and drying bacteria immobilized in alginate. It was established that after lyophilization by the traditional method, from 80 to 85% of viable bacteria remain in the biomass, while at the same time, from 90 to 95% of living organisms remain in the granules when dried.
