Известен способ оценки уровня гибридности семян кукурузы, который состоит в том, что из эндосперма кукурузы осуществляют экстракцию белка. Полученный белок разделяют на пять групп с последующим определением относительной электрофоретической подвижности каждого белкового компонента. На основании анализа электрофореграмм составляют белковые формулы для гибридов и их родительских линий. Гибридным считается такое зерно, зеиновый спектр которого включает все позиции белковых компонентов отцовской и материнской линии. Цель изобретения - обеспечение возможности оценки относительной степени гомо- и гетерозиготности с учетом селекционно значимых признаков образцов кукурузы. Цель достигается тем, что из оцениваемых образцов получают матроклинные гаплоиды с применением генетических индуцирующих линий. Полученные семена с гаплоидным зародышем высевают на полевом участке. Технология выращивания гаплоидных растений - общепринятая для кукурузы. Затем определяют степень изменчивости гаплоидных растений по оцениваемым морфологическим признакам. После чего проводят ранжирование изучаемых образцов по вариации их гаплоидов, при этом к относительно гомозиготным относят те образцы, гаплоиды которых имеют наименьшую вариацию признака. Объектом исследований служили два образца кукурузы - хорошо отселектированная заведомо гомозиготная линия и потомство F4, полученное в результате трехкратного самоопыления гибрида F1, заведомо имеющее определенную степень гетерозиготности. Данные образцы во время цветения растений были опылены гаплоиндуцирующей линией Зародышевый маркер Саратовский. Из полученных семян отбирали те, которые содержат гаплоидный зародыш. Отбор проводят по наличию антоциановой метки на эндосперме и отсутствию ее у зародыша. Как правило, количество отобранных гаплоидов составляет 0,5-3,0% от общего количества семян. Гаплоиды высевают на полевом участке общепринятым способом. Технология возделывания растений не отличается от таковой для обычной диплоидной кукурузы. Измерения морфологических признаков проводят после окончания роста растений. Всего было изучено 96 диплоидных и 97 гаплоидных растений. Измеряют следующие признаки: высота растения, длина метелки, длина листа, ширина листа и количество листьев. В таблице представлены полученные коэффициенты вариации. Из данных, представленных в таблице, видно, что вариация гаплоидных растений, полученных из гомозиготной линии 092, ниже вариации гаплоидов потомства F4 по всем изучаемым признакам. Так, например, по высоте растения коэффициенты вариации гаплоидов линии 092 и потомства F4 составили 14,4 и 22,7% соответственно; по длине метелки - 18,6 и 27,6. В то же время анализ изменчивости признаков диплоидных растений не позволяет отличить оцениваемый в данном случае уровень гомо- и гетерозиготности. Так, вариация потомства F4 по трем признакам из пяти оказалась ниже вариации гомозиготной линии 092. Т.е. выбранный уровень различий в гомо- и гетерозиготности линии 092 и потомства F4 не может быть оценен при измерении диплоидных растений.PRI me R.
